Abstract

PURPOSE:To measure the quantity of enzyme by arranging at least three optical absorption detectors in consecsutive positions with respect to the proceeding of an enzyme reaction so that the detected value for use in calculation may be selected in accordance with the values obtained by the first and second detectors. CONSTITUTION:In case glutamic acid oxalacetic transminase (GOT) in serum is to be measured by the production rate method using malic dehydrogenase enzyme (MDH) and nicotinic acid amide adenine dinucleotide of reduction type, the concentration of NADH2 is measured at the times t1, t2 and t3 after the start of the enzyme reaction to obtain the measured absorption values d1, d2 and d3. Then, the preset absorption variation level DELTAd0 is compared with the value obtained from equation II so that, if the latter is higher, the value of equation II is used as the value DELTAE of equation I and so that, if the latter is lower, the value of equation III is used as DELTAE of equation I thereby to calculate the value of GOT. Here, V indicates the liquid quantity of the final reaction; TF indicates a temperature coefficient; epsilon indicates the absorption coefficient at the measured wavelength of NADH2; v indicates the quantity of serum; and a indicates the length of the optical path.
