Abstract
PURPOSE: To prepare a methanol-containing agar medium which gives highly developed colony of methanol-assimilable bacteria and is suitable for the conservation of the bacteria, by sterilizing agar and the other medium component separately, and mixing both components.
CONSTITUTION: Agar is mixed with a part of water to be added to a medium, usually 5W40pts.wt. water per 1pt.wt. of agar, and sterilized with steam at e.g. 110W130°C (about 2 atm.) for 10W30min. Separately, the other component of the medium is mixed with the rest part of the water, and sterilized by filtration or by heating under the above conditions. Both liquid compositions are cooled pref. immediately after sterilization to about 50W60°C, mixed together, and poured dividedly into cultivation vessels to obtain methanol-containing agar medium. The addition of methanol may be before or after the sterilization similar to the conventional method of medium preparation. The concentration of methanol of the medium is adjusted below 3wt%. The components of the medium may be those used in the conventional culturing medium.
