More specifically, since the area of the opening 1 has a size of about 20 to 80 cm 2 , it is extremely easy to insert and remove the medium 2 and the tissue to be cultured 3, and the opening 1 Since the surface has an angle of 60 ° to 90 ° with respect to the vertical, it is easy to put the medium 2 and the tissue to be cultured 3 in and out, and prevent bacteria and foreign substances from dropping into the container from above, and Breeding does not occur, aeration is performed between the inside and the outside of the container, and the amount of ventilation can be adjusted, the containers can be stacked, and a shelf is not required when arranging them in the vertical direction. The tissue culture container has a greater culturing capacity than that of 10 to 30 mm and can be used even when the tissue 3 to be cultured grows vertically upwardly to a maximum height of 110 mm. .

After the medium 2 has cooled to a predetermined temperature (for example, 20 ° C.), the dew on the inner wall of the main body 30 is wiped off with sterilized and dry tissue paper. A clean bench for transplanting the tip of the stem of the male plant of asparagus placed in the tissue culture container 100 and the dish prepared in this way (with an operation box having a glass door that opens and closes at the front, It can be opened halfway and the transplanter can perform the transplantation manual work in the box through the lower opening.) 20, and the glass door 21 of the clean bench 20 is lowered so that the worker can perform the manual work. The operator uses a gas burner (not shown) in the clean bench to sterilize a main part of the tissue culture container 100, for example, the opening 1 by stacking.

Then, the tip of the stem of the male strain of asparagus, which is the tissue to be cultured 3, is placed at a predetermined position of the medium 2 inside the main body 30 through the knob opening 1 with the pincette. The lid 50 having the packing 14 mounted thereon is screwed onto the neck portion 9 of the main body 30 to complete the transplanting operation. At the time of this transplantation, it is desirable that the inside of the clean bench is shielded from the outside as much as possible in order to protect the aseptic condition.
Should be as low as possible. In this tissue culture container 100, the opening 1 is opened in the lateral direction, and the movement of the worker's hand is substantially horizontal in the transplanting work. Therefore, the glass door 21 of the clean bench 20 is opened by the amount of the worker's arm. The transplant work is possible only by itself. Therefore, the work is simple and the sterile condition is well protected.

When culturing is continued for about 1 month, asparagus grows like hair buds. Here, the buds are transferred to another tissue culture vessel containing a growth medium containing a hormone of special composition for growing. At this time, the sprouted asparagus is taken out from the opening 1 using tweezers, and then transferred into another tissue culture container containing a growth medium through the opening. After this, asparagus sprouts grow 5 to 10 times in culture for about one month.

The tissue culture container 100 is used as described above. However, since the two side walls 6, 6'and the rear wall 7 are perpendicular to the bottom wall 7, the medium area can be widely used.
Further, since the container 30 has a sufficient height H10, it can also be used for the tissue to be cultured 3 that grows strongly vertically upward.
Further, since the area of the opening 1 is sufficiently large, the medium 2 and the tissue to be cultured 3 can be easily put in and taken out, and the fact that the surface of the opening 1 is slightly upward means that the container 100 is placed on the table of the clean bench 20. Place the cultured tissue 3 on the container 10
It is very suitable for wrist movement when putting in and out of 0. In addition, the amount of the medium 2 is sufficient and the culturing ability is extremely large.

A vent hole 15 is provided in the lid 50 to allow ventilation inside and outside the container 100.
The air flow rate can be adjusted by packing a breathable filler, for example, glass fiber, a foamed resin composed of open cells, specifically, a plug having a specific structure made of silicon foam, ceramics, or the like.

Further, the lid 50 and the packing 14 are easy to sterilize, and no germs propagate in the vicinity of the screwed portion with the main body 30 and airtightness can be maintained. The tissue culture container can be used not only when the tissue to be cultured is a plant but also when it is an animal. Also, the opening can be used as the upper surface.
