Claims (18)
- 1.遺伝子プロモーター配列、コーディング領域及び遺伝子ターミネーターを順 次に有する植物DMからなる組換え体DNAであって、上記コーディング領域は 、リグニンの生合成に必要な酵素をエンコードする内在植物遺伝子又はその一部 によってエンコードされるmRNAと実費的に相同性であるか又は相補性である mRNAをエンコードするヌクレオチド配列からなり、従って、形質転換により 植物ゲノム中に組込まれた場合、上記コーディング領域から転写されたmRNA は内在植物遺伝子からの前記酵素の産生を抑制するものである、組換え体DNA 。
- 2.コーディング領域は前記の内在遺伝子によってエンコードされるmRNAに 対してアンチセンス配向にあるmRNAをエンコードする、請求の範囲1に記載 の組換え体DNA。
- 3.コーディング領域は前記内在遺伝子をエンコードするDNAの非転写鎖から 単離される、請求の範囲2に記載の組換え体DNA。
- 4.コーディング領域は前記の内在遺伝子と同一の配向である、請求の範囲1に 記載の組換え体DNA。
- 5.コーディング領域は50塩基の最小寸法を有する、請求の範囲1〜4のいず れかに記載の組換え体DNA。
- 6.前記酵素はシンナミルアルコールデヒドロゲナーゼ(CAD)、シンナモイ ル:CoAレダクターゼ(CCR)及びカテコール−O−メチルトランスフェラ ーゼ(COMT)からなる群から選択される、請求の範囲1〜5のいずれかに記 載の組換え体DNA。
- 7.プロモーターはCaMV35S、GPAL2、GPAL3及び内在CAD遺 伝子の発現を制御する内在植物プロモーターからなる群から選択される、請求の 範囲1〜6のいずれかに記載の組換え体DNA。
- 8.遺伝子プロモーター配列、コーディング領域及び遺伝子ターミネーターを順 次に有する植物DNAからなる組換え体DNAであって、上記コーディング領域 は、リグニンの生合成に必要な酵素をエンコードする内在植物遺伝子又はその一 部によってエンコードされるmRNAと実質的に相同性であるか又は相補性であ る,mRNAをエンコードするヌクレオチド配列からなり、従って、形質転換に より植物ゲノム中に組込まれた場合、上記コーディング領域から転写されたmR NAは内在植物遺伝子からの前記酵素の産生を抑制するものである組換え体DN Aを、形質転換により植物のゲノム中に安定に組込むことからなる、植物内での リグニンの生合成を抑制するか又は変更する方法。
- 9.リグニンを形成するための通常の能力より低い能力を有する形質転換植物で あって、上記植物はそのゲノム中に請求の範囲1〜7のいずれかに記載の組換え 体DNAが安定に組込まれていることを特徴とする形質転換植物。
- 10.植物内はアルファルファ、トウモロコシ、アブラナ、ユーカリ、ポプラ、 ロリウム又はフェスツカである、請求の範囲9に記載の形質転換植物。
- 11.英国、アバーデイーン所在のThe National Collect ion ofIndustrial and Marine Bacteria に、E.coli菌株 XL1 Blueを宿主として、それぞれ、1991年 4月17日に40404の寄託番号で、1991年4月8日に40401の寄託 番号で寄託されているプラスミドpTCAD14又はpTCAD19から誘導さ れたタバコCAD遺伝子及びこれを含有する組換え体DNA。
- 12.英国、アバーデイーン所在のThe National Collect ion ofIndustrial and Marine Bacteria に、E.coli菌株 XL1 Blueを宿主として、1992年4月2日に 40501の寄託番号で寄託されているプラスミドpZCAD1から誘導された トウモロコシCAD遺伝子及びこれを含有する組換え体DNA。
- 13.英国、アバーデイーン所在のThe National CoHecti on ofIndustrial and Marine Bacteriaに 、E.coli菌株 XL1 Blueを宿主として、1992年4月2日に4 0500の寄託番号で寄託されているプラスミドpPOPCAD1から誘導され たポプラCAD遺伝子及びこれを含有する組換え体DNA。
- 14.英国、アバーデイーン所在のThe National Collect ion ofIndustrial and Marine Bacteria に、E.coli菌株 XL1 Blueを宿主として、1992年4月2日に 40502の寄託番号で寄託されているプラスミドpEUCAD1から誘導され たユーカリCAD遺伝子及びこれを含有する組換え体DNA。
- 15.プロモーターGPAL2の制御下にあるアンチセンスCAD遺伝子を含有 する組換え体DNA。
- 16.プロモーターCPAL3の制御下にあるアンチセンスCAD遺伝子を含有 する組換え体DNA。
- 17.プロモーターCaMV35Sの制御下にあるアンチセンスCAD遺伝子を 含有する組換え体DNA。
- 18.内在CAD遺伝子を制御する内在植物プロモーター制御下にあるアンチセ ンスCAD遺伝子を含有する組換え体DNA。
