Abstract
本発明の目的は、TLRシグナルにおけるIKKαの役割を解明し、インターフェロン-αの制御剤及び制御方法を提供することである。本発明によれば、IKKαの阻害剤を含む、インターフェロン-α産生抑制剤が提供される。

背景技術
[0002] Toll様受容体 (TolHike receptors; TLRs)は、榭状細胞(DC)等の抗原提示細胞上 に発現しており、多様な微生物由来の分子構造の認識に関与している。 TLRリガンド としては、細菌及びウィルスに由来する脂質、タンパク質及び核酸成分が挙げられる 。 TLRの多くは、 IL-6などの炎症性サイト力インの誘導、共刺激分子の発現などを含 む、 TLRに共通の榭状細胞活性ィ匕作用を有している力 各 TLRは各 TLRに特有の機 能を示す。
[0003] TLR7及び TLR9はそれぞれ一本鎖 RNA及び非メチル化 CpG DN Aを認識でき る。これらの核酸を認識する TLRは、抗ウィルス免疫に必要な IFN— α及び IFN— 等の I型インターフェロン(IFN)を誘導できることを特徴とする(Wagner, H, Trends Immunol 25, 381-6 (2004))。 TLR7及び TLR9は、細胞質内アダプター分子である MyD88と会合する。 MyD88は、榭状細胞が TLRリガンドに応答して、 I型インター フエロンのみならず炎症性サイト力インを産生するのに必要である。転写因子 IRF— 7も、 TLR— 7Z9により誘導される I型インターフェロンの産生に重要である(Honda, K et al., Nature, 434, 772-7 (2005))。 IRF7は MyD88と腫瘍壊死因子(TNF)受容 体関連因子 6 (TRAF6)に会合して、 IFN— α遺伝子を誘導する(Kawai, T. et al, Nat Immunol 5, 1061—8 (2004) ;及び Honda, K. et al., Proc Natl Acad Sci USA 101, 15416-21 (2004)

発明の開示
発明が解決しょうとする課題
[0006] 本発明は、 TLRシグナルにおける ΙΚΚ αの役割を解明し、インターフェロン一 ocの 制御剤及び制御方法を提供することを解決すべき課題とした。
課題を解決するための手段
[0007] 本発明者らは、 IKK a欠損マウスを分析して、 IKK aが TLRにより誘導される作用に いかに関与するのかを調べた結果、 TLR7,9刺激による榭状細胞力 の IFN- a産生 誘導に ΙΚΚ αが必要であることを見出し、本発明を完成するに至った。
[0008] 即ち、本発明によれば、 IKK aの阻害剤を含む、インターフェロン a産生抑制剤 が提供される。
好ましくは、 IKK aの阻害剤は、 IKK aの発現を阻害する物質、又はキナーゼ活 性を欠損する IKK a変異体である。
[0009] 本発明の別の側面によれば、細胞内の IKK a活性を阻害することを含む、細胞か らのインターフェロン一 aの産生を抑制する方法が提供される。

発明を実施するための最良の形態
[0012] 以下、本発明の実施の形態について説明する。
本発明は、 IKK αの阻害剤を含むインターフェロン α産生抑制剤、並びに IKK a、又は IKK aの発現を増大させる物質を含むインターフェロン a産生誘導剤に 関するものである。本明細書中では、インターフェロン a産生抑制剤及びインター フエロン ひ産生誘導剤を総称して、インターフェロン a制御剤と称する場合があ る。

[0014] 本発明で用いる IKK αの阻害剤としては、 IKK aの発現を阻害する物質、キナー ゼ活性を欠損する IKK a変異体、又は IKK aに作用して IKK aの活性及び機能を 阻害する物質等が含まれる。本明細書において、「阻害」との用語は、抑制又は低減 との意味を含む。
[0015] IKK aの発現を阻害する物質としては、 RNAi、アンチセンス法又はリボザィム法を 利用した物質等が挙げられ、特に限定されないが、 RNAiを利用した siRNAsが好まし V、。キナーゼ活性を欠損する IKK a変異体の具体例としては、 44番目のリジン残基
