Abstract
PURPOSE: To prepare the titled substance, easily, in high efficiency, by breaking the peptide bonds of swine-type insulin with an enzyme acting specifically to the bond, and forming peptide bonds using the same enzyme.
CONSTITUTION: An enzyme acting specifically to the Pro-Lys bond in the B- chain of swine-type insulin, e.g. peptidase produced by the bacteria belonging to Flavobacterium genus or peptidase separated from the kidney of a lamb, is added to an aqueous medium. Swine-type insulin is hydrolyzed in the aqueous medium to obtain desdipeptide-B29,B30-insulin. The amino group of the desdipeptide-B29,B30-insulin is protected to obtain X2-desdipeptide-B29,B30-insulin, which is condensed with N -X-Lys-Thr in the presence of the above enzyme. The reaction is carried out by using the latter reactant in excess to the former one, and rapidly removing the reaction product from the reaction system.
