Abstract
PURPOSE: To obtain an L-form bacterium having a good antitumor activity and no side effects, e.g. phlogogenic and pyrogenic properties, by subjecting a bacterium body of a hemolytic streptococcus to the action of a hemolytic enzyme, and incubating the resulting protoplast in a culture medium containing a cell wall synthesis inhibitor.
CONSTITUTION: The bacterium body of a hemolytic streptococcus is subjected to the action of a hemolytic enzyme to form a plastoplast, which is then incubated in a culture medium containing a cell wall synthesis inhibitor to give an L-form bacterium of the hemolytic streptococcus. For example, a bacterium body of a hemolytic streptococcus is suspended in a hypertonic solution, and a hemolytic enzyme is added to the solution and treated at 10W40°C for 10W120min. The resulting protoplast is then incubated in a solid culture medium for the L-form bacterium containing an inhibitor for the cell wall synthesis, e.g. penicillin, at 25W40°C. In the incubation, the hemolytic streptococcus is in oculated in the solid culture medium for the L-form bacterium, and a paper disk impregnated with the inhibitor tfor the cell wall synthesis is placed on the culture medium to incubate the protoplast effectively.
