Claims (1)
- 【特許請求の範囲】 1.遺伝子型物質および表現型物質の各々の特異的結合を目的とした少なくとも 2つの同時結合性有する基質材料。 2.前記基質材料の特定の表面領域に遺伝子型物質を結合し、かつ、該基質材料 の局所的に分離した表面領域に表現型物質を結合しうるか、または該基質材料の 同じ領域が遺伝子型および表現型物質の両方を結合する事を特徴とする請求項1 記載の基質材料。 3.前記核酸が遺伝子型物質として結合し、かつタンパク質またはペプチドが表 現型物質として同時に結合することを特徴とする請求項1および2のいずれか1 項記載の基質材料。 4.前記遺伝子型および表現型を結合する各々の領域が互いに10〜1000μm、望 ましくは10nm〜100μm離れていることを特徴とする請求項1〜3の少なくとも1 項記載の基質材料。 5.前記領域がイオン交換能または疎水相互作用などの親和性または複合体形成 性、またはこれらの組み合わせた性質を有することを特徴とする請求項1〜4の 少なくとも1項記載の基質材料。 6.前記領域型の1つが親和性を有し、かつ、他の領域型がイオン交換性を有す ることを特徴とする請求項5記載の基質材料。 7.前記親和性を有する領域が結合する遺伝子型または表現型と可逆的な相互作 用をすることで、結合する分子型の少なくとも1つの溶出が可能であることを特 徴とする請求項1〜6記載の少なくとも1項記載の基質材料。 8.前記基質材料がメンブレンまたはメンブレン集合体上に二次元的に存在する か、またはメンブレン中に埋設されているか、または、キャピラリー中または繊 維または不織布のような繊維状混成構造物上に存在するか、またはこれらの組み 合わせ物上に存在することを特徴とする請求項1〜7の少なくとも1項記載の基 質材料。 9.前記親和性を有する物質およびアニオン交換物質が不織布などのマトリクス 中に存在することを特徴とする請求項1〜8の少なくとも1項記載の基質材料。 10.前記親和性を有する領域が少なくとも1つのタンパク質またはペブチドを特 異的に結合し、かつ、同時に核酸を結合しうることを特徴とする請求項1〜9の 少なくとも1項記載の基質材料。 11.前記領域がイオン交換性を有することを特徴とする請求項1〜10の少なくと も1項記載の基質材料。 12.前記アニオン交換性を有する領域が表面処理された大孔または閉孔粒子によ って提供されることを特徴とする請求項1〜11の少なくとも1項記載の基質材料 。 13.前記アニオン交換性を有する領域が、さらに、1M NaClに相当するイオン強 度条件下でもDNA分子を結合しうることを特徴とする請求項11および12のいずれ か1項記載の基質材料。 14.前記親和性を有する領域がビオチン誘導体、ストレプトアビジン誘導体、ア ビジン誘導体、核酸、抗体断片、金属キレート、タンパク質および/またはペプ チドリガンドで構成されることを特徴とする請求項1〜13の少なくとも1項記載 の基質材料。 15.前記親和性を有する領域がニッケル/ニトリロトリ酢酸キレートに基づく金 属キレートで構成され、かつ、アニオン交換性を有する領域が核酸を結合しうる 材料で構成されていることを特徴とする請求項1〜14の少なくとも1項記載の基 質材料。 16.生体高分子の進化論的最適化の方法であって、遺伝子型および表現型を1つ の基質材料に同時に結合し、ついで選択的または同時にこれらを単離する方法。 17.1つの反応容器中で行われる請求項16記載の方法であって、遺伝子型を提示 する構造型に突然変異を誘発し、少なくとも1回の該構造型の複製ステップを介 し、発現後、該発現産物を遺伝子型と共に基質材料に結合し、発現産物の評価を 行うことを特徴とする請求項16記載の方法。
