Abstract
PURPOSE:To efficiently secrete a protein having sufficient lysozyme activity into a culture medium by recombinant DNA technique, by using a gene coding a variant polypeptide obtained by changing part of amino acid sequence of human lysozyme. CONSTITUTION:A variant human lysozyme gene coding variant polypeptide obtained by replacing cysteine in 77th and 95th site of amino acid sequence of natural human lysozyme with alanine is prepared and the gene is inserted into a proper manifestation vector together with a nucleotide sequence (preferably obtained by giving modification to nucleotide sequence coding signal peptide of egg white lysozyme) coding signal peptide and host is transformed using the resultant manifestation vector (e.g. plasmid pERI 8716) and the resultant transformant is cultivated and a variant human lysozyme secreted into a culture medium is separated and obtained.
