発明の名称
抗腫瘍剤としてのインシユリン状成長因子II
技術分野
本発明は腫瘍の治療のため、とくに皮膚黒色腫の治療用の薬剤の製造にIGF
−IIを使用することに関する。
技術的背景
イギリスにおける皮膚黒色腫の発生率はおよそ1年につき8000件で、毎年
増加しており、この病気の治療は通常効果がない。初期の黒色腫は手術で取り除
かれかつ生化学的治療を受けることができるが、これは僅かに成功するのみであ
る。最近の研究はインターロイキン−2を基礎にした型式がおよそ20%部分/
完全応答率を結果として生じ得ることを示したが散在性の腫瘍を治療するための
新規な治療を開発する必要がある。
毛様体および脈絡膜の黒色腫は成人における最も一般的な初期の眼球内悪性腫
瘍でありそして患者のおよそ半分において致命的である。イギリスにおける発生
率は1年につき7/100万である。死亡は、初期の治療に続いて15年以内に
発生し、そしてほとんど独占的に肝臓における転移が事実上すべての腫瘍関連死
亡の原因である。摘出または局部除去の必要をうまく回避することにより視力を
保つことができる技術の開発にも拘わらず、全体の患者の生存は変わらないまま
であつた。最も致命的な場合において、腫瘍散在は多分患者が治療に行く前
に発生した。超自然的な転移病の駆除に向けられる定義されない形の治療と結合
される、早期の腫瘍の広がりの認知、または転移を生じる多くの危険での患者の
識別は今までのところではこれらの腫瘍の将来の処理に関する我々の一番の希望
である。
インシユリン状成長因子II(IGF−II)は血漿および他の体液中に存在
するペプチドである。それは、3個の二硫化物結合を含む67のアミノ酸からな
りかつその最初のシーケンスはIGF−1に対して64%の同族関係を示す。そ
れは細胞型の広い範囲の成長を刺激することが可能である。IGF−IIはヒト
血漿から精製されかつ完全なアミノ酸シーケンスが知られている。ヒトIGF−
IIに対する広範囲の同族関係を有するシーケンスが他の種の血漿から精製され

インシユリン状成長因子IGF−IおよびIGF−IIはインシユリンに対す

示された。それらは多数の胎児のおよび未発達の細胞型における差異を支持する
ことができかつ幾つかの癌に関して成長パターンはIGF−IおよびIGF−I
I遺伝子発現を反映するかも知れない。
ハインツ−エリアン氏等著、内分泌学、(1991年)第129巻、第4号、
第1769頁は、細胞分裂促進および成長の規則において両IGFレセプターの
必須の役割があることを報告する。

今では容易に達成され得る。
シヨーフイールドおよびエングストローム氏はインシユリン状成長因子の構造
および生物学的機能、第240頁乃至第257頁(1992年、OUP)におい
て広範囲のヒト腫瘍によるIGF−IIの発現に関する論評を掲載している。
IGF−II腫瘍発現における高度の関心の理由は
1)肝外成人組織が通常多量のIGF−IIを作らないためおよび
2)IGF−IIが多数の細胞型の有糸分裂促進物質であるためである。
論理的結論はIGF−IIが腫瘍成長を開始または促進し得るかまたはし得な
い予定外の細胞増殖を発生するように作用するかも知れないということである。
しかしながらデータは系統から系統へ異なり、そして記述的なデータから潜在的
なまたは実際のオート/パラクリン成長を示すことができるけれども、細胞培養
または異種移植片培養に依存して、実験的データは腫瘍成長に関してIGF−I
Iの作用を制限するそれらの潜在率の評価において異なる。
シヨーフイールド氏等(英国雑誌、キヤンサー、第63巻、第687頁乃至第
692頁、1991年)は、腫瘍成長についてのIGF−IIの潜在的な役割を
検査するために、IGF−IIcDNAを含有しかつ発現する組み換えレトロウ
イルスを構成しかつこれをクローニングされた腫瘍形成のマウス繊維芽細胞細胞
線に感染させるのに使用した。細胞がヌードマウスに感染させられたときDNA
構成からIGF−IIを発現するこれらのクローンは親の、非IGF発現細胞線
より非常に長い潜伏期間により腫瘍を起こすことが認められた。公表された実験

されるIGF−IIが腫瘍成長に関して抑制または遅延作用を有するがIGF−
Iがこの作用を持たないことを示した。これは予期しないかつ意外なことであり
そして以前に公表された結果により予測されなかつた。IGF−IIは細胞増殖
抑制または細胞破壊剤よりむしろ有糸分裂促進物質であるように伝統的に支持さ
れる。したがつてこの結果は完全に直観に反している。日付けを記載すべきIG

