Abstract

PURPOSE: To obtain the title compound having improved antibacterial action commercially advantageously, by reacting cephamycin C having protected amino group with a tetrazole, and by reacting the compound having the protected carboxyl groups with an acyl chloride.
CONSTITUTION: The amino group of cephamycin C is protected by acetyl chloride, etc., and the cephamycin C is reacted with 1-methyl-5-merceptotetrazole in the presence of NH3 in an aqueous solution. The carboxyl groups of the reaction product are protected by isobutene, etc., and the protected compound is then reacted with cyanomethyl thioacetyl chloride in the presence of molecular sieve or a silylating agent, e.g. N-(trimethylsilyl) acetamide. The protective groups of the carboxyl groups are removed, if necessary, by the conventional method to give 7β-cyanomethyl thioacetamido-7β-methoxy-3-(1-methyl-1H-tetrazole-5-yl)thiomethyl-3- cephem-4-carboxylic acid.
