Claims (1)
- 【特許請求の範囲】 1.実質的に純粋なRXR-相互作用性タンパク質。 2.(a)図4に示すRIP14-1のアミノ酸配列(配列番号:1)、 (b)図4に示すRIP14-2のアミノ酸配列(配列番号:2)、 (c)図5に示すRIP15のアミノ酸配列(配列番号:3)、 (d)図10に示すRIP110のアミノ酸配列(配列番号:4)、または (e)図11に示すRIP13のアミノ酸配列(配列番号:5) と実質的に同一なアミノ酸配列を含む、請求項1記載のタンパク質。 3.ポリペプチドが哺乳類由来のものである、請求項1記載のタンパク質。 4.哺乳類がヒトである、請求項3記載のタンパク質。 5.ポリペプチドが、RXRの存在下においてβ-RAREと結合する、またはRXRの存 在下においてEcRE部位と結合する、請求項1記載のタンパク質。 6.請求項1記載のタンパク質をコードする配列を含む精製されたDNA。 7.DNAがヒトRXR-相互作用性タンパク質をコードする、請求項6記載の精製され たDNA。 8.(a)図4に示すRIP14-1のDNA配列(配列番号:6)、 (b)図4に示すRIP14-2のDNA配列(配列番号:14)、 (c)図5に示すRIP15のDNA配列(配列番号:7)、 (d)図10に示すRIP110のDNA配列(配列番号:8)、または (e)図11に示すRIP13のDNA配列(配列番号:9) と実質的に同一なDNA配列を含む、請求項6記載のDNA。 9.請求項6記載の精製されたDNAを含むベクター。 10.請求項6記載の精製されたDNAを含む細胞。 11.細胞内において発現する位置にあるRXR-相互作用性タンパク質をコードする DNAによって形質転換された細胞を提供すること、DNA発現条件下において該形質 転換細胞を培養すること、および該組換えRXR-相互作用性タンパク質を単離する ことを含む組換えRXR-相互作用性タンパク質の作製方法。 12.請求項6記載の精製されたDNAの発現によって産生されるRXR-相互作用性タン パク質。 13.(a)(i)タンパク質結合部位に機能的に結合させたレポーター遺伝子と、 (ii)該タンパク質結合部位に特異的に結合することができる結合部分に 共有結合したレチノイドX受容体タンパク質を含む第一の融合タンパク質を発現 する第一の融合遺伝子と、 (iii)遺伝子活性化部分に共有結合した該被験タンパク質を含む第二の融 合タンパク質を発現する第二の融合遺伝子とを含む宿主細胞を提供すること、 (b)該被験タンパク質が該レチノイドX受容体タンパク質と相互作用しうるか否 かを示す指標として、該被験タンパク質が該レポーター遺伝子の発現を増強する か否かを判定することを含む、被験タンパク質にレチノイドX受容体(RXR)タン パク質と相互作用する能力があるか否かを判定する方法。 14.レチノイドX受容体と結合するリガンドで宿主細胞を処理すること、および 該細胞の該リガンドによる処理後にレポーター遺伝子の発現を増強する能力によ ってリガンド依存的相互作用性タンパク質を同定することをさらに含む、請求項 13記載の方法。 15.レチノイドX受容体と結合するリガンドで宿主細胞を処理すること、および 該リガンドの処理下および非処理下の両方においてレポーター遺伝子の発現を増 強する能力によってリガンド非依存的相互作用性タンパク質を同定することをさ らに含む、請求項13記載の方法。 16.レチノイドX受容体と結合するリガンドで宿主細胞を処理すること、および 該リガンドの処理下ではなく、非処理下において、レポーター遺伝子の発現を増 強する能力によってリガンド感受性相互作用性タンパク質を同定することをさら に含む、請求項13記載の方法。 17.遺伝子活性化部分がB42の遺伝子活性化部分である、請求項13記載の方法。 18.リガンドが9-cis-RAである、請求項14記載の方法。
