本発明は、特にシグマE(σE)遺伝子を発現し、かつ胞子形成を全くせず、または胞子形成をほとんどせず、または成育可能な胞子を産生しないことを特徴とするBa cillus thuringiensisの菌株に関する。本発明は、Baci llus thuirngiensisの上記菌株を含む有害生物駆除組成物にも関する。

【発明の詳細な説明】
Bacillus thuringiensisの新規菌株およびそれらを含む有害生物駆除組成物
本発明は、Bacillus thuringiensis(バチルス チューリンゲンシス)の新規
な菌株、それらを用いる有害生物駆除組成物、並びに目的のタンパク質を発現す
るためのこれらの菌株の使用に関する。
Bacillus thuringiensis(Bt)は、特に多数の昆虫の幼虫に対して殺虫特性
を有するタンパク質を産生するグラム陽性菌である。これらの細菌は、場合によ
っては不活性化の後に、穀物または疾患のベクターに有害な昆虫、特に蚊に作用
することを意図した有害生物駆除組成物に用いられる。
現在では、Bt血清型3a 3bが特に穀物の有害生物に対して用いられ、血
清型H14は蚊の幼虫を殺すのに用いられている。
Bacillus thuringiensisによって産生される有害生物駆除活性を有するタンパ
ク質はδ−内毒素と呼ばれ、胞子形成の際に多量に産生される。それらはパラ胞
子結晶性封入体の形態で蓄積し、胞子形成細胞の乾燥重量の25%まで存在する
ことができる。
δ−内毒素の多数の遺伝子はクローニングされ、配列決定され、配列相同性お
よび毒性スペクトルに基づいて5つの群に分類されている。相当する遺伝子は、
cry遺伝子と呼ばれる。
Bacillus thuringiensisに基づいた処方物は、様々な商品名で30年近く生物
学的有害生物駆除剤(biopesticides)として用いられてきた。生物学的抑制剤と
してのBacillus thuringiensisの使用は、化学的有害生物駆除剤に関して多数の
利点を有し、実際に、これは宿主スペクトルが狭くかつ極めて特異的であり、標
的でない昆虫に対しては作用せず、またこれは脊椎動物または環境に望ましくな
い作用を持たない。
しかしながら、環境でのδ−内毒素の持続性が僅かであり、処方物中に胞子が
存在することは、Bacillus thuringiensisを基剤とする生成物を発売する上での
2つの不都合な点である。これらの2つの問題を解決するために、特許出願EP
−192,319号明細書では、特にCry1Ac毒素を発現するPseudomonas fluorescens
型細胞を用いてあるいは特許出願PCT WO94/25612号
明細書には、spo0A遺伝子における冒された胞子形成しない突然変異体でC
ry1IIA毒素を発現することによる細胞膜に毒素をカプセル封入することが提
案されてきた。この後者の方法は、cryIII A遺伝子の発現の様式が他のcr
y遺伝子の発現の様式とは異なっているために可能であり、実際にこのcryII
I A遺伝子は、胞子形成の開始に関与しているすべての遺伝子または胞子形成に
関与している因子とは独立している。
Bacillus thuringiensisにおいて、胞子形成はそれぞれシグマ[σ]35およ
びシグマ[σ]28と呼ばれる2種類のシグマ因子の発現によって変化し、Baci llus subtilis
におけるシグマ(σ)Eおよびシグマ(σ)K因子との相同性が
大きいことを考慮して、以下において用いられるこの後者の名称は相当する遺伝
子と同様にsigEおよびsigKと呼ぶ。
本発明は、σEを発現するが、胞子形成はしないか、または胞子形成をほとん
どしないか、または成育可能な胞子を生成しないBacillus thuringiensisの菌株
に関する。
本発明は、sigEを発現するが、sigKは発現しないBacillus thuringie nsis
の突然変異体が、相当する野生の菌株と実質的に同一の毒素を多量に産生す
るが、一方胞子形成はしないかまたは成育可能な胞子を生成しないという事実の
実証に基づいている。
これは、詳細には菌株がsigK-株である場合である。
このような菌株の構築は、1)生物学的有害生物駆除剤で処理する際の胞子の環
境への散布が回避され、2)それらをカプセル封入するため環境での毒素の持続性
が増加するという2つの利点を有する。
試験は、シグマK遺伝子sigK-)を発現しないBt株は、供給源菌株と同
等な毒素を多量に蓄積することができるが、胞子は生成しない。これは、cry
遺伝子、または発現がσEタンパク質の産生によって変化する関連遺伝子によっ
てコードされる毒素をほぼ全量産生することができた。
BtのsigK-突然変異体を得るには、任意のDNA配列を導入することに
よる挿入、または欠失、またはsigK遺伝子の相の変化による中断法を用いる
のが特に有利であり、更に、このDNA配列を突然変異体に選択特性を付与する
ように選択することができ、例えば、これは中断を施した菌株が選択される抗生
物質、特にカナマイシンに対する耐性であることができる。
sigK-突然変異体も同様に、調節領域を有するまたは持たないsigK遺
伝子のヌクレオチド配列に相当するヌクレオチド配列の総てまたは一部を欠失す
ることによって得ることができる。
