- 【特許請求の範囲】 1.配列表に配列番号11として示されるアミノ酸配列をコードする、単離され た核酸。 2.前記核酸が発現するに適切なベクター中の請求項1に記載の単離された核酸 。 3.前記核酸が発現するに適切な宿主中の請求項2に記載のベクター。 4.長さが少なくとも15ヌクレオチドの単離された核酸であって、ポリメラー ゼ連鎖反応の下で請求項1に記載の核酸と選択的にハイブリダイズし得る、核酸 。 5.長さが少なくとも15ヌクレオチドの単離された核酸であって、60℃およ び5×SSCのストリンジェンシー条件下、これに続く、室温、2×SSCおよび0.1 %SDSである最初の洗浄条件下、および50℃、0.5×SSCおよび0.1%SDSのスト リンジェンシー条件での2回の第2の洗浄下で請求項1に記載の核酸と特異的に ハイブリダイズし得る、核酸。 6.請求項5に記載の単離された核酸によってコードされる、精製された抗原性 ポリペプチド。 7.配列表に配列番号11として示されるアミノ酸配列を有する、精製された抗 原性ポリペプチド。 8.配列表に配列番号7として規定されるヌクレオチド配列中のヌクレオチドか らなる、単離された核酸。 9.前記核酸が発現するに適切なベクター中の請求項8に記載の核酸。 10.前記核酸が発現するに適切な宿主中の請求項9に記載のベクター。 11.高ストリンジェンシー条件下で請求項8に記載の核酸と特異的にハイブリ ダイズする単離された核酸であって、そして該核酸がハイブリダイズする鎖のセ グメントと少なくとも85%の配列相補性を有する、核酸。 12.請求項8に記載される核酸によってコードされる、精製された抗原性タン パク質。 13.請求項8に記載される核酸によってコードされる、抗原性ポリペプチドフ ラグメント。 14.配列表に配列番号5として規定されるヌクレオチド配列中のヌクレオチド を含有する、Rochalimaea henselaeに特異的な単離された核酸。 15.配列表に配列番号6として規定されるヌクレオチド配列中のヌクレオチド を含有する、Rochalimaea quintanaに特異的な単離された核酸。 16.配列表に配列番号1、配列番号2、配列番号3および配列番号4として示 される核酸の混合物。 17.被検体における現在および以前のネコ引っ掻き病(cat scratch disease) を診断する方法であって、該被検体由来のサンプルにおけるRochalimaea hensel aeの存在を検出する工程を包含し、ここで、該検出工程が、 該被検体由来の液体サンプルまたは組織サンプルと、配列表に配列番号11と して示されるポリペプチドと特異的に結合する、検出可能量の精製された抗体と を接触させる工程、 該抗体とポリペプチドとの結合を検出する工程、および 特異的結合の存在と、該被検体におけるネコ引っ掻き病とを相関させる工程、 を包含する、方法。 18.被検体における現在および以前のネコ引っ掻き病を診断する方法であって 、該被検体由来のサンプルにおけるRochalimaea henselaeの存在を検出する工程 を包含し、ここで、該検出工程が、 該被検体由来の液体サンプルまたは組織サンプルと、配列表に配列番号7とし て示されるポリペプチドと特異的に結合する、検出可能量の精製された抗体とを 接触させる工程、 該抗体とポリペプチドとの結合を検出する工程、および 特異的結合の存在と、該被検体におけるネコ引っ掻き病とを相関させる工程、 を包含する、方法。 19.R.henselaeと特異的に結合する抗体の存在を検出することによって、被 検体における現在および以前のネコ引っ掻き病を診断する方法であって、ここで 、該検出工程が、 該被検体由来の、抗体を含有する液体サンプルまたは組織サンプルと、配列表 に配列番号7として示される、所定量の精製されたポリペプチドとを接触させる 工程、 該ポリペプチドと該抗体との結合を検出する工程、および 該ポリペプチドと特異的に結合する抗体の存在と、該被検体におけるネコ引っ 掻き病とを相関させる工程、 を包含する、方法。 20.R.henselaeと特異的に結合する抗体の存在を検出することによって、被 検体において現在および以前のネコ引っ掻き病を診断する方法であって、ここで 、該検出工程が、 該被検体由来の、抗体を含有する液体サンプルまたは組織サンプルと、配列表 に配列番号11として示される所定量の精製されたポリペプチドとを接触させる 工程、 該ポリペプチドと該抗体との結合を検出する工程、および 該ポリペプチドと特異的に結合する抗体の存在と、該被検体におけるネコ引っ 掻き病とを相関させる工程、 を包含する、方法。 