- 【特許請求の範囲】 1.4重量パーセントを上回る酵母固形分を含有する、少なくとも約1,50 0リットルという容積の栄養素培地において、適当な条件で培養するとき、酵母 乾燥固形分を基準に少なくとも約3,000ppmのアスタキサンチンを産生するP haffia rhodozyma菌株。 2.酵母乾燥固形分が6重量パーセントを上回る請求項1の菌株。 3.栄養素培地の容積が少なくとも30,000リットルである請求項2の菌 株。 4.酵母乾燥固形分を基準に少なくとも約4,000ppmのアスタキサンチン を産生する請求項3の菌株。 5.4重量パーセントを上回る酵母固形分を含有する、少なくとも約1,50 0リットルという容積の栄養素培地において、適当な条件で培養するとき、酵母 乾燥固形分を基準に少なくとも約3,000ppmという量のアスタキサンチンを 産生する能力を保持しているATCC−74218,ATCC−74219,A TCC−74220およびATCC−74221として寄託されている菌株、な らびにその突然変異体および誘導体より成る群から選ばれる請求項1の菌株。 6.菌株が、(i)急速にその細胞が数を増して、アスタキサンチンを産牛し 、かつエネルギー源が所定のレベルを上回るほどは培地に蓄積されないような速 度で菌株に給送される生育期に続き(ii)細胞数の増加は減じるが、アスタキサ ンチンの産生は生育期間中の速度と少なくとも同等の速度で継続する成熟期を経 験するようなレベルに、培養の少なくとも一部が終るまで保たれる急速に代謝さ れるエネルギー源を含有する栄養素培地において、適当な条件で菌株の細胞を培 養することを含むPhaffia rhodozyma菌株からアスタキサンチンを生成させる改 良発酵法において、成熟期を延長させることを含む改良法。 7.菌株を光源に曝露することによって成熟期を延長させる請求項6の方法。 8.光源が、発酵ブロス1キロリットル当り約100ワットを下回る光を菌株 に与える請求項7の方法。 9.露光が成熟期の初めから発酵が終るまで実質的に継続する請求項8の方法 。 10.光源が約250から700ナノメートルの間の波長の光を菌株に与える請 求項9の方法。 11.生育期の終りに、菌株への急速に代謝されるエネルギー源の給送速度を低 下させることによって、成熟期を延長させる請求項6の方法。 12.生育期の終りに菌株にエネルギー源を給送した速度の僅か約50%にエネ ルギー源の給送速度を低下させる請求項11の方法。 13.エネルギー源がグルコースである請求項12の方法。 14.成熟期の初めに、急速に同化されるエネルギー源を、徐々に同化されるエ ネルギー源で置き換えることによって成熟期を延長させる請求項6の方法。 15.急速に代謝されるエネルギー源がグルコースであり、徐々に代謝されるエ ネルギー源がグリセロール;糖みつやコーンシラップ中に存在するグルコース、 スクロースや他の多糖類の重合形態;一価アルコール類;およびカルボン酸類よ り成る群から選ばれる請求項14の方法。 16.徐々に代謝されるエネルギー源が、発酵の生育期の間、急速に代謝される エネルギー源とともに菌株に給送される請求項15の方法。 17.徐々に代謝されるエネルギー源が、急速に代謝されるエネルギー源の給送 停止後に菌株に給送される請求項15の方法。 18.請求項6の方法でつくったアスタキサンチン。 19.請求項7の方法でつくったアスタキサンチン。 20.請求項11の方法でつくったアスタキサンチン。 21.請求項14の方法でつくったアスタキサンチン。 22.請求項1の菌株からつくったPhaffia rhodozyma酵母クリーム。 23.請求項22の酵母クリームからつくった乾燥生成物。 24.可食酸化防止剤、食用油、および可食乳化剤を含む請求項23の乾燥生成 物。 25.乾燥生成物の重量を基準に、酸化防止剤の濃度が約0.02ないし約2重 量パーセントの間、油の濃度が約0.1ないし約10重量パーセントの間、およ び乳化剤の濃度が約0.1ないし約10重量パーセントの間である請求項24の 乾燥生成物。 26.酸化防止剤が、エトキシキン、ビタミンE、アスコルビルパルミテート、 ブチル化ヒドロキシアニソール、およびブチル化ヒドロキシトルエンより成る群 から選ばれる請求項25の乾燥生成物。 27.油がベニバナ油、コーン油、ヒマワリ油、大豆油、および前記油の2種以 上の混合物より成る群から選ばれる請求項26の乾燥生成物。 28.乳化剤が大豆レシチンおよびソルビタンエステル類より成る群から選ばれ る請求項27の乾燥生成物。 29.酵母細胞の過半数が破壊されていない請求項23の乾燥生成物。 30.酵母クリームを噴霧乾燥またはドラム乾燥して調製した請求項29の乾燥 生成物。 31.酵母細胞の過半数が破壊されている請求項23の乾燥生成物。 32.酵母細胞が微粉砕されている請求項31の乾燥生成物。 33.請求項23の乾燥生成物を含む動物飼料。 34.請求項24の乾燥生成物を含む動物飼料。 35.動物飼料の重量を基準に、約0.1ないし約10重量パーセントの間の該 乾燥生成物を含む請求項34の動物飼料。 36.動物飼料の重量を基準に、約1ないし約3重量パーセントの間の該乾燥生 成物を含む請求項34の動物飼料。 37.サケ科魚類、甲殻類およびタイ科魚類より成る群から選ばれる動物用に配 合された請求項35の動物飼料。 38.動物の飼養法において、請求項23の飼料を動物に与えることを含む方法 。 39.動物が、サケ科魚類、甲殻類およびタイ科魚類より成る群から選ばれる請 求項38の方法。