遺伝子の中断の技術は知られており、それらは本質的にはsigK遺伝子を運
ぶDNA配列の水準で任意のDNA配列を導入することにあり、菌株に導入され
る全体が相同組換えを生成し、sigK遺伝子は中断されたsigK遺伝子によ
って置換されている。選択特性により、この時点で目的の突然変異体を選択する
ことができる。
形質転換をしようとする菌株として極めて変化したまたは極めて顕著な毒素産
生を有するBacillus thuringiensisの菌株を選択するのが特に有利であるのはも
ちろんである。実際に、上記したように、sigK遺伝子の中断の事実は、胞子
形成を遮断するだけであり、毒素の産生は遮断しない。
Btは、Bacillus thuringiensisの任意の菌株を意味するものとして理解され
る。
従って、中断を施そうとする菌株では、工業用菌株を用いることができる。例
えば、Dulmage H.T.(1970)によって記載されたBt subsp .kurstaki HD-1、ま
たはBt israelensis、またはBt aizawai 7-29(この菌株は、第T07029号
でIEBCから入手可能である)のような野生菌株である。
本発明は、更に詳細には、それぞれ1995年10月26日に第I−1634
号でおよび1996年10月22日に第I−1776号でザ・ナショナル・コレ
クション・オブ・マイクロオーガニズム・カルチャーズ・オブ・ザ・インスティ
テュート・パスツール(the National Collection of Microorganism Cultures
of the Institut Pasteur )に寄託された菌株 Bacillus thuringiensis 407 Si
gK-(pHT410)並びに組換え体菌株Bacillus thuringiensis Kto SigK-(pHTF3-IC/
A)(b)-IRS-T-Δ)に関する。
毒素の産生を増加させるために、本発明によるsigK-菌株に自己複製プラ
スミド系を導入して、同様に専門家に知られている構築物によって上記毒素を確
実に発現させることも可能である。
この菌株によって発現されるタンパク質は、求められる有害生物駆除活性の種
類に依存し、Cry1は類に対して毒性を有し、Cry1Iは鱗翅類及び双翅類
に対して毒性を有し、Cry1Vは双翅類に対して毒性を有する。
概していえば、cryIC、cryIA、cryIVA、B、Dのような天然で
所定数の毒素を発現する野生菌株のsigK-を生成することが可能である。ま
た、本発明により記載された突然変異体菌株であって、sigK-でありかつ染
色体またはプラスミドに組み込んだ後Btゲノムに関して同種または異種タンパ
ク質をコードする遺伝子を発現する菌株を用いることも可能である。
利用可能な技術の実例は、Biotechnology,1992,vol.10,p.418(Lereclus
et al)に記載されている。
遺伝子、例えばcyrIC遺伝子を、同種組換えによってBt sigK-に
導入することができる。組換え体は、cryIC遺伝子を得るが、任意の異種D
NAを保持しない。
このようにして、数種類の毒素遺伝子を相同組換えによって細菌染色体に加え
ることができ、または常在性プラスミドに組み込むことができる。
例えば、Bt kurstaki の菌株であって、それ自身のプロモーターの制御下でc
ryIAc遺伝子およびcryIII A遺伝子のプロモーターの制御下でcryI
C遺伝子を同時に発現するものである。
これらの構造体で利用可能なcry遺伝子の中には、cryI、cryII、c
ryIVおよびcytを挙げることができる。
発現される遺伝子の導入のもう一つの方法は、pHT304およびpHT31
5プラスミドに関する特許出願PCT WO93/02199号明細書などに記
載のBtの機能的複製源を有するグラム陽性菌プラスミドを使用することからな
っている。
本発明の方法によって得られるsigK-菌株は、場合によっては不活性化し
た後に、有害生物駆除組成物、特に幼虫、詳細には昆虫の幼虫を殺す目的で用い
られる殺虫組成物に利用することができる。この有害生物駆除組成物は、自体知
られている手法によって調製され、すなわちこれが必要な場合には、関連のBaci
llus毒素の活性を最適にする不活性または非不活性担体との混合物として調製さ
れる。
ある国では、胞子形成菌株の利用に本質的な菌株の不活性化は、本発明に関し
て構築されるsigK-突然変異体の場合には任意である。この不活性化は、任
意の物理的または化学的方法によって、特に照射によって行うことができ、これ
によって菌株を成育不可能とする。本発明による菌株は成育可能な胞子を持たず
、それらの不活性化は、胞子形成した菌株の場合より容易である。
上記のように、毒素は細菌の内部に保持されているので、環境での毒素の有効
期間を増加することができる(毒素は幼虫が細菌を消化する際にだけ放出される
)。しかしながら、本発明によるある種の突然変異体は極めて例外的な耐性を有
し、この場合には、選択された特異的な菌株を用いて処理を行う昆虫で良好に消
化されるようにし、または例えば界面活性剤のような化学的処理、物理的処理、
超音波処理、または生物学的処理、特定の要素を(遺伝子組換え技術などにより
