Abstract

PURPOSE:To obtain an enzyme electrode responding quickly to a change in the concn. of substrate by adsorbing and immobilizing coenzyme on a conductive carrier, and immobilizing oxidoreductase to the resultant carrier by a cross-linking method. CONSTITUTION:A material which is conductive, has good adsorbing and holding power for coenzyme and is chemically stable, such as acetylene black, is molded under pressure as a binder, whereby an electrode substrate 1 is made. An aq. soln. of, for example, coenzyme nicotinamide adenine nucleotide (NAD) is developed on the substrate 1, after which it is dried, whereby an adsorbing and immobilizing layer 2 for coenzyme is provided. Next, an aq. soln. of oxidoreductase alcohol dehydrogenase is developed on the layer 2 and is dried, whereby an immobilizing layer 3 is formed. Thence these are caused to crosslink and react in the vapor of glutaraldehyde, whereby the enzyme electrode is obtained. This enzyme electrode is mounted in the bottom end part of a holder 7, and is immersed in buffer 8 contg. substrate (ethanol) 8, so that the analysis of good response speed is accomplished by using a device constituting a counter electrode 10, a salt bridge 9, a reference electrode 9, a potentiostat 5, and a recorder 4.
