Abstract

PURPOSE:To easily measure over all ranges of concentration of the sample without radiation damage even when the concentration range of the sample is extremely wide, by using a spectral sensitizing dye of photograph as a labeling agent. CONSTITUTION:An antigen or an antibody is labeled with a labeling agent of organic dye, and this labeled substance, the samping contg. the antigen or antibody to be measured and the antibody or antigen which reacts with the antigen or antibody specifically, are reacted competitively. In this case, the labeling agent of organic dye is a spectral sensitizing dye for photograph, i.e., which has an absorbing range in the longer side of wavelength than that of inherent range of the silver halide, and absorbed to the grain of silver halide, as well as being capable of spectral sensitization. The resulted reaction product of antigen-antibody is brought into contact with silver halide, and exposed to the light of wavelength which is absorbed by the spectral sensitizing dye. The exposed silver halide is developed, and the optical density of the resulted chromophoric dye is measured. According to this process, the sensitivity is controlled freely by adjusting the intensity of radiation, therefore the determination is carried out over a wide range.