)微生物の壁に導入して、微生物が摂取されたときに毒素を一層良好に消化され
または一層容易に接近し得るようにする必要があることを示すことが可能となっ
た。
このBtの新規な菌株は、有害生物駆除組成物に要素の一つを供給することが
できるが、Btのゲノムに関して同種または異種遺伝子を発現するベクターとし
ても有用であり、上記遺伝子は、自己複製プラスミドによってまたは細菌のゲノ
ムに関する相同組換えによってBtのsigK-突然変異体でクローニングされ
る。
例えば、プロテアーゼ、リパーゼ、または任意の他の種類のタンパク質を発現
することができるベクター系の構築は、特許出願PCT WO94/25612
号明細書に記載されているものと同様であることができる。
本発明は、SigE遺伝子を含み、任意の活性なSigK遺伝子を含まず、目
的の遺伝子をコードする配列を含むヌクレオチド配列にも関する。
本発明の他の特徴および利点は、下記の実施例を読めば明らかになるであろう
。
第1図は、Bacillus thuringiensisのsigEおよびsigK染色体遺伝子の
中断を表す。pAB1およびpAB2プラスミドが相同組換えによってBt染色
体に組み込まれ、相同組換えの第二の場合には、総てのpRN5101配列が失
われる。矢印はそれぞれアンピシリン、エリスロマイシンおよびカナマイシンに
対する耐性を付与する遺伝子に相当するApR、EmR、およびKmR遺伝子の転
写方向を示し、三角形はそれぞれpBR322(oriEc)の複製源およびp
E194ts(orits)の複製源を表す。
第2図は、Bacillus thuringiensis における転写の分析のためのプラスミド
の構築を表す。pHT304−18Zは以前に報告されている(Agaisse and Ler
eclus 1994b)。矢印はermC、blaおよびlacZの転写方向、およびE.c
oli(oriEc)における機能的複製方向を示し、ori1030はBt p
HT1030プラスミドの複製源であり(Lereclus and Arantes 1992)、破線矢
印は上記のようにプロモーターpsigE、psigK、Bt IおよびBt
IIから開始した転写方向を示す(Rong et al.,1986; Sandman et al.,1988; Wo
ng et al.,1983)。spoIID、cotAおよびcryIAa遺伝子のプロモー
ター領域を有するHindIII −BamHI断片は、pHT304−18Zにお
いてクローニングされている。
第3図は、spoIIDおよびcotA遺伝子のプロモーターの制御下でのBt
におけるβ−ガラクトシダーゼの発現を表す。細胞は30℃でSP培地で成育さ
せ、時間0は対数増殖期の終了を示し、tnは時間0の前または後の時間数であ
る。AはpHTspoIIDを有するBt株のβ−ガラクトシダーゼ活性であり、
BはpHTcotAを有するBt株のβ−ガラクトシダーゼ活性である。β−ガ
ラクトシダーゼの特異活性はSpo+407(■)、407SigE(●)、お
よび407sigK-(○)で示される時間に測定した。
第4図は、30℃のSP培地で成育したpHTcryIA2を有するBtの菌
株におけるcryIAaの制御下で指示されたβ−ガラクトシダーゼの発現を表
す。β−ガラクトシダーゼ活性は、Spo+407(■)、407SigE(●
)、および407sigK-(○)で示される時間に測定し、
第5図は、pHTF3−1C/A(b)−IRS−Tプラスミドを表す。この
プラスミドはpBluescripIIKS-から誘導され(pBluescri
pIIKS-のDNAは「bla+oriEc」ボックスによって表され)、pB
luescripIIKS-、およびBacillus cereus 由来のテトラサイクリン耐
性を付与するtet遺伝子の直ぐ両側に位置したTn4430トランスポゾン(L
ereclus et al.,1986)の内部分割部位(internal resolution site)(IRS)
を含む2個の配列を有し、これは、更にcryIII Aのp3プロモーターの制御
下でcryIC/A(b)キメラ遺伝子のコード部分(Agaisse and Lereclus,1
994)、およびB .thuringiensisのpHT1030プラスミドの複製源(Lereclus
and Arantes,1992)を含んでいる。
第6図は、Kto SigK-菌株に含まれるTn4430トランスポゾンの
TnpIインテグラーゼによって触媒される2個のIRS部位の間の組換え反応
を表す。部位特異的組換えから生じるプラスミドはpHTF3−IC/A(b)
−IRS−T−Δと呼ばれる。実施例1 材料および方法 細菌株および培地
Bt407(血清型H1)およびそのアクリスタリフェラス誘導体(acrystall
iferous derivative)(Cry-)は、上記のようにO.Arantesによって単離され
た(Lereclus et al.,1989)。E.coli K-12株TG1(Δ(lac−proAB
)supE thi hud D5(F′traD36 pro+proB+la
clq lacZ ΔM15))を、クローニング実験に用いる( Gibson,1984
)。Bt株を30℃でルリア培地(LB)、およびHCT培地( Lecadet et al.
