Abstract
PURPOSE: Heat-resistant acetate kinase is collected from a thermophilus Bacillus bacterium and made to react with a carrier of porous glass for immobilization, thus producing the titled substance showing high enzymatic activity after immobilization.
CONSTITUTION: Bacillus stearothermophilus or the like is cultured and acetate kinase is collected at any stage, e.g., from the culture mixture, culture filtrate or separated cells or obtained as the purified product. A previously activated carrier for immobilization such as porous glass, polysaccharide derivative or polyacrylic acid amide and the above heat-resistant acetate kinase are dispersed in a buffer solution of 6W9.5 pH and they are stirred at 0W70°C, when necessary irradiated with UV or visible rays to give the objective heat-resistant immobilized acetate kinase.
