128の重鎖および軽鎖のヌクレオチド配列およびアミ
ノ酸配列、およびトロンボスポンジンと結合した抗原。
活性化した血小板が関与する疾患、例えば血栓症、具体
的には血栓分解治療における心筋梗塞または発作の場合
の血栓症、深静脈血栓症、肺塞栓症、血管損傷、アテロ
ーム性動脈硬化症性斑、化膿性感染症および腸虚血のイ
ン・ビボでの検出および治療に特に好適である。
Description
合する単クローン性抗体および部分、単クローン性抗体
MAbBW2128の重鎖および軽鎖のヌクレオチド配
列およびアミノ酸配列、およびトロンボスポンジンと結
合した抗原に関する。ハイブリドーマ技術により、画定
したまたは未画定の抗原上の任意の所望なエピトープに
対し、選択的な単クローン性抗体(MAbs)を作成す
ることが可能になっている。これに用いることができる
免疫原は、高純度の画定された抗原或いは抗原混合物、
細胞または組織であり、選択性の高いMAbsを見出す
ためには、好適な免疫原を見出すことも必要である。こ
の技術を用いることにより、静脈注射の後に、例えば人
体の腫瘍、炎症工程および血管閉塞に結合することがで
きるMAbsを開発することが可能になってきている。
MAbsの対応する標的構造(腫瘍、炎症工程、血管閉
塞)へのこの優先的な結合は、核医学診断において既に
用いられている。標的構造を検出するための放射能標識
したMAbsの注射を扱う具体的な核医学的方法は、イ
ムノシンチグラフィである(Mach, J.P.,Buchegger,
F., Forni, M., Ritschard, J., Berche, C., Lumbras
o, J.-O., Schreyer, M., Girardet, C., Accolla, R.
C., Carrel, S., (1981), Immunologytoday, 2, 239-24
9 )。現在のところ、血小板に選択的に結合し、他の正
常なヒトの組織には本質的に結合しないMAbsは、こ
れには利用されていない。しかしながら、血小板のα−
顆粒タンパク質に向けられ且つ実際に活性化した血小板
に優先的に結合するMAbsでも、他の組織上のエピト
ープへのはっきりした結合を示す。例えば、トロンボス
ポンジン(TSP)、すなわちα−顆粒パク質(Baenzi
ger, N.L., Brodie, G.N., Majerus, P.W. (1971), Pro

るMAbsは、皮膚および肺の腺上皮、真皮と表皮との

SPMAbsを調製する際に、本発明者らは意外にも、
活性化した血小板に対する特異的MAb(MAb BW
2128)は、Towbin T. およびGordon J. ((197

って記載されたウェスターンブロットにおいて、TS

たは組織とも反応しないことを見出した。MAbは、T
SPと結合した抗原を免疫沈澱させ、等電点は実験誤差
の限界内ではTSPとは識別できないが、還元条件下で
は約160kDaのTSPよりも明らかに小さな分子量
を有する。

(このTSP調製物は、他の血小板タンパク質が混入す
ることがある。)生成するハイブリドーマの上澄液を、
間接APAAP法を用いて低温保存したヒト組織につい
ての特異性について検討を行った(Cordell, J.L., Fal
ini, B., Erber, W.N.ら、(1984), J. Histochem. Cyto
chem., 32, 219)。通常は、これらの上澄液は、活性化
したヒト血小板、肝臓の管上皮、腎臓の血管、肺上皮、
脾臓および骨髄(巨核球)と反応した。それ故、これら
は文献既知のTSP特異性を示した。しかしながら、意
外なことには、活性化したヒト血小板と特異的に反応す
るが前記の正常な組織とは反応しない上澄液も見出され
た。引用文献には活性化したヒト血小板に特異的なMA
bを示唆する証拠はないので、この結果は極めて驚くべ
きことであった。このMAbを分泌するハイブリドーマ
細胞を限定希釈法によって3回クローニングを行い、液
体窒素で凍結した(ハイブリドーマ細胞2128)。特
異性の詳細な免疫組織化学的分析を表3(第II表)に
示す。APAAP法(Cordell ら、上記)を用いて示さ
れた血小板に対するその高い特異性を上回って、MAb
BW2128は活性化されていない血小板とよりもトロ
ンビンで活性化した血小板と10倍以上強く反応する。
血小板のホルムアルデヒド固定は、MAbの血小板への
結合をごくわずか抑制するだけである。MAbを活性化
されていないヒト血小板に加えると有意な活性化を生じ
るが、トロンビン0.5単位/mlを加えると活性化マー
カー(PF4)が顕著に分泌されるようになる(表4
(第III表)。顆粒球上のエピトープに特異的なMA
bs(例えば、BW250)、またはマイコプラズマ上
のエピトープに特異的なもの(例えば、BW227)お
よびGPIa/IIaに特異的なもの(例えばBW4)
は、MAbBW2128と同様に、休止している血小板
の活性化に何んらの効果を示さない(K. Bossletら、(1
988), 14, 523-528 )。第III表に記載した条件下で
血小板をインキュベーションした後のPF4の測定のた
めのELISA法は、Pelzer, H., Heimburger, N. (19
86), J. Biomed. Mat. Res., 20, 1401-1409およびOsei
-Bonsu, A., Cafourek, G., Reiter, S., Popovic, R.,