をァラニン残基に置換した変異体などが挙げられる。また、 ΙΚΚ αに作用して ΙΚΚ α の活性及び機能を阻害する物質としては、低分子化合物及び抗体等が挙げられる。
[0016] 抗体としては、例えば、 IKK aの全長又は部分配列を有するペプチドを免疫源とし て作製した抗体を用いることができる。全長の ΙΚΚ αとしては、例えば組み換え ΙΚΚ α等を用いることができる。抗体の作製は慣用の方法に従って行えばよい。抗体は モノクローナル抗体が好ましい。ペプチドとしては、例えば、 ΙΚΚ αの部分配列を有 するペプチド等が挙げられる。
[0017] RNAi (RNA interference)は、細胞に導入された 2本鎖 RNA力 同じ配列を持つ遺伝 子の発現を抑制する現象を言う。 RNAiにより ΙΚΚ αの発現を阻害する物質の具体例 としては、下記に説明するような siRNA又は shRNA等が挙げられる。
[0018] siRNAとは short interfering RNAの略称であり、約 21〜23塩基程度の長さの二本 鎖 RNAをいう。 siRNAは RNAiを引き起こすことができる限り、どのような形態のものでも よぐ例えば、化学合成もしくは生化学的合成、又は生物体内の合成で得られた siRN A、ある ヽは約 40塩基以上の二本鎖 RNAが体内で分解されてできた 10塩基対以上 の短鎖二本鎖 RNA等であればよい。 siRNAの配列と、 IKK αの mRNAの部分配列と は 100%—致することが好まし 、が、必ずしも 100%—致して 、なくてもょ 、。
[0019] siRNAの塩基配列と、 IKK a遺伝子の塩基配列との間で相同性のある領域は、 IK Κ α遺伝子の翻訳開始領域を含まないことが好ましい。相同性を有する配列は、 ΙΚ K a遺伝子の翻訳開始領域から 20塩基離れて ヽることが好ましぐ 70塩基離れて ヽ ることがより好ましい。相同性を有する配列としては、例えば、 ΙΚΚ α遺伝子の 3'末端 付近の配列でもよい。
[0020] RNAiにより IKK aの発現を阻害する物質としては、 siRNAを生成する約 40塩基以 上の dsRNA等を用いてもよい。例えば、 ΙΚΚ α遺伝子の核酸配列の一部に対して約 70%以上、好ましくは 75%以上、より好ましくは 80%以上、より好ましくは 85%以上 、さらに好ましくは 90%以上、特に好ましくは 95%以上、最も好ましくは 100%の相 同性を有する配列を含む、二本鎖部分を含む RNA又はその改変体を使用することが できる。相同性を有する配列部分は、通常は、少なくとも 15ヌクレオチド以上であり、 好ましくは約 19ヌクレオチド以上であり、より好ましくは少なくとも 20ヌクレオチド以上
であり、さらに好ましくは 21ヌクレオチド以上である。
[0021] RNAiにより ΙΚΚ αの発現を阻害する物質としては、 3'末端に突出部を有する短い ヘアピン構造から成る shRNA (short hairpin RNA)を使用することもできる。 shRNAと は、一本鎖 RNAで部分的に回文状の塩基配列を含むことにより、分子内で二本鎖 構造をとり、ヘアピンのような構造となる約 20塩基対以上の分子のことである。また、 s hRNAとしては 3'突出末端を有するのが好ましい。二本鎖部分の長さは特に限定され ないが、好ましくは 10ヌクレオチド以上であり、より好ましくは 20ヌクレオチド以上であ る。ここで、 3'突出末端は、好ましくは DNAであり、より好ましくは少なくとも 2ヌクレオ チド以上の DNAであり、さらに好ましくは 2〜4ヌクレオチドの DNAである。
[0022] RNAiにより ΙΚΚ αの発現を阻害する物質は、人工的に化学合成してもよいし、セン ス鎖及びアンチセンス鎖の DNA配列を逆向きに連結したヘアピン構造の DNAを Τ7 R ΝΑポリメラーゼによってインビトロで RNAを合成することによって作製してもよい。イン ビトロで合成する場合は、 Τ7 RNAポリメラーゼ及び Τ7プロモーターを用いて、铸型 D ΝΑからアンチセンス及びセンスの RNAを合成することができる。これらをインビトロで アニーリングした後、細胞に導入すると、 RNAiが引き起こされ、 ΙΚΚ αの発現が抑制 される。細胞への導入は、例えば、リン酸カルシウム法、又は各種のトランスフエクショ ン試薬(例えば、 oligofectamine、 Lipofectamine及び lipofection等)を用いた方法等に より行うことができる。
