- 【特許請求の範囲】 1.図1または図2のアミノ酸配列に本質的に対応するアミノ酸配列からなるヒ ト由来キチナーゼ、または該ヒト由来キチナーゼと実質的に同様のキチン加水分 解活性を有する該ヒト由来キチナーゼの改変体であることを特徴とする、実質的 に単離または精製されたキチナーゼ。 2.遺伝子工学的に処理された宿主細胞によって産生される、該宿主細胞または 該宿主細胞の培養に用いた培地から単離される、図1または図2のヌクレオチド 配列に本質的に対応するヌクレオチド配列によってコードされるアミノ酸配列か らなることを特徴とする、請求項1に記載のキチナーゼ。 3.本質的に図1のアミノ酸配列に対応するアミノ酸配列からなり、分子量が約 50kDaであることを特徴とする、請求項1に記載のキチナーゼ。 4.本質的に図2のアミノ酸配列に対応するアミノ酸配列からなり、分子量が約 39kDaであることを特徴とする、請求項1に記載のキチナーゼ。 5.請求項1〜4のいずれかに記載のキチナーゼ及び薬学的 に許容される担体または希釈剤を包含してなる医薬組成物。 6.キチン含有病原体による感染に対するヒトの治療または予防用の医薬組成物 であって、治療または予防に有効な量の請求項1〜4のいずれかに記載のキチナ ーゼ及び薬学的に許容される担体または希釈剤を包含してなる医薬組成物。 7.更に治療または予防に有効な量のヒト由来β−1,3−グルカナーゼを含有 することを特徴とする、請求項6に記載の医薬組成物。 8.請求項1〜4のいずれかに記載のキチナーゼ及び担体または希釈剤を包含し てなる医薬品以外の組成物。 9.細胞培養用の培地である、請求項8に記載の組成物。 10.ヒト由来細胞の培養用の培地である、請求項8に記載の組成物。 11.化粧用品(例:ボディローション)、歯科用品(例:練り歯磨き、含嗽剤 )または食品(例:牛乳、チーズ等の乳製品)である、請求項8に記載の組成物 。 12.請求項1〜4のいずれかに記載のキチナーゼを、キチンを加水分解するの に十分な量含有することを特徴とする、キチンを母材とする製造物。 13.薬物放出の制御を目的とする、薬物を含有する担体あるいは植込剤である 、請求項12に記載のキチンを母材とする製造物。 14.過渡的な機能性植込剤である、請求項12に記載のキチンを母体とする製 造物。 15.請求項5〜7のいずれかに記載の医薬組成物をヒトに投与することを含む 、キチン含有病原体による感染に対するヒトの治療方法または予防方法。 16.図1または図2のアミノ酸配列に本質的に対応するアミノ酸配列からなる ヒト由来キチナーゼ、または該ヒト由来キチナーゼと実質的に同様のキチン加水 分解活性を有する該ヒト由来キチナーゼの改変体の製造方法であって、 該ヒト由来キチナーゼまたは該改変体の産生能を有する、遺伝子工学的に処理 された宿主あるいは宿主細胞を増殖し、 該宿主、該宿主細胞またはその培養に用いた培地より、産生されたキチナーゼ を単離することを含む製造方法。 17.該キチナーゼのアミノ酸配列が、図1または図2のヌクレオチド配列に本 質的に対応するヌクレオチド配列によってコードされていることを特徴とする、 請求項16に記載の製造方法。 18.図1または図2のアミノ酸配列に本質的に対応するアミノ酸配列からなる ヒト由来キチナーゼ、または該ヒト由来キチナーゼと実質的に同様のキチン加水 分解活性を有する該ヒト由来キチナーゼの改変体を産生しうる、遺伝子工学的に 処理された宿主細胞。 19.図1または図2のアミノ酸配列に本質的に対応するアミノ酸配列をコード するヌクレオチド配列、またはそれに相補的なヌクレオチド配列を包含してなる 組換え核酸。 20.図1または図2のヌクレオチド配列に本質的に対応するヌクレオチド配列 、またはそれに相補的なヌクレオチド配列を包含してなることを特徴とする、請 求項19に記載の組換え核酸。 21.図1または図2のヌクレオチド配列に対応するか、もしくは相補的なヌク レオチド配列である、少なくとも約8塩 基からなるオリゴヌクレオチドであって、完全に近い相補性を要するハイブリダ イゼーション条件下において、請求項1〜4のいずれかに記載のヒト由来キチナ ーゼをコードする核酸とハイブリダイゼーションにより結合し得ることを特徴と するオリゴヌクレオチド。 22.図1または図2のアミノ酸配列より得られるアミノ酸配列を有する少なく とも8アミノ酸残基からなるペプチドであって、 請求項1〜4のいずれかに記載のヒト由来キチナーゼのエピトープ、あるいは 模擬エピトープであることを特徴とするペプチド。 23.図1または図2のアミノ酸配列に対応するアミノ酸配列からなり、抗原性 を有することを特徴とする、請求項22に記載のペプチド。 24.請求項1〜4のいずれかに記載のヒト由来キチナーゼに結合し得ることを 特徴とする抗体。 25.モノクローナル抗体であることを特徴とする、請求項24に記載の抗体。 26.請求項24または25に記載の抗体、並びに抗原または抗体検出用の診断 用キットに含まれる従来の成分を含んでなる診断用キット。 27.請求項22または23に記載のペプチド、並びに抗原または抗体検出用の 診断用キットに含まれる従来の成分を含んでなる診断用キット。 28.請求項21に記載のオリゴヌクレオチド、並びに核酸検出用の診断用キッ トに含まれる従来の成分を含んでなる診断用キット。 29.請求項19または20に記載の組換え核酸、ならびに核酸検出用の診断用 キットに含まれる従来の成分を含んでなる診断用キット。 30.請求項1〜4のいずれかに記載のヒト由来キチナーゼ、ならびに抗原また は抗体検出用の診断用キットに含まれる従来の成分を含んでなる診断用キット。 31.キチンの加水分解条件下において、請求項1〜4のいずれかに記載のヒト 由来キチナーゼにキチンを接触させることを包含する、キチンの分解方法。
