Claims (1)
- 【特許請求の範囲】 1.DNA配列のある標的位置の塩基を同定する方法にして、試料DNAを増幅 処理に付し、その増幅DNAを固定化した後で二本鎖解離処理に付して、標的位 置の直ぐ隣にハイブリダイズするような伸長プライマーから非固定化鎖を取り除 いておき、固定化された一本鎖DNAの4つのアリコートの各々を1種類のジデ オキシヌクレオチド存在下でのポリメラーゼ反応に付し、その際、各アリコート について異なるジデオキシヌクレオチドを用いて標的位置の塩基に相補的なジデ オキシヌクレオチドだけが組込まれるようにしておき、次に、4つのアリコート を4種類すべてのデオキシヌクレオチドの存在下での伸長反応に付して、各アリ コート中においてジデオキシヌクレオチドと反応していないDNAが伸長して二 本鎖DNAを形成する一方で、ジデオキシヌクレオチドでブロックされたDNA は非伸長鎖DNAとして残るようにしておき、しかる後に、二本鎖及び/又は非 伸長鎖DNAの同定を行なって、どのジデオキシヌクレオチドが導入されていて 、したがってどの塩基が標的位置に存在しているのかを明らかにすることを特徴 とする方法 2.請求項1記載の方法にして、固定化されているか或いは固定化用手段の備わ っている第一のプライマーを用いるインヴィトロ増幅反応によって、試料DNA を増幅することを特徴とする方法。 3.請求項2記載の方法にして、前記第一プライマーが、標識保有DNA結合タ ンパク質に対する認識部位を二本鎖形のときに含んでいる領域を有していて、鎖 伸長反応による二本鎖DNAの形成を上記標識タンパク質への結合によって同定 することを特徴とする方法。 4.請求項2又は請求項3記載の方法にして、前記第一プライマーが固定化用手 段としてビオチンを有することを特徴とする方法。 5.請求項2乃至請求項4のいずれか1項記載の方法にして、前記インヴィトロ 増幅に、前記標的DNAの3′末端側の領域A及び/又はその領域Aからさらに 3′側に伸びた領域Bにハイブリダイズする第二のプライマーであって、上記標 的配列のA及び/又はBの少なくとも一部分にハイブリダイズする3′末端配列 を有していてかつその5′末端にはAと実質的に同一の配列を有する第 二のプライマーを使用し、この増幅処理によって、標的配列の3′末端に、領域 Aとループを形成し得る領域と配列Aに相補的な領域A′とを以上の順序で有す る二本鎖標的DNAを生じさせ、しかる後に、増幅二本鎖DNAを固定化形で二 本鎖解離処理に付して、そうすることによって固定化されていない標的鎖を遊離 させて、領域A′を領域Aに自然に或いは人為的にハイブリダイズさせてループ を形成させることを特徴とする方法。 6.請求項1乃至請求項5のいずれか1項記載の方法にして、二本鎖DNAの形 成を、鎖伸長反応中に放出されたピロリン酸の検出又は推計によって同定するこ とを特徴とする方法。 7.請求項6記載の方法にして、発光をピロリン酸のインジケーター又は指標と するルシフェリン/ルシフェラーゼ反応によってピロリン酸を検出又は推計する ことを特徴とする方法。 8.請求項1記載の方法を実施するためのキットにして、少なくとも以下の構成 成分: (a)その検査に特異的な伸長プライマーであって、前記標的位置がそのプラ イマーの3′末端の直ぐ隣になるように試料DNAにハイブリダイズする伸長プ ライマー; (b)ポリメラーゼ; (c)ジデオキシヌクレオチドとデオキシヌクレオチド;及び (d)任意成分としての固体担体 を含んでなることを特徴とするキット。 9.請求項8記載のキットにして、さらに少なくとも以下の構成成分: (i)少なくとも一方のプライマーがそのプライマーを固定化するための手段 を有している一対のPCR用プライマー; (ii)ポリメラーゼ、好ましくはTaq1ポリメラーゼのような耐熱性のポリ メラーゼ; (iii)PCR反応用の緩衝液;及び (iv)デオキシヌクレオチド をも含んでいることを特徴とするキット。