[0023] RNAiにより IKK aの発現を阻害する物質としては上述の siRNA又は shRNAをコード する核酸配列を含む発現ベクターを用いてもょ ヽ。さらに該発現ベクターを含む細胞 を用いてもよい。上記した発現ベクターや細胞の種類は特に限定されないが、既に 医薬として用いられて 、る発現ベクターや細胞が好ま 、。
[0024] 本発明では、 IKK a、又は IKK aの発現を増大させる物質をインターフェロン α 産生誘導剤として用いることができる。 ΙΚΚ αの発現を増大させる物質としては、転 写因子等のタンパク質や低分子化合物などが挙げられる。
[0025] 本発明のインターフ ロン α制御剤の投与経路は特に限定されず、経口投与又 は非経口投与 (例えば、静脈内投与、筋肉内投与、皮下投与、皮内投与、粘膜投与 、直腸内投与、膣内投与、患部への局所投与、皮膚投与等)のいずれの投与経路に
より投与してもよい。経口投与に適する製剤形態としては、固形又は液体の形態が挙 げられ、非経口投与に適する製剤形態としては、注射剤、点滴剤、坐剤、外用剤、点 眼剤、点鼻剤等の形態が挙げられる。本発明のインターフ ロン α制御剤は徐放 剤の製剤形態であってもよい。本発明のインターフェロン—ひ制御剤は、その製剤形 態により必要に応じて薬学的に許容可能な添加剤が加えられていてもよい。薬学的 に許容可能な添加剤の具体例としては、例えば、賦形剤、結合剤、崩壊剤、滑沢剤 、抗酸化剤、保存剤、安定化剤、等張化剤、着色剤、矯味剤、希釈剤、乳化剤、懸 濁化剤、溶媒、フィラー、増量剤、緩衝剤、送達ビヒクル、希釈剤、キャリア、賦形剤 及び Ζ又は薬学的アジュバント等が挙げられる。
[0026] 経口用の固形製剤形態の本発明のインターフェロン α制御剤は、例えば、有効 成分である IKK aの阻害剤、 IKK a、又は IKK aの発現を増大させる物質に賦形 剤を加え、さらに必要に応じて結合剤、崩壊剤、滑沢剤、着色剤、又は矯味剤などの 製剤用添加物を加えた後、常法により錠剤、顆粒剤、散剤、カプセル剤として調製す ることができる。経口用の液体製剤形態の本発明のインターフェロン α制御剤は、 有効成分である IKK aの阻害剤、 IKK a、又は IKK aの発現を増大させる物質に 矯味剤、安定化剤、又は保存剤など製剤用添加物の 1種又は 2種以上を加え、常法 により内服液剤、シロップ剤、エリキシル剤等として調製することができる。
[0027] 本発明のインターフェロン α制御剤を液体製剤として処方するために使用される 溶媒は、水性又は非水性のいずれでもよい。液体製剤は当該分野において周知の 方法により調製することができる。例えば、注射剤は、生理食塩水、 PBSのような緩衝 液、滅菌水等の溶剤に溶解した後、フィルタ一等で濾過滅菌し、次いで無菌容器 (例 えば、アンプル等)に充填することにより調製することができる。この注射剤には、必 要に応じて、慣用の薬学的キャリアを含めてもよい。また、非侵襲的なカテーテルを 用いる投与方法を用いてもよい。本発明で用いることができるキャリアとしては、中性 緩衝化生理食塩水、又は血清アルブミンを含む生理食塩水等が挙げられる。
[0028] IKK aの siRNAまたは siRNA発現ベクターなど遺伝子送達の種類に関しては、適用 される細胞内で IKK aの siRNAをコードする RNAまたは siRNA発現ベクターの発現を 得る限り特に方法は限定されるものではなぐ例えば、ウィルスベクター、リボソームを
用いた遺伝子導入を用いる事が可能である。ウィルスベクターとしては、例えば、レト ロウィルス、ワクシニアウィルス、アデノウイルス、シンリンセムリキウイルス等の動物ゥ ィルスが挙げられる。
[0029] RNAiにより ΙΚΚ αの発現を阻害する物質は、細胞に直接注入してもよい。