21.R.henselaeと特異的に結合する抗体の存在を検出することによって、被 検体における現在および以前のネコ引っ掻き病を診断する方法であって、ここで 、該検出工程が、 該被検体由来の、抗体を含有する液体サンプルまたは組織サンプルと、配列表 に配列番号11として示されるアミノ酸配列をコードする核酸と特異的にハイブ リダイズし得る核酸によってコードされる所定量の精製されたポリペプチドとを 、60℃および5×SSCのストリンジェンシー条件下、これに続く、室温、2×S SCおよび0.1%SDSである最初の洗浄条件下、および50℃、0.5×SSCおよび0.1 %SDSのストリンジェンシー条件での2回の第2の洗浄下で接触させる工程、 該ポリペプチドと該抗体との結合を検出する工程、および 該ポリペプチドと特異的に結合する抗体の存在と、該被検体におけるネコ引っ 掻き病とを相関させる工程、 を包含する、方法。 22.被検体における現在および以前のネコ引っ掻き病を診断する方法であって 、該被検体由来のサンプルにおけるRochalimaea henselaeの存在を検出する工程 を包含し、ここで、Rochalimaea henselaeの存在が、配列表に配列番号11とし て示されるアミノ酸配列をコードする核酸の存在を検出することによって決定さ れる、方法。 23.前記核酸が、核酸増幅技術を利用して検出される、請求項22に記載の方 法。 24.前記被検体がヒトである、請求項22に記載の方法。 25.被検体における細菌性血管腫症を診断する方法であって、 該被検体由来のサンプルにおけるRochalimaea henselaeの存在を検出する工程 であり、Rochalimaea henselaeの存在が、配列表に配列番号5として示されるア ミノ酸をコードする核酸の存在を検出することによって決定される、工程、およ び、 Rochalimaea henselaeの存在と、該被検体における細菌性血管腫症の存在とを 相関させる工程、 を包含する、方法。 26.被検体における細菌性血管腫症を診断する方法であって、 Rochalimaea quintanaに特異的な核酸の存在を検出することによって、該被検 体由来のサンプルにおけるRochalimaea quintanaの存在を検出する工程であり、 該Rochalimaea quintanaに特異的な核酸が、配列表に配列番号6として規定され るヌクレオチド配列中のヌクレオチドからなる、工程、および、 Rochalimaea quintanaの存在と、該被検体における細菌性血管腫症の存在とを 相関させる工程、 を包含する、方法。 27.被検体における細菌性血管腫症を診断する方法であって、 Rochalimaea quintanaに特異的な核酸の存在を検出することによって、該被検 体由来のサンプルにおけるRochalimaea quintanaの存在を検出する工程であり、 該Rochalimaea quintanaに特異的な核酸が、配列表に配列番号11として規定さ れるヌクレオチド配列中のヌクレオチドからなる、工程、および、 Rochalimaea quintanaの存在と、該被検体における細菌性血管腫症の存在とを 相関させる工程、 を包含する、方法。 28.被検体におけるネコ引っ掻き病または細菌性血管腫症を診断する方法であ って: (a)配列番号1、配列番号2、配列番号3および配列番号4からなるプライマ ー混合物を使用して該被検体由来のDNAを増幅する工程; (b)工程(a)由来の増幅DNAと配列番号5の核酸からなるプローブとを接触さ せ、そして該プローブと該増幅DNAとのハイブリダイゼーションを検出する工程 であり、ハイブリダイゼーションの存在がR.henselaeの存在を示し、これがネ コ引っ掻き病と相関づけられる、工程;および、 (c)工程(a)由来の増幅DNAと、配列番号6の核酸からなるプローブとを接触 させ、そして該プローブと該増幅DNAとのハイブリダイゼーションを検出する工 程であり、ハイブリダイゼーションの存在がR.quintanaの存在を示し、これが 細菌性血管腫症と相関づけられる、工程 を包含する、方法。
