Claims (9)
- 1.全血サンプル中のアナライトの存在、欠如、あるいは量を検出する横方向流 デバイスであって、一般的に平面的な配置で: i)第一の固体多孔性マトリックスを含有し、横方向流でii)と接続されるサ ンプル受容ゾーン、 ii)該アナライトと結合する能力あるいは捕獲試薬に対して該アナライトと兢 合する能力を有する標識試薬を含有する第二の固体多孔性マトリックスを含有し 、横方向流でiii)と接続される標識ゾーン、 iii)標識アナライトと特異的に結合するか、あるいはアナライトと兢合する 標識と特異的に結合する捕獲試薬を含有する第三の固体多孔性マトリックスを含 有し、横方向流でiv)と接続される捕獲ゾーン、 iv)吸収ゾーン; を包含するデバイス: ここで、該サンプル受容ゾーン、および標識ゾーンの少なくとも1つが、赤血球 表面抗原と結合する試薬を含有する。
- 2.前記サンプル受容ゾーンが、赤血球表面抗原と結合する試薬を含有する、請 求項1に記載のデバイス。
- 3.前記標識ゾーンが、赤血球表面抗原と結合する試薬を含有する、請求項1に 記載のデバイス。
- 4.前記標識試薬が、前記のアナライトと結合するように前記アナライトに対す る特異的結合パートナーと結合した可視部分を含有する、請求項1に記載のデバ イス。
- 5.アナライトに対する前記特異的結合パートナーが、モノクローナル抗体、ポ リクローナル抗体、およびそれらのF(ab′)2、Fab′およびFabフラ グメントからなる群がら選択される、請求項4に記載のデバイス。
- 6.前記標識試薬が、前記捕獲試薬に対する前記アナライトの競合物と結合した 可視部分を含有する、請求項1に記載のデバイス。
- 7.第一、第二および第三固体多孔性マトリックスが非吸収性である、請求項1 に記載のデバイス。
- 8.赤血球表面抗原に結合する前記試薬が、モノクローナル抗体、ポリクローナ ル抗体およびそれらのF(ab′)2、Fab′およびFabフラグメントから なる群から選択される、請求項1に記載のデバイス。
- 9.以下の工程を包含する全血サンプル中のアナライトの存在、欠如または量を 決定する方法: (a)全血サンプルを i)個体多孔性マトリックスを含有し、横方向流でii)と接続されるサンプル 受容ゾーン、 ii)該アナライトと結合する能力あるいは捕獲試薬に対して該アナライトと競 合する能力を有する標識薬を含有する固体多孔性マトリックスを含有し、横方向 流でiii)と接続される標識ゾーン、 iii)標識アナライトと特異的に結合するか、あるいはアナライトと競合する 標識と特異的に結合する捕獲試薬を含有する固体多孔性マトリックスを含有し、 横方向流でiv)と接続される捕獲ゾーン、 iv)吸収ゾーン; と接触させる工程、(ここで、該サンプル受容ゾーン、および標識ゾーンの少な くとも1つが、赤血球表面抗原と結合する試薬を含有する)、および、 (b)アナライトの存在、欠如または量の関数として標識の結合を観察する工程 。
