PURPOSE:To readily produce the titled substance in a large quantity, by inserting a gene coding PDG prepared by a bacterium of genus Bacillus into a plasmid vector capable of multiplying in a bacterium of genus Bacillus and subsequently introducing the inserted vector into a bacterium of genus Bacillus. CONSTITUTION:A plasmid pSDR 0023 containing a gene coding PGD (3- phosphoglycerate dehydrogenase) is prepared from a gene DNA of bacillus amyloliquefaciens SD-31 (FERM No.6840). A 0.9 mega dalton ECoRI fragment of the plasmid is recloned in a Bacillus subtilis pLS 15 to give a plasmid pSDM 00350. The plasmid is introduced into a genus Bacillus host to give a transformed strain Bacillus amyloliquefaciens SD-1021 or Bacillus subtilis SD-1022. These strains are cultured in a medium containing anthranilic acid, indole or a salt thereof to provide L-tryptophane.
