Abstract

PURPOSE: To obtain the titled compound which is an intermediate for tryptophan in high yiled under mild conditions, by reacting an N-(substituted methylene)glycine ester with a (3-indolylmethyl)trialkylammonium salt as raw materials.
CONSTITUTION: An N-(substituted methylene)glycine ester of formula I [R1 and R2 are H, alkyl, (un)substituted aryl or (non)cyclic thioketal residue; R3 is ester residue] is reacted with a (3-indolylmethyl)trialkylammonium salt of formula II (R4, R5 and R6 are alkyl; X- is halide ion, sulfate ion, alkylsulfate ion, hydrogensulfate ion, hydroxyl ion, etc.) to give an N-(substituted methylene)tryptophan ester of formula III. The raw materials of formulas I and II are readily obtained in high yield. The reaction is preferably carried out in a homogeneous solvent and between two phases of solvents. The compound of formula III is hydrolyzed to afford tryptophan readily.
