Abstract

PURPOSE: A 2-thioxopenam-3-carboxylic derivative is made to react with an alkylating reagent or hydroxy compound and an azo compound to produce the titled compound used as an antibacterial or synthetic intermediate thereof.
CONSTITUTION: The reaction between a compound of formula 2 (R7 is H, lower alkyl, lower alkoxy; R9 is carboxyl-protecting group) and another compound of formula 4 (R2 is H, lower alkyl; R8 is amino-protecting group; A is lower alkylene with a branched chain; X is halogen) affords a compound of formula 3. Or the reaction between the compound of formula 2, a compound of formula 5 and the third compound of R10OOCN=NCOOR10 (R10 is alkyl) is conducted in the presence of triphenylphosphine to give the compound of formula 3. When necessary, the protecting groups are removed or the substituted groups are exchanged to produce the compound of formula 1 (R1 is R7, lower hydroxyalkyl; R3 is H, R8; R4 is H, R9) and its salt.