[0030] 本発明のインターフ ロン oc制御剤の投与量は、使用目的、疾患の重篤度、患 者の年齢、体重、性別、既往歴、又は有効成分として用いる物質の種類等を考慮し て、当業者が決定することができる。本発明のインターフェロン—ひ制御剤の投与量 は、例えば、有効成分量として、成人一人当たり、約 0.1 ng〜約 100 mg/kg、好ましく は約 1 ng〜約 10 mgであり、ウィルスベクター又は非ウィルスベクターとして投与され る場合は、通常、 0.0001〜100 mg、好ましくは 0.001〜10 mg、より好ましくは 0.01〜1 mgで 3Dる。
[0031] 本発明のインターフェロン a制御剤の投与頻度は、例えば、一日一回〜数ケ月 に 1回であればよい。 RNAiにより ΙΚΚ αの発現を阻害する物質を用いる場合は一般 に投与後 1〜3日間効果が見られるので、毎日〜 3日に 1回の頻度で投与することが 好ましい。発現ベクターを用いる場合には 1週間に 1回程度の投与が適する場合もあ る。

血管炎、反応性脊椎関節症、強直性脊椎炎、炎症性慢性腎状態 (糸球体腎炎、ル 一ブス腎炎、膜性腎炎等)、リウマチ熱、シエーダレン症候群、ベーチェット病、甲状 腺炎、 I型糖尿病、皮膚筋炎、慢性活動性肝炎、重症筋無力症、グレーヴス病、多発 性硬化症、原発性胆汁性肝硬変、自己免疫性血液疾患 (溶血性貧血、真性赤血球 性貧血、特発性血小板減少症、再生不良性貧血等)、橋本病、ぶどう膜炎、接触皮 膚炎、乾癬、川崎病、 I型アレルギー反応が関与する疾患 (アレルギー性喘息、アトピ 一性皮膚炎、蓴麻疹、アレルギー性結膜炎、花粉症、湿疹、食物過敏症、アレルギ 一性鼻炎等)、ショック (敗血性ショック、アナフィラキシー性ショック、成人型呼吸窮迫 症候群等)、サルコイドーシス、ゥ ゲナー肉芽腫症、ホジキン病、癌 (肺癌、胃癌、結 腸癌、肝癌等)が例示される。また、種々の微生物感染症、特に各種ウィルスによる 急性(たとえばインフルエンザウイルス、単純へルぺスウィルス、水疱性口内炎ウィル スなど)、慢性 (たとえば B型肝炎ウィルス、 C型肝炎肝炎ウィルスなど)感染症、各種 細菌感染症、各種真菌感染症、各種寄生虫感染症等も例示される。

実施例
[0035] 実施例 1 : TLR7,9刺激による骨髄由来 in vitro榭状細胞力 の IFN- a産生誘導にお ける IKK aの必要'性
ΙΚΚ α +/-マウス同士を交配させることにより、胎生 13. 5-15. 5日の胎児を得た。こ の胎児の中から、 IKK a +/+,-/-の遺伝形質を有するマウスを PCRにて選別したのち 、胎児肝細胞を採取した。さらに、胎児肝細胞を放射線照射野生型 C57BL6-CD45.1 マウスに静注したのち 6-10週後に IKK a +/+ある!/、は IKK a -/-骨髄キメラマウスとし て使用した。
[0036] このキメラマウス骨髄細胞を 5xl06個/ mlで、 10%FCS, 100ng/ml human Flt31igand
(Peprotech)を含む RPMI1640培地で 6〜8日間培養することにより、 Flt3L誘導骨髄榭 状細胞 (Flt3L BM DC)を調整した。この Flt3L BM DCを非刺激 (med)あるいは 100ng/ ml Bacterial lipopeptide (BLP, Invivogen), 50 μ g/mi polyinosinic- polycytidylic acid、 poly (I:C), Amersham) (但し、図 lcでは、 lipofectamine 2000(Invitrogen)と混合した最
終濃度 25 μ g/ml poly(I:C)を使用), 100ng/ml Salmonella Minnesota Re595由来 LPS (Sigma), ΙΟΟηΜ R848 (Invivogen),合成 DNA (1 ^ M 1668: 5 '-tccatgacgttcctgatgct-3 ' (配列番号 1) , 3 ^ M D19:5'-ggTGCATCGATGCAgggggG-3' (配列番号 2)、小文 字 ίま phosphorothioate DNA,大文子 ίま phosphodiester DNA), Lipofectamine 2000 (In vitrogen)と混合した最終濃度 20 μ g/ml polyuridylic acid (PolyU)により 20- 24時間刺 激し、培養上清中の IL-12p40、 TNF- αの量を Genzyme Techne社の ELISAにて、 IFN - αの量を PBL社の ELISAにて測定を行った(図 la、 b及び c)。
