Abstract
PURPOSE:To easily produce the titled compound useful as a raw material for pharmaceuticals, on an industrial scale, by using an easily available 5- halogenopyrimidine derivative as a starting raw material and reacting the derivative with an alkylmagnesium halide in the presence of a metal complex catalyst. CONSTITUTION:The objective 5-alkylpyrimidine derivative of formula III (e.g. 5-methyluridine) is produced by reacting a 5-halogenopyrimidine derivative of formula I [R<1> is H or (protected) pentose; X<1> is halogen] with an alkylmagnesium halide of formula II (R<2> is alkyl or aryl; X<2> is halogen which is the same as or different from X<1>) in the presence of a metal complex catalyst [e.g. chlorinated diphenylphosphinoethane nickel (II)]. The halogenopyrimidine derivative of formula I can be easily produced by reacting a pyrimidine derivative of formula IV with an anionic halogenation agent.
