Claims (1)
- 【特許請求の範囲】 1. 表面もしくは膜結合抗原または他のタンパク質をコードし、かつ所望の バクテリア産物の生産に対して温度調節的手法で発現される、プラスミド内遺伝 子を有するバクテリアの培養方法であって、 このバクテリアがそれらのプラスミドを保持し、その上なんらの発現も起こさ ない温度条件下で上記バクテリアを先ず培養基中で接種物へと培養し、次いで、 発現が起こる温度条件下で上記接種物を培養基中でさらに培養し、バクテリア を、それらのプラスミドを失う前に収集し、その後、所望の産物を分離すること を特徴とする、バクテリアの培養方法。 2. 接種物への上記最初の培養を室温で行い、上記のさらなる培養を哺乳動 物の体温で行う、請求項1記載の方法。 3. 上記室温が約20℃であり、哺乳動物の上記体温が34〜39℃である 、請求項2記載の方法。 4. 接種物への最初の培養を、先ず培養平板上で、次いで液体培養基中で行 う2段階工程で実施する、請求項1ないし3のいずれか1項記載の方法。 5. さらなる培養を液体培養基中で行う、請求項1ないし4のいずれか1項 記載の方法。 6. このバクテリアが、CFA/l、CS1、CS2、CS3、CS4、C S5およびCS6からなる群から選択されたコロニー化因子抗原の少なくとも1 つのタイプを発現するEscherichia coliである、請求項1ない し6のいずれか1項記載の方法。 7. 所望のバクテリア産物が、CFA/l、CS1、CS2、CS3、CS 4、CS5およびCS6からなる群から選択されたコロニー化因子抗原の少なく とも1つのタイプを保有するE.coliである、請求項6記載の方法。 8. 所望のバクテリア産物が、CFA/l、CS1、CS2、CS3、CS 4、CS5およびCS6からなる群から選択された少なくとも1つのコロニー化 因子抗原である、請求項6記載の方法。
