Abstract

PURPOSE: To determine pyruvic acid accurately even in the presence of other salts, by subjecting a pyruvic acid solutio containing a hypotaurine to the action of an ω-amino acid-pyruvic acid transaminase to generate SO2 gas.
CONSTITUTION: A pyruvic acid solution containing a hypotaurine is subjected to the action of an ω-amino acid -pyruvic acid tansaminase which is an enzyme having very high specificity for the pruvic acid. The amount of SO2 gas in th resultant substances is measured to determine the pyruvic acid specifically. The purified and crystallized ω- amino acid -pyruvic acid transaminase has a molecular weight of 167,000±3,000, shows the maximum activity at pH8.5W10.5, and is specifically active only for the pyruvic acid. Thus, the determined value is not affected even by the presence of an amino acid, e.g. glutamic acid, methionine or alanine, α-ketoglutaric acid, α-ketobutyric acid, etc.