,1980)、または胞子形成栄養培地(SP培地)(Lereclus et al.,1995)で培養
する。E.coli 株は、LB培地で37℃で培養する。細菌選択のための抗生物質
濃度は下記の通りである:アンピシリン、100μg/ml(E.coli に対する
);エリスロマイシン、5μg/ml(Btに対する);カナマイシン、E.col
i に対して10μg/mlおよびBtに対して200μg/ml。プラスミドおよびDNA断片
S.Grussによって供給されたpRN5101プラスミドは、グラム陽性生物の
熱感受性複製源プラスミドであり、これはpBR322のClaI部位にpE1
94ts(Villafane et al.,1987)の挿入によって構築された。Bluescr
iptプラスミドはStratageneから入手し、pHT304−18ZおよびpHT
410プラスミド構築物は既に報告されている(Agaisse and Lereclus 1994b; L
ereclus et al.,1989)。Cry1Aa遺伝子のプロモーター領域を含む362
bp断片の増幅に用いられるオリゴヌクレオチド(Cry1A−1およびCry
IA−2)を、第1表に示す。Cry1A−1プライマーはHindIII 制限部
位を含む5′末端に7bpの伸長部を有し、Cry1A−2プライマーはBam
HI制限部位を有する8bpの伸長部を有する。この2つの制限部位を導入して
、pHT304−18Zにおけるクローニングを促進する。Bt sigEおよ
びsigK遺伝子を中断するため、相当する遺伝子の5′および3′領域を5′
末端に適当な制限部位を有するオリゴヌクレオチドを用いるPCRによって増幅
し、pBS KS-において別個にサブクローニングする。sigEおよびsi
gK
遺伝子の5′領域は、それぞれBamHI−XbaIの857および611bp
の制限断片である。3′領域は、それぞれ807および606bpのEcoRI
−BamHI制限断片である。これらの遺伝子のそれぞれの5′および3′領域
を含むDNA断片を精製して、pRN5101プラスミドのBamHI制限部位
におけるEnterococcus faecalis のaphA3遺伝子(KmRカセット)(Trieu-
Cuot and Courvalin,1983)を有する1.5kbのXbaI−EcoRI断片に
結合する。生成する熱感受性プラスミドpAB1およびpAB2は、それぞれs
igEおよびsigK遺伝子においてカナマイシン耐性遺伝子によって中断され
たコピーを有する。
B .subtilis のspoIIDおよびcotA遺伝子のプロモーター領域をそれぞ
れ有するプラスミドpDG675およびpDG676は、Dr.P.Stragier(Ins
titut de Biologie Physico-Chimique,パリ,フランス)から提供された。pH
TspoIIDは、pHT304−18ZのHindIII およびBamHI制限部
位の間のpDG675の300bpのHindIII −BamHI制限断片をサブ
クローニングすることによって構築した。pHTcotAは、下記のようにして
構築した。すなわち、pDG676のEcoRI−BamHI断片の400bp
を最初にpBS KS-においてサブクローニングして、pKScotAを得る
。次いで、pKScotAのHindIII −BamHI制限断片を、pHT30
4−18ZのHindIII とBamHI制限部位の間でサブクローニングし、生
成するプラスミドをpHTcotAという名称で呼ぶ。構築および形質転換
プラスミドDNAを、標準的なアルカリリーシス法によってE.coli から抽出
する。染色体DNAを、以前に報告されている方法でBtから抽出する(Msadek
et al.,1990)。制限酵素およびT4リガーゼは、New England Biolabs 、ビバ
リー、マサチューセッツから入手する。DNA断片を、Prep-A-Gene キット(Bio
Rad Laboratories,リッチモンド,カリフォルニア)を用いてアガロースゲル上
で精製する。オリゴヌクレオチドプライマーは、Genset(パリ、フランス)によ
って合成され、PCR増幅はGeneAmp PCR 2400装置(Perkin-Elmer,フォスター
シティー,カリフォルニア)を用いて行う。PCR増幅に用いたDNAマトリッ
クスは、Bt407から既にクローニングしたcryAa遺伝子(Lereclus et a
l.,1989)、または407 Cry-株から抽出した染色体DNAである。反応条
件は下記の通りである。95℃で5分間インキュベーションした後、ハイブリダ
イゼーションのため57℃で1分間を30サイクル、伸長のため72℃で1分間
、および変性のため92℃で1分間、最後に72℃で10分間新たなインキュベ
ーションを行う。Taqポリメラーゼは、USB Laboratories(クリーブランド、
オハイオ)から入手する。標準的手順を用いてE.coli の形質転換を行い、Bt
株は既に記載されているように電気穿孔によって形質転換した(Lereclus et al.