るが、これらのMAbsは前記の生物学的特性に影響を
与える。免疫沈澱とウェスターンブロットにより、MA
bBW2128によって認識される血小板抗原はgp1

可能であった。

es., 49, 4852-4858, 1989にしたがって行った)を用い
て、これらのMAbが精製したTSPに強力に結合する
が、標準的な固相ELISA法並びに非還元条件下での
ウェスターンブロット分析で明らかにされたようにMA
bBW2128によって認識される血小板抗原には結合
しないことを示すことができた。これらの実験は、MA
bBW2128によって画定される血小板抗原は、前記
の抗TSPMAbによって検出された3種類のエピトー
プをTSPと共有しないことを示している。更に、2D
ゲル電気泳動(O'Farrell, P.H. (1975), J. Biol. Che
m., 250, 4007-4021)によって、還元条件下では分子量
がTSPより小さく且つ相対分子質量が約150,00
0〜170,000、好ましくは約160,000であ
るが、TSPと同様な等電点を有しており、実験誤差の
限界内では識別がつかず、好ましくは約5.5〜6.5
の範囲にある(比較実験でのTSPの分子量は180k
Daであり、等電点は5.5〜6.5であった)ことを
示すことも可能であった。この新規な抗原はTSPと結
合しており、物理的および化学的結合の両方が可能であ
り、活性化した血小板上で優先的に発現されるので、一
般的に血小板抗原と呼ばれる。この抗原は、好ましくは
第II表に詳細に示されている組織分布も示す。

体を調製し、チェックし、模倣体を調製するのに特に好
適である。

tPA変異体のような線維素溶解タンパク質をコードす
るヌクレオチド配列に結合させることができる。好まし
いtPA変異体は、例えばEP−A1−0387380
号明細書に記載の方法で調製することができる。これら
の構造は、ドイツ国特許出願第P4106389.9に
ヒューマナイズドαCEAMAbおよびヒトβ−グルク
ロニダーゼの例によって示されているように、DNAの
水準で連結して、機能性融合タンパク質として発現する
ことができる。一般的には、前記の融合タンパク質は、
血管閉塞における活性化した血小板の特異的な高親和性
結合領域と同じ分子における機能的に活性な領域とを合
せ持つので、副作用の少ない効率的な線維素溶解剤であ
る(Haber, E., Quertermous, Th., Matsueda, G.R., R
unge, M.S., Bode, Ch. (1989),Jap. Circulation J.,
54, 345-353)。

しているプラスミドクローンBを、制限マッピングおよ
び核酸配列分析によって同定した(図3)。

IIIおよびEcoRIで開裂したのと同様にKS+プ
ラスミドベクター(pBluescriptIIKS
+;ストラタジーン(Stratagene)、ラ・ジョラ、カリフ
ォルニア州)に連結した。IgG3遺伝子が5′および
3′末端においてBamHI開裂部位を有するプラスミ
ドクローンCを単離した(図4)。

連結した。式に示される配向のIgG3C遺伝子を含む
発現プラスミドDを同定した。このクローニング段階で
は、pABStopベクターはSV40停止および2つ
のBamHI開裂部位の間に位置したポリアデニル化信
号を喪失する(図5)。

作によって連結した抗体抱合体を図式的に表現したもの
である。図1bは抗体抱合体を模式的に表現したもので
あり、二特異性抗体の各種の抗原結合特異性が、V領域
を異なった陰影を付けることによって明らかになってい
る。オリゴ特異性高分子の場合には、それぞれの場合
に、CH1ドメインはポリペプチドリンカーを介して次
の、軽鎖が結合しているVHドメインに連結している。
抗体/酵素抱合体の酵素は点で表わしている。