[0037] 野生型 Flt3L誘導骨髄榭状細胞 (Flt3L BM DC)は、細菌リポペプチド (BLP, TLR2リ ガンド)、ポリ(I:C) (TLR3リガンド)、 LPS (TLR4リガンド)、 R848 (TLR7リガンド)及び 0 DN1668 (TLR9リガンド)に応答して IL- 12p40を産生した。 ΙΚΚ α - /- Flt3L BM DCも I L-12p40を産生することによってこれらの TLRァゴ-ストに応答した。 TLR7又は TLR9 シグナル伝達による IL-12p40の誘導量は僅かに減少した力 刺激後は有意に増加し た(図 la)。 TLR7又は TLR9シグナル伝達による TNF- αの誘導量は野生型及び IKK a -/- Flt3L BM DCにおいて有意差はみられなかった(図 lb)。また、 D/A型 CpG D NA(D19)は、野生型 Flt3L BM DCにおける IFN- α産生を誘導できた力 この誘導は 、 IKK a -/- Flt3L BM DCでは大きく障害されていた(図 lc)。 Poly U誘導 IFN- α産 生も ΙΚΚ α - /- Flt3L BM DCでは大きく障害されていた(図 lc)。一方、 poly (I:C)刺激 による IFN- a産生は正常であった(図 lc)。
[0038] また、 CD40と B220の発現を FACSにより解析を行った(図 ld)。 B220+ (PDC)及び B2 20— (通常の DC)の 2種類の榭状細胞の割合は正常であり、各々の DCの TLR7及び T LR9による CD40の発現誘導は、 ΙΚΚ α +/+及び IKK a -/- Flt3L BM DCの間で有意 な差を認めな力つた(図 ld)。
[0039] Flt3L BM DCにおける IFN— a , IFN— β , IL— 12p40遺伝子の発現を解析するため、 F lt3L BM DCを LPS、 D19または PolyUにより刺激したのち、 RNAを調製し、 IFN- α , IF N- j8 , IL-12p40遺伝子をプローブとして Northern blot analysisを行った(図 le)。 TLR 7又は TLR9シグナル伝達による IFN- α遺伝子誘導は、 ΙΚΚ α - /- Flt3L BM DCでは 大きく損なわれたが、 IFN- β及び IL-12p40遺伝子の TLR7又は TLR9誘導発現上昇 は、野生型及び ΙΚΚ α -/- Flt3L BM DCの間で同等であった(図 le)。この結果より、 I
KK α力 in vitro榭状細胞にお!、て TLR7又は TLR9に仲介された IFN- a誘導に必須 であることが示された。
[0040] 実施例 2: TLR7,9刺激による in vivo榭状細胞からの IFN- a産生誘導における IKK a の必要性
キメラマウスの骨髄細胞における CDl lc, B220の発現を FACSにて解析した (図 2a)。 その結果、 ΙΚΚ α +/+及び IKK « -/-キメラは共に、 CDl lc+B220+細胞を骨髄細胞中 に含んでいた。
[0041] また、骨髄細胞を lxlO6個/ mlで、 10%FCSを含む RPMI 1640培地で非刺激、ある いは lOOnM R848, 1 μ Μ 1668, 3 ^ M D19, Lipofectamine 2000 (Invitrogen)と混合し た最終濃度 g/ml polyUの刺激後 20〜24時間の培養上清中の IL-12p40の量を G enzyme Techne社の ELISAにて、 IFN- αの量を PBL社の ELISAにて測定を行った(図 2b)0 TLR7又はTLR9誘導IFN- Q;産生は、 ΙΚΚ α - /-キメラ骨髄細胞において大きく 減少していた。一方、 IL_12p40誘導は軽度減少していた(図 2b)。
