Abstract

PURPOSE: To prepare peptidase, which is an enzyme of the endo type hydrolyzing the peptide bonds between proline and other amino acids specifically, from the bodies of peptidase-producing bacteria belonging to the genus Flavo bacterium.
CONSTITUTION: Peptidase-producing bacteria, e.g. Flavobacterium TY-78-74 (FERM-P No.4849), which belong to the genus Flavobacterium, are incubated, and the bacterium bodies are destroyed to obtain the cell fluid, which is salted out with ammonium sulfate to form a precipitate. The precipitate is subjected to chromatography with a cation exchanger to separate a peptidase-containing fraction, which is passed through a hydroxy apatite column, and purified by gel filtration.
