Claims (1)
- 【特許請求の範囲】 1.ビソクラミス・ニヴェア(Byssochlamys nivea)、ネオサルトリア・フィシェ リ(Neosartorya fischeri)及びジゴサッカロマイセス・バイリ(Zygosaccharomyc es bailii)の検出を、プライマーとしてリボゾーム単位の内部転写スペーサー( ITS)中に含有される配列を用いて標的微生物類のゲノムDNAの増幅によっ て可能とする方法; ビソクラミス・ニヴェア(Byssochlamys nivea)に対して、 −配列番号1に対応するITS1による −配列番号2に対応するITS2による ネオサルトリア・フィシェリ(Neosartorya fischeri)に対して、 −配列番号3に対応するITS1による −配列番号4に対応するITS2による ジゴサッカロマイセス・バイリ(Zygosaccharomyces bailii)に対して、 −配列番号5に対応するITS1による −配列番号6に対応するITS2による。 2.ITS1及びITS2配列中において、さらに詳細には、 −ビソクラミス・ニヴェア(Byssochlamys nivea)に対して、配列番号7及び8、 −ネオサルトリア・フィシェリ(Neosartorya fischeri)に対して、配列番号9及 び10、 −ジゴサッカロマイセス・バイリ(Zygosaccharomyces bailii)に対して、配列番 号11及び12 が選択される請求項1に記載の方法。 3.前記増幅がPCR法によって実施される請求項1又は2のうちの1項に記載 の方法。 4.食品、特に果物をベースにした食品、特に苺をベースにした食品中におけ るこれらの微生物類の検出方法で、その胞子または細胞を遊離させかつ濃縮する ために前記果物を含有する試料を前処理し、Taqポリメラーゼの阻害剤の作用を 低減または抑制し、さらに前記胞子または細胞からDNAを抽出する請求項1〜3 のうちの1項に記載の方法。 5.果物を含有する試料を、それらを液状化するためにセルラーゼ/ヘミセルラ ーゼの混合物で処理し、次に、DNAを抽出できる細胞を含有する産物を得るこ とが可能となる条件下で、濾過しかつ遠心分離する請求項4に記載の方法。 6.配列番号1から12までの配列から選択された配列を有するプライマー。 7.DNA増幅による検出と請求の範囲第3項乃至第5項のうちの1項に記載の 方法を実施するためのキットで、請求の範囲第6項に記載のプライマー類からな ることを特徴とするキット。
