Abstract

PURPOSE: To eliminate the positive error which occurs in the determination of a specific component of a biospecimen containing ascorbic acid, by adding heparin, Mn ions and ascorbic acid oxidase to the specimen thereby decomposing the ascorbic acid.
CONSTITUTION: A specific component (e.g. α-liporotein) in a biospecimen (e.g. serum) containing ascorbic aicd is determined as follows. The ascorbic acid in the specimen is decomposed by adding Mn ions and heparin as a polysaccharide sulfate to the specimen, and at the same time or prior to the addition of the above components, ascorbic acid oxidase is added thereto. The amount of the specific component can be determined without positive error caused by the presence of ascorbic acid, by determining the amount of the oxidative substance (e.g. H2O2) originated from the specific substance and produced correlatively with the amount of the substance.