,1989)。
タンパク質分析は、Bt株の培養および超音波処理の後に行い、この分析は、
0.1%SDS−12%PAGE上で行う。殺虫活性のバイオアッセイ
製材の毒性を、第二段階でのPlutella xylostella の幼虫および以前に記載さ
れている自由摂取法(Sanchis et al.,1988)を用いて評価する。実施例2 BtのSigE−およびsigK−突然変異体の構築
それぞれsigEおよびsigKのKmRによって中断された遺伝子のコピー
を含むpAB1およびpAB2の熱感受性プラスミドを、電気穿孔によってBt
407 Cry-株に導入する。sigEおよびsigK遺伝子のsigE:
:KmおよびsigK::Kmで中断したコピーによる置換は、非許容温度(4
0℃)でカナマイシンの存在下での形質転換体の連続培養によって得られる(第
1
図を参照されたい)。第1図から分かるように、pAB1またはpAB2プラス
ミドによって形質転換されたBt株は、いずれも37℃でエリスロマイシンおよ
びカナマイシンに耐性である。
sigEまたはsigK遺伝子がその中断されたコピーに交換された形質転換
体を非許容温度、すなわちプラスミドの複製が遮断された温度で培養する。それ
らは、カナマイシンに対するそれらの耐性によって選択することができる。Sp
o 突然変異体(下記の407−SigE-および407−SigK-)は、カナ
マイシンに耐性であるが、エリスロマイシンには感受性である。Bt sigE
およびsigK遺伝子のそれらの中断されたコピーによる置換はPCR分析によ
ってチェックし、選択された突然変異体の染色体DNAをPCRのマトリックス
として用い、それぞれの相補的外部配列をそれぞれsigE−4およびsigK
−4オリゴヌクレオチドと組合わせてプライマーとして用いる。PCR生成物の
大きさは、KmRによって中断される遺伝子に相当する。
Bt SigE-およびSigK-突然変異株は、胞子形成を行うことができな
い。耐熱性胞子は、HCTまたはSP培地で30℃で72時間成育した後に生成
しない。同様な成育条件では、野生株の細胞の少なくとも90%が24時間また
は48時間後に胞子形成する。位相差顕微鏡による細胞の検討では、sigE-
突然変異株が初期の胞子形成段階で遮断され(段階II)、非対称隔膜を形成した
後、母細胞と胞子区画とに分裂する。sigK-突然変異株は後期胞子形成期(
段階IV)で遮断される。細胞の極の一方に位置する灰色の前胞子を、細胞の内部
に観察することができる。
lacZ遺伝子と融合したBacillus subtilis のspoIIDおよびcotA遺
伝子のプロモーター領域を有するpHTspoIIDおよびpHTcotAプラス
ミド(第2図を参照されたい)を構築して、Btの胞子形成の際のσEおよびσ
K因子の出現および消失を観察する。spoIIDを、σE因子を含むRNAポリ
メラーゼによって転写する(Lopez-Diaz et al.,1986; Rong et al.,1986)。こ
の遺伝子は、段階IIにおける胞子の形態学的発生に関与している(Young and Man
delstam,1979)。cotA遺伝子は、胞子嚢タンパク質をコードし、その転写は
σK(Sandman et al.,1988)によって変化する。pHTspoIIDおよびpHT
cotAプラスミドを電気穿孔によってBt 407 Cry-Spo+、407
−SigE-および407−SigK-に導入し、β−ガラクトシダーゼの合成を
SP培地での成育中に観察する(第3A図および3B図)。Spo+株では、s
poIIDプロモーターの制御下でのβ−ガラクトシダーゼの合成はt2で開始し
、t5ではタンパク質が最大約10,000U/mgに達した後、減少する。β
−ガラクトシダーゼの合成がcotAプロモーターの制御下にある菌株では、こ
れはt6だけで検出され、t11にはタンパク質が最大4000U/mgに達す
る。spoIIDまたはcotA′−′lacZ転写融合体に対する407−Si
gE-突然変異体では、β−ガラクトシダーゼの発現は検出されない(タンパク