[0042] また、キメラマウスの脾臓細胞から、磁気ビーズを用いたソーティング (MACS)により CDl lc陽性細胞を調製し、その CDl lc, B220の発現を FACSにて解析した (図 2c)。 C Dl lc陽性細胞における B220陽性細胞の数は、 ΙΚΚ α+Λ及び ΙΚΚ α -/-キメラ脾臓 細胞の間で同等であった(図 2c)。
[0043] また、この CDl lc陽性脾細胞を骨髄細胞と同様の条件で刺激し、 IL-12p40,IFN- αの産生量を ELISAにより測定した (図 2d)。 ΙΚΚ α - /- CDl lc陽性脾細胞は、 ΙΚΚ α +/+ CDl lc陽性脾細胞よりも IL-12p40の産生量が軽度減少していた力 \KK a ~/~ C Dl lc陽性脾細胞からの TLR7/9で誘導される IFN— aの産生は大きく低下していた( 図 2d)。
[0044] キメラマウスに 1匹あたり 50nmolの R848を静注し、静注後 1,3,6時間に採血を行い、 血清中の IFN- a濃度を ELISAにより測定した(図 2e)。血清中の IFN- a濃度の TLR- 7リガンド(R848)によって誘導される上昇は、 PDCからの産生に依存している力 IKK a -/-キメラで大きく低下していた(図 2e)。
[0045] 実施例 3 : MyD88依存性経路による IFN- aプロモーター活性化における IKK aの機 能
(1)プラスミドの調製
pUNO-MyD88, pUNO— TRAF6: Invivogenより購入した。
pUNO: pUNO- TRAF6を Agel+Nhelで切断し、インサートを除去後、 self annealingを 行い、コントロールベクターとして使用した。
pEF-BOS-FLAG-mIRF-7:プライマー 5し GGGTCGACATGGACTACAAAGACGA TGACGACAAGGCTGAAGTGAGGGGGGTCCAGCGA-3' (配列番号 3)と 5し GGG CGGCCGCTCAAGGCCACTGACCCAGGTCCATGAG-3' (配列番号 4)による PCR にて、 GM-CSFによって誘導されたマウス骨髄由来榭状細胞の cDNAライブラリーか ら、 IRF7 cDNAを増幅し、プラスミド pEF—BOS (S. Mizushima, S. Nagata. Nucleic Acid s Res. 18:5322, 1990.)の Sail- Notl部位に挿入した。
[0046] pSR a -6myc-mIKK a:徳島大学松本満先生より供与を受けた。野生型 IKK a (IKK a -WT)の発現ベクターとして使用した。
pSR a -6myc-mIKK a (K44A): pSR a - 6myc- mIKK aを基に、 44番目のリジン残基 をァラニン残基に変換するように in vitro mutagenesisを行い、キナーゼ活性欠損 IKK a (IKK a— KD)の発現ベクターとして使用した。
pSR a -6myc: pSR a - 6myc- mIKK aから mIKK aを EcoRI+XhoIにより切断、末端を 修復し、 self annealingを行った。
[0047] pIFN- a 4- luc: IFN- a 4プロモーター領域をセンスプライマー 5し CCCCCACACT TTACTTTTTTGAC AGAA-3 ' (配列番号 5)とアンチセンスプライマー 5'- TACAGG TTCTCTGAGAGCCTGCTGTGT-3 ' (配列番号 6)にて C57BL6マウス DNAから PCR にて増幅を行った。このプロモーター領域を pGL3 (Promega)の Xhol-Hindlll部位に サブクロー-ングすることにより pIFN- a 4-lucを作成した。
pELAMl-luc: Dr. D. T. Golenbockより供与。 R丄. Delude et al. J. Immunol. 161:30 01, 1998.
