Abstract

PURPOSE: To prepare 1,2-dehydrotestosterone (17β-hydroxy-androsta-1,4-dien-3- one; DHT) in large quantity, by culturing microorganisms capable of producing androsta-1,4-diene-3,17-dione (ADD), under controlled aeration.
CONSTITUTION: In the preparation of DHT from a sterole having a side chain at 17-position, using microorganisms capable of ADD, the aeration rate is controlled to >0.1VVM in the first 2/5 of the ADD fermentation period, and to ≤0.1VVM after an arbitrary time between 2/5 and 4/5 of the fermentation period. The microorganism of this invention is a mutant capable of producing ADD from steroles, or a strain capable of producing ADD in the presence of an oxidation inhibitor of a chelating agent.
