Claims (1)
- 【特許請求の範囲】 1. リガーゼ活性をもつ核酸分子を得るための方法において、 a) 各々がランダム配列領域をもつ、候補核酸分子の集団を提供する段階; b) 該集団を、 (i) 基質核酸分子;及び (ii) 該基質核酸分子の3'領域の一部分及び該集団内の候補核酸分子の各 々の5'領域の一部分に対し相補的であり、かつ該集団からの候補核酸分子及び 該基質核酸分子に対するその結合が該3'及び5'領域を並置し、該3'又は該5' 領域のいずれかの末端ヌクレオチドが活性化された基を含んでいる外部鋳型; と接触させる段階; c) 該集団からリガーゼ活性をもつ核酸分子の下位集団を分離する段階; d) インビトロで該下位集団を増幅させる段階; e) 該増幅された下位集団について任意に段階b〜dを反復する段階;及び f) 該増幅された下位集団からリガーゼ活性をもつ該核酸分子を分離する段 階; からなる方法。 2. 段階b〜dの任意の反復が外部鋳型の非存在下で実施される請求項1に記 載の方法。 3. リガーゼ活性をもつ核酸分子又は基質核酸分子がDNAである請求項1に 記載の方法。 4. 基質核酸の5'末端ヌクレオチドがビオチン部分を含有する請求項1に記 載の方法。 5. 活性化された基が基質の3'末端ヌクレオチド上の3'−ホスホルイミダゾ リドである請求項1に記載の方法。 6. リガーゼ活性をもつDNA分子を得るための方法において、 a) 各々がランダム配列領域をもつ、候補DNA分子の集団を提供する段階 ; b) 基質核酸分子と該集団を接触させる段階; c) 該集団からリガーゼ活性をもつDNA分子の下位集団を分離する段階; d) 該下位集団をインビトロで増幅する段階; e) 該増幅された下位集団のために任意に段階b〜dを反復する段階;及び f)該増幅された下位集団からリガーゼ活性をもつ該DNA分子を分離する段 階; からなる方法。 7. 基質核酸分子がDNAである請求項6に記載の方法。 8. 基質核酸の5'末端ヌクレオチドがビチオン部分を含有する請求項6に記 載の方法。 9. 活性化された基が、基質の3'末端ヌクレオチド上の3'−ホスホルイミダ ゾリドである請求項6に記載の方法。 10. 触媒として作用できるDNA分子。 11. 核酸基質内のリボヌクレオチドの存在を必要としない触媒として核酸基 質に対し作用することのできるDNA分子。 12. リガーゼ活性を有する核酸分子。 13. 核酸分子がDNAである請求項12に記載の核酸分子。 14. リガーゼ活性がDNAリガーゼ活性である請求項12に記載の核酸分子 。 15. 第2の基質核酸に対して第1の基質核酸を連結する能力をもつ核酸分子 において、該連結の速度が、同じ反応条件下で鋳型処理することにより該基質核 酸を連結する速度よりも速い核酸分子。 16. 第1の基質核酸を第2の基質核酸に連結する能力をもつ触媒DNA分子 において、該第1の基質核酸が配列3'−S1−S2−5'を含み、該第2の基質核 酸が配列3'−S3−S4−5'を含み、それ自体配列5'−E'−TTT−E2−A GA−E3−E4−E5−E6−3'を含む触媒DNA分子であって、ここで、 S1は、S2の3'末端に隣接して位置づけされた2個以上のヌクレオチドを含 み、このS1ヌクレオチドは、該TTTの5'末端に隣接して位置づけされたE1 内の同等の数のヌクレオチドに対し相補的であり; S2は、1〜3個のヌクレオチドを含み、S3は1〜6個のヌクレオチドを含み 、S2の5'末端ヌクレオチド及びS3の3'末端ヌクレオチドが交互に活性化され た 基又はヒドロキシル基を含んでおり; S4は、S3の5'末端に隣接して位置づけされた2個以上のヌクレオチドを含 み、このS4ヌクレオチドはE5の3'末端に隣接して位置づけされたE6内の同等 の数のヌクレオチドに対して相補的であり; E1は、該TTTの5'末端に隣接して位置づけされた2個以上のヌクレオチド を含み、このE1ヌクレオチドはS2の3'末端に隣接して位置づけされたS1内の 