質10U/mg未満)。sigKの転写はσEによって変化するので(Sandman e
t al.,1988)、この突然変異株では因子Kは産生されない。407−SigK-
突然変異株でcotAプロモーターから開始するlacZの発現は検出されず、
spoIIDプロモーターから開始する発現は野生株と同様にt6で最大となる。実施例3 BtのSigE-およびSigK-突然変異株におけるcry1Aa′−′lac Zの発現
Btの野生株およびSpo-突然変異株におけるcry1Aa遺伝子のプロモ
ーターの一過性調節(temporal regulation)を測定するため、cry1Aa′−
lacZ転写融合を含むプラスミドを構築した。cry1Aa遺伝子のプロモー
ター領域を含む領域を、記載されている方法でPCRによって増幅した後、la
cZリポーター遺伝子のpHT304−18Z上流でクローニングする。pHT
cry1A2という名称で呼ばれる生成したプラスミドを、電気穿孔によってB
t 407 Cry-Spo+、407−SigE-および407−SigK-株に
導入する。Spo+407 Cry-株におけるβ−ガラクトシダーゼの産生はt
2に開始し、t7に最初の、t11に第二の2個のピークを有する(第4図)。
spoIID′−′lacZおよびcotA′−′lacZ融合体について示され
ているように、t7およびt11はσEおよびσKの発現の最大期に相当する。
cry1Aaのプロモーター領域によって指示されたβ−ガラクトシダーゼの合
成の発現は、407−SigE-では顕著に減少する(第4図)。しかしながら
、若干のβ−ガラクトシダーゼ活性がt2で検出され、t10でタンパク質20
0U/mgの最大となる。cry1Aaのプロモーター領域によって指示された
β−ガラクトシダーゼ合成はt2で開始し、突然変異株407−SigK-では
t7で最大値9000U/mgのタンパク質を示す(第4図)。Spo+株で後
期胞子形成時の第二の発現ピークはSigK-突然変異株では現れず、これは、
後期胞子形成期中のcry1Aa遺伝子の転写におけるσK因子の関与を示して
いる。実施例4 BtのSigEおよびSigKにおけるCry1Aa毒素の産生
Bt 407の野生株のcry1Aa遺伝子を有するpHT410プラスミド
(Lereclus et al.,1989)を、電気穿孔によってBt 407 Cry-Spo+
、407−SigE-および407−SigK-株に導入する。
形質転換体を30℃でHCTおよびSp培地で培養し、結晶性封入体(crystal
line inclusion)の産生を位相差顕微鏡法および電子顕微鏡法によって検討する
。HCT培地で48時間成育したところ、大きな二ピラミッド型結晶が407−
Spo+および407−SigK-形質転換体で見られる。しかしながら、Spo+
株での結晶は、SigK-突然変異株の結晶が細胞壁にカプセル封入されたまま
であるのに、遊離する。HCT培地で72時間成育した後でも、SigK-突然
変異株からの結晶性封入体は遊離しない。pHT410プラスミドを有する40
7−SigE-では、結晶は観察されない。
HCT培地で成育した細胞からの結晶−細胞および胞子−結晶製剤に含まれる
タンパク質のSDS−PAGE分析では、pHT410を有する407−Sig
E-株は、pHT410を含む407 Cry-Spo+株から得られるのと同様
な毒素を産生する同じプラスミドを有する407−SigK-とは異なり、13
0kDaのCry1Aaポリペプチドを産生しないことを示している。
胞子−結晶および細胞−結晶製剤の殺虫活性を、第二段階でのPlutella xylos
tella 鱗翅類の幼虫を用いて分析する(第2表)。407−SigK-突然変異
株におけるCry1Aa以外のタンパク質の存在を考慮すれば、この菌株の結晶
性製剤での毒素の濃度を正確に測定することはできない。これは、LD50を培
養容積によって定義して、これらの生成物の殺虫活性を評価するからである。こ
のバイオアッセイは、407−SigK-で産生されるCry1Aa毒素は、P.
