Abstract

PURPOSE: To carry out the anaerobic cultivation of microorganisms easily, by the presence of an oxygen absorbing agent in a chamber separated from a culture medium through a thin film having the water impermeability and oxygen permeability.
CONSTITUTION: A thin film, e.g. polypropylene or polyethylene, is used as a material having the water impermeability and oxygen permeability. A culture medium is sealed in a container consisting of the material, sterilized and inoculated with a microorganism. The inoculated culture medium is then introduced into a large-sized container containing an aqueous solution of a sulfite as an oxygen absorbing agent and cultivated. The cultivation may be carried out by the following other methods: (1) The culture medium is introduced into a general sealed container for the cultivation, and a container, molded from the material, and containing the oxygen absorbing agent is introduced into the culture medium. (2) One of the materials capable of absorbing oxygen on mixing, e.g. ferrous sulfate or pyrogallol and an aqueous solution of sodium hydroxide, is sealed in an ampul, sealed with the other in the container, and then mixed therewith on breaking the ampul in use.