Claims (29)
- 【請求項1】寄託番号がDSM ACC2024であ
る、ハイブリドーマ2128。 - 【請求項2】ハイブリドーマ2128(寄託番号DSM
ACC2024)に由来する単クローン性抗体。 - 【請求項3】請求項2に記載の単クローン性抗体によっ
て認識されるエピトープに結合する単クローン性抗体ま
たはその部分。 - 【請求項4】活性化したヒト血小板に優先的に結合し、
且つ請求項2に記載の単クローン性抗体によって認識さ
れるエピトープに結合する生物学的特性を有する、表1
および/または表2に示されるアミノ酸配列またはそれ
らの任意の対立形質変種または変異体を含む、単クロー
ン性抗体またはその部分。 - 【請求項5】抗体がキメラ、ヒューマナイズド、二−ま
たはオリゴ特異性抗体である、請求項3または4に記載
の単クローン性抗体またはその部分。 - 【請求項6】抗体がmruフラグメント、単一ドメイン
フラグメント、一本鎖フラグメント、F(ab)フラグ
メントまたは1または2以上のヒンジ領域を有するF
(ab′)2フラグメントである、請求項3〜5のいず
れか1項に記載の単クローン性抗体。 - 【請求項7】キメラ抗体がBW Chi 2128であ
る、請求項5に記載の単クローン性抗体。 - 【請求項8】好ましくはTc−99mで標識されてい
る、請求項1〜7のいずれか1項に記載の単クローン性
抗体。 - 【請求項9】請求項2〜4のいずれか1項に記載の単ク
ローン性抗体によって認識されるエピトープに結合する
ことを特徴とする、模倣体。 - 【請求項10】請求項2〜7のいずれか1項に記載の単
クローン性抗体またはその部分または請求項9に記載の
模倣体、および免疫グロブリン類に属さないタンパク質
またはその機能性部分から構成される融合タンパク質。 - 【請求項11】免疫グロブリン類に属さないタンパク質
が線維素溶解タンパク質またはその機能性部分である、
請求項10に記載の融合タンパク質。 - 【請求項12】線維素溶解タンパク質がストレプトキナ
ーゼ、ウロキナーゼ、tPA、tPA変異体またはその
機能性部分である、請求項11に記載の融合タンパク質。 - 【請求項13】請求項2に記載の単クローン性抗体によ
って認識されるエピトープに結合する生物学的特性を有
する表1および/または表2に示されるアミノ酸配列ま
たはそれらの任意の対立形質変種または変異体を含むポ
リペプチド。 - 【請求項14】請求項2に記載の単クローン性抗体によ
って認識されるエピトープに結合する生物学的特性を有
するポリペプチドをコードする1または2以上の核酸配
列を含むポリヌクレオチド。 - 【請求項15】請求項14に記載のポリヌクレオチドを
含むベクター。 - 【請求項16】請求項14に記載のポリヌクレオチドを
含む発現ベクター。 - 【請求項17】ベクターL1またはベクターL2であ
る、請求項16に記載の発現ベクター。 - 【請求項18】請求項14に記載の組換えポリヌクレオ
チドまたは請求項15〜17のいずれか1項に記載のベ
クターを含む原核または真核宿主細胞。 - 【請求項19】宿主細胞がCOS、CHOまたはBH
K、好ましくはBHK細胞である、請求項18に記載の
真核宿主細胞。 - 【請求項20】請求項14に記載のポリヌクレオチドに
よる原核または真核宿主細胞の形質転換またはトランス
フェクションにより宿主細胞が前記のポリペプチドを発
現するようにする、請求項13に記載のポリペプチドの
製造法。 - 【請求項21】請求項18に記載の原核または真核宿主
細胞、好ましくは真核宿主細胞を培養し、前記のポリペ
プチドを単離することを特徴とする、請求項13に記載
のポリペプチドの製造法。 - 【請求項22】請求項2〜8のいずれか1項に記載の単
クローン性抗体、請求項9に記載の模倣体、請求項10
〜12のいずれか1項に記載の融合タンパク質または請
求項13に記載のポリペプチドを含む、医薬組成物。 - 【請求項23】請求項2〜8のいずれか1項に記載の単
クローン性抗体、請求項9に記載の模倣体、請求項10
〜12のいずれか1項に記載の融合タンパク質または請
求項13に記載のポリペプチドを含む、診断用助剤。 - 【請求項24】請求項2〜8のいずれか1項に記載の単
クローン性抗体、請求項9に記載の模倣体、請求項10
〜12のいずれか1項に記載の融合タンパク質または請
求項13に記載のポリペプチドの、活性化したヒト血小
板を検出するための診断用助剤の製造への使用。 - 【請求項25】請求項2〜4のいずれか1項に記載の抗
体によって認識される抗原またはその免疫学的に反応性
を有する部分。 - 【請求項26】還元条件下の相対分子質量が約150,
000〜170,000である、請求項25に記載の抗
原。 - 【請求項27】実験誤差の限界内でTSPとは識別がつ
かない等電点を有する、請求項25または26に記載の