xylostella の幼虫にとって極めて毒性が高いことを示している。しかしながら
、これらの生成物の殺虫活性は、音波処理によって著しく増加する。
上記の実験条件下ではシグマKタンパク質を発現しないかまたは極めて弱くし
か発現せず、第I−1634号でCNCMに寄託されているBacillus thuringie nsis
株を、下記の条件下で構築する。
sigK遺伝子を含む菌株をカナマイシンに対する耐性を付与するaphA3
遺伝子によって中断する。このようにして構築した菌株を、Cry1Aa遺伝子
およびermC遺伝子を有するpHT410プラスミドによって形質転換して、
エリスロマイシンに対する耐性を付与する。この非胞子形成株は、静止期1aに
Cry1Aa毒素を多量に産生する。実施例5 cryIII A遺伝子のプロモーターの制御下でCry1C/Cry1A(b)キ メラδ−内毒素をコードする遺伝子を発現するKto SigK-(pHTF3 −IC/A(b)−IRS−T−Δ)と命名したB.thuringiensisの組換え菌株 の構築
Kto株はB .thuringiensisの天然の胞子形成株であり、この菌株はCry1
A(c)型のδ−内毒素を合成する。このδ−内毒素は、米国および欧州におけ
るトウモロコシ作物の主要な害虫であるOstrinia nubilalis(アワノメイガ)の
幼虫に対して殺虫活性を有する。一方、このδ−内毒素(従って、菌株Kto)
は、Spodoptera littoralis 、Spodoptera exigua またはMamestra brassicaeの
ようなヤガ科に属する他の重要な有害生物に対しては余り活性ではない(第3表
を参照されたい)。逆に、Cry1C δ−内毒素、または以下においてCry
IC/Cry1A(b)と呼ばれるCry1C/Cry1A(b)キメラδ−内
毒素であって、その構築(PHT81プラスミド)がSanchis et al.(1989)に
よって追加的に記載されているものは、S .littoralis に対して活性を有するが
、O .nubilalisに対しては余り活性でない(第3表)。
Kto株の活性のスペクトルを増加するには、Kto株にcry1C遺伝子ま
たはキメラcry1C/A(b)遺伝子を導入することが重要であった。しかし
ながら、既に1種類以上の他のδ−内毒素遺伝子を含み、その発現も同様にシグ
マEおよびシグマK胞子形成因子によって変化するB .thuringiensisの1菌株へ
のcry1型遺伝子(胞子形成に依存性)の導入は、δ−内毒素の総生成量の増
加によっては説明されないことが示されている。従って、cry1型の様々な遺
伝子を含む組換え菌株は、一層広い活性スペクトルを有するが、δ−内毒素のそ
れぞれは余り産生せず、従って、標的とする昆虫のそれぞれに特異的な単独のδ
−内毒素を産生する菌株よりも標的昆虫のそれぞれに関する有効性は低くなる。
この現象は、この菌株に存在する様々なcry1遺伝子のプロモーターによる胞
子形成のシグマ因子の滴定効果によって説明することができる。この問題を解決
するため、最近になり(Sanchis et al.,1996)、Cry1Cタンパク質をコード
する遺伝子を、発現がシグマ胞子形成因子(Agaisse and Lereclus,1994)とは独
立しているcryIII A遺伝子のプロモーターの制御下に置くことができること
が示された。
cryIII Aプロモーターの制御下でcry1C遺伝子をKto株に導入して
、組換えKto(pHTF3−1C−IRS−Δ)株を得た(Sanchis et al.,1
996)。この組換え菌株は、同時にCry1A(c)およびCry1C毒素を産生
し、産生されるδ−内毒素の量は、親菌株と比較して1.5〜2倍だけ増加する
。菌株Kto(pHTF3−1C−IRS−Δ)で得られる2種類のδ−内毒素
Cry1A(c)およびCry1Cの総生成量の増加は、恐らくはこの菌株にお
けるcry1C遺伝子の発現は胞子形成の特異的シグマ因子によって変化しない
ことによるものであり、胞子形成に依存するcry1A(c)遺伝子の発現を妨
害しない。本明細書に記載のKto SigK-(pHTF3−1C/A(b)
−IRS−T−Δ)株を構築するため、S .littoralis に対する活性がCry1
Cよりも若干勝っているキメラδ−内毒素Cry1C/A(b)をコードする遺
伝子を、cry1C遺伝子(Sanchis et al.,1996)について以前に記載した通り
にcryIII A遺伝子のプロモーターの制御下に置いた。
同様に、Kto株のsigK-突然変異株(このsigK遺伝子がaphA3
遺伝子によって中断されている)を、pAB2プラスミド(第1図を参照された
い)を用いて実施例2に記載の方法で構築した。BtのSpo-突然変異株であ
るKto SigK-をHCT培地で30℃で48時間培養すると、かなりの量
のCry1A(c) δ−内毒素を産生し、これは結晶性封入体の形態で蓄積し
、細胞中にカプセル封入されたままになり、リーシスを受けない。O .nubilalis
に
対するKto SigK-株の活性は、Kto SigK-株が予め音波処理され
ていてもまたはされていなくとも、Kto親株の活性と同等である。
次いで、Kto SigK-株を、pHTF3−1C/A(b)−IRS−T
プラスミドで形質転換した(第5図を参照されたい)。pBluescript
II KS-由来のこのプラスミドは、Tn4430_トランスポゾンの内部分割
部位(internal resolution site)(IRS)を含む2個の配列を有する(Lereclu
s et al.,1986)。これらの2個のIRSは、pBluescriptII KS-
、およびテトラサイクリンに対する耐性を付与しかつBacillus cereus 由来のt