同等の数のヌクレオチドに対して相補的であり; E2は、0〜12個のヌクレオチドを含み; E3は、該AGAの3'末端に隣接して位置づけされた2個以上のヌクレオチド を含み、該E3ヌクレオチドは、E6の5'末端に隣接して位置づけされたE5内の 同等の数のヌクレオチドに対し相補的であり; E4は、3〜200個のヌクレオチドを含み; E5は、E6の5'末端に隣接して位置づけされた2個以上のヌクレオチドを含 み、該E5ヌクレオチドは、該AGAの3'末端に隣接して位置づけされたE3内 の同等のヌクレオチド数に対して相補的であり、 E6は、E5の3'末端に隣接して位置づけされた2個以上のヌクレオチドを含 み、該E6ヌクレオチドは、S3の5'末端に隣接して位置づけされたS4内の同等 の数のヌクレオチドに対して相補的である、触媒DNA分子。 17. E2が3〜4個のヌクレオチドを含む、請求項16に記載の触媒DNA 分子。 18. S2の最5'ヌクレオチドがE2の最5'ヌクレオチドと相補的であり;S3 の最3'ヌクレオチドがE2の最も5'から2番目のヌクレオチドに対して相補的 であり、S3の最も3'から2番目のヌクレオチドがE2の最も5'から3番目のヌ クレオチドに相補的である、請求項17に記載の触媒DNA分子。 19. E2が4つのヌクレオチドを含み、S3の最も3'から3番目のヌクレオ チドがE2の最も5'から4番目のヌクレオチドに対し相補的である、請求項18 に記載の触媒DNA分子。 20. a) S2が1つのヌクレオチドを含み; b) S3が3つのヌクレオチドを含み; c) E4が5つのヌクレオチドを含み; d) E5及びE3が各々5つのヌクレオチドを含む、請求項16に記載の触媒 DNA分子。 21. 第2の核酸分子に対して第1の核酸分子を連結する方法において、該第 1及び第2の核酸分子をリガーゼ活性をもつ核酸分子と接触させる段階を含む方 法。 22. リガーゼ活性をもつ該核酸分子がDNAである、請求項21に記載の方 法。 23. リガーゼ活性がDNAリガーゼ活性である請求項21に記載の方法。 24. a) 各々がランダム配列領域をもつ、候補核酸分子の集団を提供する 段階; b) 該集団を、 (i) 基質核酸分子:及び (ii) 該基質核酸分子の3'領域の一部分及び該集団からの候補核酸分子の 各々の5'領域の一部分に対し相補的であり、かつ該集団内の候補核酸分子及び 該基質核酸分子に対するその結合が該3'及び5'領域を並置し、該3'又は該5' 領域のいずれかの末端ヌクレオチドが活性化された基を含んでいる外部鋳型; と接触させる段階; c) 該集団からリガーゼ活性をもつ核酸分子の下位集団を分離する段階; d) インビトロで該下位集団を増幅させる段階; e) 該増幅された下位集団について任意に段階b〜dを反復する段階;及び f) 該増幅された下位集団からリガーゼ活性をもつ該核酸分子を分離する段 階;により得られるリガーゼ活性をもつ核酸分子。 25. 段階b−dの任意の反復が外部鋳型の非存在下で実施される請求項24 に記載の核酸。 26. リガーゼ活性をもつ核酸分子がDNAである請求項24に記載のリガー ゼ活性をもつ核酸分子。 27. a) 基質核酸の5'末端ヌクレオチドがビオチン部分を含むか;又は b) 活性化された基が該基質の3'末端ヌクレオチド上の3'−ホスホルイミ ダゾリドである; 請求項24に記載のリガーゼ活性をもつ核酸分子。 28. a) 各々がランダム配列領域をもつ、候補DNA分子の集団を提供す る段階; b) 基質核酸分子と該集団を接触させる段階; c) 該集団からリガーゼ活性をもつDNA分子の下位集団を分離する段階; d) 該下位集団をインビトロで増幅する段階; e) 該増幅された下位集団のために任意に段階b〜dを反復する段階;及び f) 該増幅された下位集団からリガーゼ活性をもつ該DNA分子を分離する 段階; によって得られるリガーゼ活性をもつDNA分子。 29. a) 基質核酸の5'末端ヌクレオチドがビオチン部分を含むか、又は b) 活性化された基が該基質の3'末端ヌクレオチド上の3'−ホスホルイミ ダゾリドである請求項28に記載のリガーゼ活性をもつDNA分子。
