Abstract
PURPOSE:To easily produce the titled substance in large quantity, by inserting a structural gene of a BanI restriction endonuclease originate from Bacillus aneurinolyticus into a vector plasmid pBR322 and introducing the plasmid into E.coli. CONSTITUTION:A chromosome DNA containing BanI restriction endonuclease gene originated from Bacillus aneurinolyticus IAM1077 is prepared and introduced into BamHI site of pBR322 to obtain a plasmid pBAN11 of 8kb. The plasmid is introduced into Escherichia coli HB101 strain to obtain a transformed Escherichia coli HB101 (pBAN11) (FERM P-9424). The strain is cultured in a nutrient medium to obtain BanI restriction endonuclease without necessitating the removal of BanIII and BanII restriction endonuclease.
