Description
本発明は歯磨組成物、さらに詳しくは、炭酸カ
ルシウム、リン酸カルシウムのようなカルシウム
イオンを溶出する基剤を用いる歯磨組成物におい
て、アラントインまたはアラントイン誘導体を安
定に配合した歯磨組成物に関する。
アラントインおよびアラントイン誘導体(以
下、単にアラントインという)は壊死組織の除去
作用を有し、かつ、それ自体低毒性、無刺激性
で、抗刺激性、抗アレルギー性を有することから
医薬品、化粧品の薬効剤として広く使用されてお
り、歯磨組成物においても歯槽膿漏、歯肉炎など
の予防を目的として配合されている。
しかして、従来、アラントインを歯磨組成物に
配合する場合、基剤としては、もつぱら、水酸化
アルミニウムやピロリン酸カルシウムなどのよう
なカルシウムイオンを溶出しないものが望ましい
とされており、通常の歯磨組成物によく用いられ
る炭酸カルシウムやリン酸カルシウムなどのよう
なカルシウムイオンを溶出する基剤を用いると、
経日により有効アラントイン量(可溶性アラント
イン量)が著しく減少し、アラントインの配合効
果が損われる。これは、アラントインが、経日に
より基剤から溶出したカルシウムイオンと何らか
の結合を生じ、不溶性もしくは難溶性物質を形成
することによるものと考えられる。
本発明者らは、通常の歯磨組成物によく用いら
れるカルシウムイオンを溶出する基剤を用いた歯
磨組成物にアラントインを安定に配合するために
鋭意研究を重ねた結果、モノフルオロリン酸塩あ
るいはポリリン酸塩を共存させるとアラントイン
の安定化が図れることを見出し、本発明を完成す
るにいたつた。
すなわち、本発明は、カルシウムイオンを溶出
する基剤を用いるアラントイン含有歯磨組成物に
おいて、アラントイン安定化剤としてモノフルオ
ロリン酸塩およびポリリン酸塩から選ばれる化合
物を配合したことを特徴とする歯磨組成物を提供
するものであり、本発明の歯磨組成物はカルシウ
ムイオンを溶出する基剤を用いても、経日による
有効アラントイン量の減少が著しく抑制され、歯
槽膿漏、歯肉炎の予防にきわめてすぐれた効果を
発揮する。
このモノフルオロリン酸塩あるいはポリリン酸
塩によるアラントイン安定化の作用機序は明確で
はないが、これらが、炭酸カルシウムあるいはリ
ン酸カルシウムのような基剤の粒子表面に吸着さ
れ、あたかも基剤を被覆したような形となつて、
基剤からのカルシウムイオンの溶出を抑制するた
めに歯磨系内に溶出されるカルシウムイオン量が
著しく抑制されると共に、溶出した少量のカルシ
ウムイオンは歯磨系内に溶存するモノフルオロリ
ン酸塩、ポリリン酸塩のキレート作用によつてイ
オンの形を失ない、アラントインとの結合が弱ま
つてアラントインの不溶化もしくは難溶化が抑制
されることによるものと推定される。
本発明で用いるモノフルオロリン酸塩として
は、一般式:M2PO3F(式中、Mはナトリウム、
カリウムなどのアルカリ金属またはアンモニウム
カチオンを意味する。以下同様)、好ましくは、
モノフルオロリン酸ナトリウムが挙げられ、歯磨
組成物に対して0.05%(重量%、以下同様)以上
配合することが好ましい。なお、多量に配合する
と歯磨組成物の物性が低下するので0.05〜2%、
好ましくは、0.1〜0.76%(フツ素として約130〜
約1000ppm)の範囲が望ましい。
また、ポリリン酸塩としては、ピロリン酸塩
(M4P2O7またはM2H2P2O7)、トリポリリン酸塩
(M5P3O10)、トリメタリン酸塩(M3P3O9)、テト
ラメタリン酸塩(M4P4O12)、ヘキサメタリン酸
塩(M6P6O18)、不溶性メタリン酸塩
〔(MPO3)o〕またはこれらの混合物などが挙げら
れ、歯磨組成物に対して0.05%以上配合すること
が好ましい。なお、多量に配合すると歯磨組成物
の物性が低下し、また、内面塗装のないアルミチ
ユーブに充填するような場合、アルミチユーブの
内面腐蝕が促進されるので、例えば、ピロリン酸
ナトリウムでは0.05〜2%、トリポリリン酸ナト
リウムでは0.05〜2%、不溶性メタリン酸ナトリ
ウムでは0.1〜4%の範囲が好ましい。
用いるカルシウムを溶出する基剤としては、炭
酸カルシウム、第二リン酸カルシウム(二水化
物、無水物)などが挙げられる。
また、アラントインとしてはアラントインまた
はアルミニウムアラントイネートのようなアラン
トイン誘導体が挙げられ、これらの配合割合は歯
磨組成物の処方に応じて適宜選択されるが、通
常、0.05〜2%の範囲が好ましい。
本発明の歯磨組成物は常法に従つて製造でき、
練歯磨、歯磨軟膏、水歯磨などの剤形とすること
ができる。用いる他の配合成分としては通常の歯
磨組成物に用いられるものでよく、例えば、発泡
剤として、ラウリル硫酸ナトリウムのようなアル
キル硫酸ナトリウム、アシルサルコシンナトリウ
ム、α―オレフインスルホネート、シヨ糖脂肪酸
エステルなど;粘結剤として、カルボキシメチル
セルロースナトリウム、カラギーナン、アルギン
酸ナトリウム、ベントナイト、無水ケイ酸など;

前記実施例1および2で得られた歯磨組成物に
おける有効アラントイン量の経日変化を試験した
結果をつぎの第1表に示す。
試験は、各経日における歯磨組成物試料中の有
効アラントイン量を測定し、製造直後の有効アラ
ントイン量を100%とし、各経日において残存す
る有効アラントイン量の割合(%)を算出して行
なつた。なお、対照として、実施例1および2の
処方からモノフルオロリン酸ナトリウムを除いた
歯磨組成物を用いた(比較例1および2)。
有効アラントイン量の測定はつぎのとおりであ
る。
試料約1gを精秤し、0.01M塩化バリウム液
(日局分析用標準液、以下同じ)50mlを加え、80
℃で30分撹拌後、遠心分離(10000r.p.m.、20
分)する。この上澄液にエーテル70mlを加えて振
とうし、その水層をとり、水層中のエーテルを揮
散させた後、50mlにメスアツプして試料液とす
る。別にアラントインクロルヒドロキシアルミニ
ウム(粧原基)0.5mgを精秤し、0.01M塩化バリ
ウム液で50mlにメスアツプし、これを標準液とす
る。試料液および標準液70μlづつを、高速液体
クロマトグラフイーに付し、標準液のピークと同
一保持時間を有する試料液のピークの高さ
(HT)および標準液のピークの高さ(HS)を求
め、次式から有効アラントイン量(mg)を算出す
る。