pIFN- j8 -luc: Sato et al. Immunity 13:539, 2000.に従い、 PCRで IFN— βプロモー ター領域を増幅し、 pGL3 (Promega)の Xhol-Hindlll部位にサブクロー-ングすること により pIFN- β -lucを作成した。
pRL-TK: Promegaより購入した。
[0048] (2)ルシフェラーゼアツセィ(図 3a, b, f)
ヒト腎臓由来細胞株 293Tを 2xl05個/ mlの濃度で 0.35ml/well, 24穴プレート上で 10 %FCSを含む DMEM培地で培養した。 24時間後、 pUNO、 pUNO- MyD88(Invivogen), pUNO- TRAF6, pEF- BOS- Flag- mIRF- 7, pSR a - 6myc, pSR a - 6myc- mIKK a , pSR a -6myc-mIKK a (K44A)を種々の組み合わせで、レポーター遺伝子 pIFN- a 4- luc ( 図 3a及び b), pELAMl-luc (図 3f)あるいは pIFN— j8— luc (図 3f)、および pRL— TK(Pro mega)と共に添カ卩し、 lipofectamine 2000(Invivogen)を用いて、遺伝子導入を行った。 1 8- 24時間後、細胞抽出物を調製し、 Dua卜 Luciferase Reporter Assay System(Promeg a), 20/20nLuminometer (Turner Biosystem)を用いて luciferase活性を測定した。遺伝 子導入効率を補正するために、得られた Firefly luciferase活性を Renilla luciferase活 性で除することにより、相対活性値とした。最終的に実験データは空ベクターのみで 遺伝子導入した相対活性値を 1として、 fold inductionで示した。
[0049] TLRアダプター MyD88だけの発現では IFN- aプロモーターを活性化できな!/、が、 プロモーターの IRF-7誘導活性ィ匕を相乗的に増大することができる(図 3a)。 IKK a - K Dの発現は、 IRF-7のみによる活性ィ匕に影響しないが(図 3a)、 IRF-7共存下で認めら れる MyD88による相乗効果を阻害できた(図 3a)。一方、 IKK α -WTの発現は、 MyD8 8、 IRF-7の相乗効果をわずかながら増強した(図 3a)。 MyD88と同様、別のアダプタ 一分子である TRAF6も、 IRF-7と相乗的に IFN- aプロモーターを活性化することがで きる(図 3b)。 ΙΚΚ α - KDは、この TRAF6による誘導相乗効果も阻害できた。 MyD88も NF- κ B又は IFN- βプロモーターを活性化できるが、この活性は IKK a -KDの発現で は阻害されなかった(図 3f)。従って、 ΙΚΚ αは、そのキナーゼ活性を介して、 MyD88 又は TRAF6と相乗して IFN- aプロモーターを活性化する IRF7の能力に関与して!/、る ことが示された。

(3— 1) 293T細胞における IKK aと IRF7の会合 (図 3c)
60mm dishl枚あたり 2xl06個の 293Tを 10%FCSを含む DMEM培地でー晚培養した 。次に示されているプラスミドを全量 5 μ gとして、 lipofectamine 2000(Invivogen)を用 いて遺伝子導入を行った。遺伝子導入 20時間後、細胞を回収し、溶解液 (1% Nonid
et P-40, 150mM NaCl, 5mM EDTA, ImM PMSF, 20 μ g/ml aprotinin, lOmM NaF, 1 OmM j8 -glycerophosphate, ImM Na VO ,及び 20mM Tris- HC1, pH8.0)にて細胞抽

出を行った。 protein G- Sepharose4FF beadsと 1時間混合した後、上清をさらに 6 /z g/ ml anti-Flag M2 mAb (Sigma— Aldnch)める ヽ ί¾5 μ g/mi anti— Myc— tag clone PL14 mAb (MBL, Japan)を結合させた protein G- Sepharose4FF beadsと 4°Cで 12時間混合 を行った。ビーズを 3回洗浄した後、 Laemmli液にて 100°C5分で溶出したものを 10%S DSポリアクリルアミドゲルにて電気泳動し、 PVDFメンブレンに転写した。次にメンブレ ンをビォチン標識 antト Flag M2 mAb (Sigma-Aldrich)あるいはビォチン標識 antト M yc-tag clone PL14 mAb (MBL, Japan)と室温で 1時間混合した。 TBS- Tバッファー(0. l%Tween20, 137mM NaCl, 2.6mM KC1, 25mM Tris— HC1, pH7.0)で洗浄後、さらに s treptavidin-HRP conjugate (Amersham bioscience)にて室温 30分混合した。さらに洗 浄後、 ECL system (PerkinElmer Life sciences)を用いて、バンドの検出を行った。
[0051] その結果、 IKK aは IRF-7と一緒に免疫沈降することが確認された(図 3c)。 MyD88 又は TRAF6の共発現は、 IRF7及び IKK aの相互作用を有意に増強できた(図 3c)。
