Description
Claims (1)
- 【特許請求の範囲】 1. 複数のヘテロ環式塩基が結合された天然には存在しない主鎖構造を含む 核酸模倣体であって、 該塩基の少なくとも一つは、主鎖への該塩基の結合位置から1、2また は3原子離れた位置が少なくとも一つの立体的にかさばった置換基で置換されて いることを特徴とする核酸模倣体。 2. 該立体的にかさばった置換基が−R’、−OR’、−SR’、−N(R ’)2、−C(R’)3、−C(=X)(R’)、−C(=X) (−Y−R’) またはS(=O)1-2(−Y−R’)であり、式中: XはO、SまたはNHであり; YはO、SまたはNHであり;および 式中R’は少なくとも3原子を含んでおり、およびH、C1−C50−アルキル、 C2−C50−アルケニル、C2−C50−アルキニル、C7−C50−アルキル−アリ ール、C6−C50−アリール、C10−C50−ナフチル、C12−C50−ビフェニル 、C7−C50−アリール−アルキル、ピリジル、イミダゾリル、ピリミジニル、 ピリダジニル、キノリル、アクリジニル、ピロリル、フラニル、チエニル、イソ オキサゾリル、オキサゾリル、チアゾリルまたはビオチニルであり、式中R’は 一つまたはそれ以上の−NO、−NO2、−SO3 -、−CN、−OH、−NH2、 −SH、−PO3 2-、−COOH、−F、−Cl、−Brおよび−Iで置換可能 である請求項第1項に記載の核酸模倣体。 3. 該塩基が天然に存在するまたは天然には存在しないピリミジン塩基であ る請求項第1項に記載の核酸模倣体。 4. 該立体的にかさばった置換基が該天然に存在するピリミジン塩基のC− 6、C−5またはN−4に結合されている請求項第3項に記載の核酸模倣体。 5. 該立体的にかさばった置換基が該天然に存在するピリミジン塩基のN− 4に結合されている請求項第4項に記載の核酸模倣体。 6. 該天然に存在するピリミジン塩基がシトシンである請求項第5項に記載 の核酸模倣体。 7. 該立体的にかさばった置換基が(C=O)−R’’(式中、R’’はC1 −C20−アルキルまたはC6−C18−アリールである)である請求項第5項に記 載の核酸模倣体。 8. 該立体的にかさばった置換基が(C=O)−C6H5である請求項第7項 に記載の核酸模倣体。 9. 核酸決定のための方法であって、 核酸模倣体を提供し; 該核酸模倣体および該核酸間の二重鎖の形成に適した条件下、該核酸模 倣体および該核酸をインキュベートし; 該核酸の存在の尺度として該二重鎖の存在を決定する; の各工程を含み、 該核酸模倣体は複数のヘテロ環式塩基が結合された天然には存在しない 主鎖構造を含んでおり、 該塩基の少なくとも一つは、主鎖への該塩基の結合位置から1、2また は3原子離れた位置が少なくとも一つの立体的にかさばった置換基で置換されて いる ことを特徴とする方法。 10. 一般式: 式中: R1は少なくとも一つの−C〇〇P1、−NHP1、−OP1または−SP1基を持 つC1−C4アルキルであり;P1は水素または保護基である; R2は−COOP2、−NHP2、−OP2または−SP2で置換されたC1−C4ア ルキルであり、式中、P2は水素または保護基である; Mは1から3の炭素リンカーによりNに結合されている、天然に存在するまたは 天然には存在しないヘテロ環式基であり;および R3は3またはそれ以上の非水素原子を含む立体的にかさばった置換基である; を有する、核酸模倣体製造のための化合物。