使用されたIGF−IIはカビ・フアーマシアにより製造された。
発明を実施するための最良の形態
例1
生体外のIGF−IIの成長禁止作用
ヒト黒色腫細胞線(A375)成長についてのIGF−IIの作用を評価する
ために予定された一連の実験において、予備研究において注目されたことはIG
F−IIが成長抑制であつたということである。
a)IGF−IIは3本の細胞線、すなわちヒト黒色腫細胞線A375細胞、
A375の生体内通路により引き出された副線、NUPRIおよび後方ぶどう膜
メラニン細胞、Me166に添加された。細胞線は結成なしの媒体中で微量測定
プレートにおいて成長させられかつDNA合成が3H−チミジンにより細胞を拍
動することにより48時間で評価分析された。広範な一連の実験においてIGF
−IIの10ng/mlまたはより大きい濃度が3H−チミジン摂取により測定
されるような細胞増殖を禁止すべく示された。表1参照。
b)IGF−IIの反増殖作用が流動血球計算分析により評価されるような遅
いS−相または早いG2/Mの間中取り次がれた。これらの実験においてIGF
−II治療A375細胞はミトロマイシンおよび臭化エチジウムにより染色され
かつ細胞サイクルの増殖相が分析された。結果は細胞サイクル中のG2/Mブロ
ツクを連想させる。他の3本の黒色腫細胞線(A375,NUPRIおよびMe
166の副クローン)を使用するさらに他の研究において3本の線の2本がまた
IGF−IIにより成長禁止を受け易かった。A375による結果に関して表2
を参照。
我々の結果は皮膚黒色腫細胞線の成長がIGF−IIにより調節され、そして
この作用が多分型2レセプター
結合部位により発生することを示す。
また、IGF−IIおよびこの配位子の黒色腫細胞への結合に続いている信号
変換が細胞増殖に鋭気用を及ぼすことが示され、そして明らかにこの知見が潜在
的な治療意義からなる。
例2
混合された腫瘍中にペプチドを発現するトランス効果
腫瘍細胞成長の抑制が、IGF−IIについて期待され得るような、分泌され
る徴候により取り次がれたかどうかまたは構成からの発現が細胞のアントノマス
特性に作用していたかどうかの調査。
この例はシヨーフイールド氏等の以前の研究を確認しかつ発現IGF−IIの
効果がトランス中にあり(すなわち拡散性因子を通って)そして投与反応に従う
ことを示す。
異なる比率において細胞を含有している腫瘍形成および非腫瘍形成構成がヌー
ドマウスに注入する前に混合された。腫瘍形成細胞の数は5×108/移植片に
維持されかつ抑制された細胞が20×この数まで加えられた。対照実験はこれが
接種材料の大きさに依存様式において抑制された細胞の腫瘍潜伏を減少する一方
非トランスフエクシヨン細胞の潜伏の増加が不釣り合いであることを示した。
一定数の腫瘍形成細胞(5×108)が第1図に付与された示された比率で等
しいかまたはより大きい数の抑
制された細胞に沿って単一の部位に接種された。Balb/cは親腫瘍形成クロ
ーン細胞を意味し
Bal12はIGF−II発現細胞を意味し
Balb/c/Bal12はそれらの細胞の混合物を意味する。
Bal9はIGF−IIならば低レベルを発現する細胞を意味し
Bal9/Bal12はそれらの細胞の混合物を意味し異なる接種材料でのBa
l12は対照として使用され、すなわち混合された移植片におけると同一の細胞
の数が各部位に単独で置かれた。
第1図において腫瘍形成細胞が種々の比率において正常な細胞とともに接種さ
れたときの潜伏日数が示される。
第1図に示されたデータは増加する抑制されたIGF−II分泌細胞の数が親
細胞線の腫瘍形成を低減可能であることを示す。これはIGF合成に関連付けら
れない代謝禁止相互作用または細胞を支持するIGF−II構成のトランス作用
特性に依存することが可能であつた。Bal9細胞線を発現する低いIGF−I
Iの使用はより少ない腫瘍形成移植片が、親Balb/c線に比較されるとき、
抑制された細胞の非常に低い入力で抑制された細胞により相互に移稙される抑制
効果をより受け易いことを示した。これはトランス効果がIGF−II発現の直
接的なまたは間接的な結果であることを示唆する。
例3
腫瘍潜伏についての外因性IGF−IIの効果
5×106の細胞が通常の肩甲骨上の部位でマウスに移植されかつアルゼツト
浸透性小型ポンプが、近くに移植された、燐酸塩緩衝塩水50μg/mlBSA
、牛血清アルブミン、(RIA等級)中に溶解されたIGF−IIにより負荷さ
れた。幾つかの場合において短いカテーテル管がポンプの端部に取着されかつ単
一の縫合を経由してそれを安定化するために下に横たわる筋肉層に取着された。
各場合において移植片へのポンプ出力の方向および近接が、移動が発生しないこ
とを保証するために、12時間後およびその後毎日チエツクされた。7日後ポン
プが移植の後部の後ろに小さい切り込みを作ることにより取り除かれかつ腫瘍部
位が24時間間隔での触診により監視された。腫瘍形成は触診できる塊りが皮膚
の下に感じられ得る2日間の第1日として記録された。切除時これは通常直径約
2〜4mmの別個の小さい腫瘍塊りからなるように認められた。
Bal12*は選択的な媒体の不存在において培養中の連続する通路に続いて
そのIGF−II構成を自然に失うBal12の線である。潜伏期は移植と第1
触診との間の日数において抑制される。各データ点が5匹の個々のマウスにおけ
る腫瘍の平均潜伏期である。グループ内の潜伏期の間の変化は各場合に平均の1
0%以内であつた。
5×106Balb/c親細胞が日数ゼロで7引きの
マウスに接種された。2匹に浸透性小型ポンプ(0.47μl/時間が21.5
ngIGF−II/時間供給すべく負荷される)が配置され、2匹が42ng/
時間供給するポンプを受容しかつ単一のマウスが83ng/時間供給するポンプ
を受容した。マウスは毎日の触診および以前に注目されたような第1の腫瘍触診
の時間により監視された。
第2図において腫瘍潜伏期についての外因性IGF−IIの効果が一定数の腫
瘍形成細胞(5×106)が示された比率において等しいまたはより大きな数の
抑制された細胞に沿って単一部位に接種されたときで示される。対照(Bal1
2)として混合された移植片におけると同一数の細胞が各部位に単独で置かれた
。
我々は7日間にわたつて供給されるIGF−IIの潜伏期の投与傾向を示すこ
とができた。高い相関関係(r2=0.753および相関係数0.868)によ
り増加したIGF−IIの投与が減少した親細胞線の腫瘍形成を結果として生じ
る。