[0052] (3 - 2) Flt3L誘導 BM DCにおける IKKaと IRF-7の会合(図 3d)
野生型 Flt3L BM DCを調製し、ゥサギ抗 IRF- 7抗体 (United States Biological),その コントロールとして正常ゥサギ抗体 IgG(Santa Cruz Biotechnology),抗 IKK α抗体 (M- 280, Santa Cruz Biotechnology)を用いて、図 3cと同様に、細胞抽出物の作成、免疫 沈降および Western blotを行った。 IRF-7のバンドの検出には HRP標識ロバ抗ゥサギ I gtj抗体 (Amersham Bioscience)を用 ヽた。

[0054] (3— 3) GM- CSF誘導 BMDCにおける ΙΚΚ αと IRF7の会合 (図 3e)
ΙΚΚ α +/+あるいは ΙΚΚ α - /-キメラマウスの骨髄細胞を lxlO6個/ mlで、 10%FCS, 1 0ng/ml GM-CSF (R&D)を含む RPMI1640培地で 6〜8日間培養することにより、 GM- C SF誘導骨髄榭状細胞 (GM-CSF BM DC)を調整した。さら〖こ、レンチウィルスベクター pCSn-EF- FLAG- mIRF7-IRES2- Venusを用いて、 GM-CSF BM DCに IRF7を強制発 現させた。 pCSII-EF-FLAG- mIRF7-IRES2-Venusは、理化学研究所三好浩之先生、 宫脇敦史先生より供与されたレンチウィルスベクター PCSII-EF-MCS-IRES2- Venus
に、実施例 3 ( 1)で作成した IRF7 cDNAを挿入することにより作成した。この GM- CSF —DCを、 Antト FLAG M2 mAb、そのコントロールとして anti—myc— tag clone PL14 mAb 、抗 IRF7抗体、及び抗 IKK α抗体 (M- 280, Santa Cruz Biotechnology)を用いて、図 3 cと同様に、細胞抽出物の作成、免疫沈降および Western blotを行った。
[0055] その結果、 IRF7と IKK aの会合が検出された(図 3e)。
[0056] 実施例 4: IKK aによる GST- IRF7のリン酸ィ匕 (図 4)
( 1)プラスミドの調製
PGST-IRF7: pEF- BOS- FLAG- mIRF- 7の 422- 457番目のアミノ酸に相当する領域 を PCRにて増幅し、 pGEX-5X-l (Amersham Biosciences)にサブクロー-ングを行つ た。
pGST-I κ Β α :マウス I κ Β αの 5- 55番目のアミノ酸に相当する領域を PCRにて増幅 し、 pGEX- 5Χ- 1 (Amersham Biosciences)にサブクロー-ングを行った。

pGEX- 5X- 1 , pGST- IRF7および pGST- 1 κ αを大腸菌 BL21に導入し、 Glutathion e sepharose 4B aftinity chromatography (Amersham Biosciences) 用いて、 GS fグン パク(コントロール)、 GST融合 IRF7および GST融合 pGST- 1 κ Β αを精製し、基質とし て使用した。 293Τ細胞に pSR a -6myc (コントロールベクター)、 pSR a -6myc-mIKK α、 pSR a -6myc-mIKK a (K44A)を導入し、細胞抽出物を作成し、 PL14を用いて免 疫沈降を行った。この免疫沈降物と基質 2 μ gを混合し、 20mM HEPES-NaOH(pH7.6 ), 10mM MgCl , 2mM MnCl , 50mM NaCl, O. lmM Na VO , lOmM β -glycerophosph

ate, lOmM PNPP, ImM DTT, lOmM NaF, 50 ^ M ATP, 370kBq[ y— 32P]ATPの存在 下で 37度 30分反応させた(図 4a)。反応後、 10%SDSポリアクリルアミドゲルにて電気 泳動、引き続きオートラジオグラフィーを行った。また、組み換えヒト ΙΚΚ α (Lake Placi d) lOOng (60units/mg protein)を用いて同様の反応を行った(図 4b)。
[0058] その結果、 IRF7は、 IKK aによりリン酸ィ匕されることが分力つた(図 4a)。
産業上の利用可能性
[0059] 本発明により、インターフェロン aの産生を制御することが可能になった。本発明 のインターフェロン α制御剤は、インターフェロン αの産生の抑制又は誘導によ
り治療や予防に有効となる各種疾患の治療や予防のために用いることができる。 図面の簡単な説明

[1] IKK aの阻害剤を含む、インターフェロン a産生抑制剤。
[2] IKK aの阻害剤が、 IKK aの発現を阻害する物質、又はキナーゼ活性を欠損する I ΚΚ α変異体である、請求項 1に記載のインターフェロン α産生抑制剤。
[3] 細胞内の IKK a活性を阻害することを含む、細胞からのインターフェロン aの産生 を抑制する方法。
[4] IKK a、又は IKK aの発現を増大させる物質を含む、インターフェロン a産生誘 導剤。
[5] 細胞に IKK a、又は IKK aの発現を増大させる物質を投与することを含む、細胞か らのインターフェロン αの産生を誘導する方法。
