PURPOSE: To prepare talampicillin, an antibacterial agent, in a short step, by reacting ampicillin with benzaldehyde, reacting the reaction product directly with a phthalide derivative without separating from the reaction mixture, and treating with an acid.
CONSTITUTION: Ampicillin (6-α-aminobenzylpenicillin) is made to react with benzaldehyde to obtain the Schiff base of formula I, which is reacted with a phthalide derivative of formula II (i.e. 1-oxo-3-hydroxyisobenzofuran). The objective talampicillin [chemical name; 6-[D(-)-α-aminophenylacetamide]penicillanic acid phthalide ester] of formula III can be prepared by the acid treatment of the reaction product.