et遺伝子の直ぐ両側に配置されている。更に、pHTF3−1C/A(b)−
IRS−Tは、cryIII Aのp3プロモーターの制御下にあるキメラcry1
C/A(b)のコード部およびB .thuringiensisのpHT1030プラスミドの
複製源を含んでいる(Lereclus and Arantes,1992)。
形質転換の後、Kto SigK-株に含まれているTn4430トランスポ
ゾンのTnpIインテグラーゼは、2個のIRS部位の間の組換え反応を触媒し
て、これらの2個の部位の間に含まれるDNAを切除する。組換えにより生じる
2個の環状分子のうち、pHT1030プラスミドの複製源とキメラcry1C
/A(b)遺伝子とを有するものだけが複製することができ、このようにして得
られたpHTF3−1C/A(b)−IRS−T−Δと呼ばれるプラスミドは、
pBluescriptII KS-およびtet遺伝子に相当するDNAを失っ
ている(第6図を参照されたい)。Kto SigK-(pHTF3−1C/A
(b)−IRS−T−Δ)組換え体菌株は、Cry1A(c)およびCry1C
/A(b) δ−内毒素をかなりの量で産生し、従って親KtoまたはKto
SigK-株よりも広い活性スペクトルを有するという利点を有する(第4表)
。
更に、このような菌株は2つの他の利点を有する。
1.)Cry1A(c)およびCry1C/A(b) δ−内毒素は、細胞中に
カプセル封入されたままである。これは、散布した後に分解および紫外線に対し
て付与される物理的保護により、処理を施した作物帯での毒素の持続性が増加す
ることによると考えられる。
2.)sigK-突然変異株は、胞子形成過程の段階IVで遮断されたSpo-突然
変異株であり、従って成育可能な胞子を産生せず、このような突然変異株の使用
により、殺虫処理中に胞子の環境中への伝播を回避することができる。

Claims (1)
- 【特許請求の範囲】 1. シグマE(σ)遺伝子を発現し、かつ胞子を形成せずまたは胞子をほと んど形成せず、または成育可能な胞子を産生しないことを特徴とする、Bacillus thuringiensis 。 2. 菌株が胞子形成しないことを特徴とする、請求の範囲第1項に記載のBa cillus thuringiensis 。 3. シグマK遺伝子を発現しない菌株であることを特徴とする、請求の範囲 第1項または2項に記載のBacillus thuringiensis。 4. SigK遺伝子が、DNA配列の導入によって中断されており、または 少なくとも部分的に欠失していることを特徴とする、請求の範囲第3項に記載のBacillus thuringiensis 。 5. SigK遺伝子がこの菌株に対する正の選択特性を付与するDNA配列 を導入することによって中断されていることを特徴とする、請求の範囲第4項に 記載のBacillus thuringiensis。 6. 正の選択特性が抗生物質耐性であることを特徴とする、請求の範囲第5 項に記載のBacillus thuringiensis。 7. Bt菌株が1以上のCry遺伝子を発現することを特徴とする、請求の 範囲第1〜6項のいずれか一項に記載のBacillus thuringiensis。 8. (1以上の)Cry遺伝子が、ベクター、例えばプラスミドによって運 ばれることを特徴とする、請求の範囲第7項に記載のBacillus thuringiensis。 9. Cry遺伝子が染色体に組み込まれることを特徴とする、請求の範囲第 7項に記載のBacillus thuringiensis。 10. 自己複製プラスミドによってまたは染色体に組み込まれたDNA配列 によって運ばれる目的タンパク質を発現することを特徴とする、請求の範囲第1 〜9項のいずれか一項に記載のBacillus thuringiensis。 11. 目的タンパク質を発現するSigK遺伝子中のDNA配列を含む事を 特徴とする、請求の範囲第10項に記載のBacillus thuringiensis。 12. SigK遺伝子の全部または一部の代わりに目的のタンパク質を発現 するDNA配列を含むことを特徴とする、請求の範囲第10項に記載のBacillus thuringiensis 。 13. 1995年10月26日にザ・ナショナル・コレクション・オブ・マ イクロオーガニズム・カルチャーズ・オブ・ザ・インスティテュート・パスツー ル(the National Collection of Microorganism Cultures of the Institut Pas teur)に番号I−1634で寄託されたBacillus thuringiensis 407 SigK-(pHT4 10)。 14. 1996年10月22日にザ・ナショナル・コレクション・オブ・マ イクロオーガニズム・カルチャーズ・オブ・ザ・インスティテュート・パスツー ル(the National Collection of Microorganism Cultures of the Institut Pas teur)に番号I−1776で寄託されたBacillus thuringiensis Kto SigK-(pHTF 3-1C/A(b)-IRS-T-Δ)。 15. 請求の範囲第1〜14項のいずれか一項に記載のBt菌株を含むこと を特徴とする、有害生物駆除組成物。 16. Bt菌が、菌株の消化性を向上させるために、あるいはタンパク質へ の接近性を向上させるために処理されたことを特徴とする、請求項第15〜19 項のいずれか一項に記載の組成物。 21. Bt菌株が音波処理によって処理したものであることを特徴とする、 請求の範囲第20項に記載の組成物。 22. SigE遺伝子を含み、任意の活性なSigK遺伝子を含まず、目的 遺伝子をコードする配列を含む、ヌクレオチド配列。
