Description
単独で用いるか、または卵胞刺激ホルモンと組み合して
用いる男性および女性の不妊症療法並びにそれらの医薬
製剤に関する。
胞刺激ホルモン(FSH)の効能は周知である。かかる
細胞によるプロゲステロン合成能の獲得は、順次、正し
い卵胞の成長および黄体の正常な機能を果すための卵母
細胞の成熟の完成を決定する。

クリックAMP産生に対して阻害作用を有し、従って卵
巣顆粒膜細胞の分化に対して阻害作用を有することが知
られている。他の成長因子、TGF−β(形質転換成長因
子−β)は、同じ細胞の分化過程の期間を通じてFSH
およびEGFの作用を調節する(Pei Feng,他−顆粒膜
細胞分化期間中の形質転換成長因子−βによる上皮成長
因子の阻害活性の調節−J.Boil.Chem.261(30),14167,19
86)。
用を有し、そしてこのゴナドトロピンの細胞標的レベル
においてFSHと相互に影響しあうことが明らかであ
る。それにもかかわらずいまだそれらの機能および相対
的な相乗作用の機構は、説明されない。

ラゲナーゼによる微小管(tubules)の処理によりラィジ
ッヒ細胞に由来する微小管(tubular)構成物の分離から
成る。分離細胞は、ウエル当たり約100,000細胞の濃度
で48ウェルコースタープレートに供した。95%のO
2/5%のCO2の雰囲気下、34℃のメタボリック・イン
キュベーター中に24時間放置し、細胞をウエルの底に
付着させた。細胞を洗浄した後、10-11Mの濃度のヨウ
素標識GRF-44の存在下、10-6Mのコールド(標識化して
いない)ヒトGRF-44の存在下又は非存在下で、TC199の
10mMのTRIS培地中で、4℃で60分間インキュベーシ
ョンした。その後、細胞を、PBS+0.5%のBSAを
用い、4℃にて3度洗浄し、次いで放射線活性を有する
細胞を0.1NのNaOHに可溶化し、そしてガイガーカウン
ターで読み取った。

を組み合せて投与するものについて実施した。

周期の13日目と14日目との間に卵胞の活性化の存在
および優性な卵胞の形跡を示す。
日、i.v.の条件下での観察: ケース3(C.A.) 34才−患者。

びにクエン酸クロミフェン+エチニルエストラジオール
および/またはゴナドトロピンによる誘導療法は、優性
なものへの進展の可能性を伴わないが黄体形成の傾向を
伴う多数の卵胞活性化の存在を示す。
コーグラフィックな徴候の不存在、並びに排卵活性化の
生化学的な徴候の存在を示した。
験の第二周期は、卵胞の活性化、優性な卵胞および卵胞
の裂開の存在を示す。卵胞に対して影響を与える大きさ
を考慮すると周期の16日目以降のエコーグラフィック
観察の消失が退縮に帰因し得るとすることを排除できな
いであろう。生化学的データは、この周期の観察におい
て排卵の存在を明らかにする。
〔メトロジン(Metrodin )〕150I.U.量を投与する条
件下で実施した。このケースでは、優性な卵胞を伴わな
い卵巣の活性化の徴候が記録された。
(150mcg/日、i.v.)+メトロジン(Metrodin )(150
I.U./日、i.m.)の投与下で実施した。この周期の期間
中に、卵巣の活性化、優先な卵胞および周期の18日目
に裂開を観察することができた。
妊症を病む患者。従前の試験は、卵胞の成熟の存在を示
している。過去には、クエン酸クロミフェンまたはゴナ
ドトロピンを用いる刺激療法を実施することにより、卵
胞の活性化および優性な卵胞の存在を示したが、しかし
ながら排卵を伴わなかった。
5日目まで卵胞の活性化および優性な卵胞の不存在を示
す。
期の試験は、排卵の不存在を示したが、卵胞形成能は活
性化された。
I.U./日、i.m.のアンプル中のFSH〔メトロジン(Me
trodin )〕の投与下で実施した。
な卵胞の形成能および排卵は示さなかった。
れi.v.およびi.m.の経路でGRF-29の100mcg/日+メトロ
ジン(Metrodin )の75I.U./日を投与する条件下で
実施した。観察期間を通じて、周期の18日目および1
9日目の間に排卵を伴う、卵胞の成熟化への進展を観察
することができた。

めた。クエン酸クロミフェンおよびゴナドトロピンを用
いる刺激療法の周期をしたが、臨床状況の解決は伴わな
かった。

黄体化の存在を示す。

ニターリングの中止を決定した。35日目に月経が再度
始った。月経期間中に行ったエコーグラフィック試験
は、卵胞形成の消失を示した。

与下で実施した。より早熟な卵巣の活性化を除いて、対
照の周期の期間を通じて観察された状況と実質的に同様
であることが示された。

た。周期の26日目に月経が自然に始った。月経期間中
に行ったエコーグラフィック検査は、この周期中に卵胞
形成の消失を示した。

期間の間に、周期の16日目と17日目との間に排卵を
伴う、卵胞のより優れた成熟の進展を観察することがで

左側の卵巣剔除術を施された観察下の患者。何等の療法
も施されていない第一周期の観察の期間を通じて、エコ
ーグラフィックな観察は可能であったが、生化学的な観
察はできず、そして卵巣の活性化の徴候および優性な卵
胞の不存在を観察することができた。
の150mcg/日、i.v.投与の下に実施した。

嚢胞性卵巣内)の増殖が観察された。

周期の14日目あたりで裂開を伴う、卵巣の活性化およ
び優性な卵胞が観察された。
も卵巣の活性化が見られない。この患者は、二次的無月
経であり、プロゲステロンの投与後だけ月経を生じる。
試験の開始に先立ち、ゲストン(Gestone )100mg、i.
m.により月経を誘発した。第一周期の観察は、何等の療
法も施さないで実施し、卵巣の活性化に対するエコーグ
ラフィックおよび生化学的な徴候が存在しないことが確
認された。
の150mcg/日、i.v.の投与で実施した。卵巣の活性化の
エコーグラフィックな徴候の存在が確認されたが、優性
な卵胞は存在しないことが確認された。
〔メトロジン(Metrodin )〕の150I.U./日、i.m.の
投与の下に実施した。卵巣の活性化のエコーグラフィッ
クな観察は可能であったが、卵巣の活性化の生化学的な
徴候は観察できず、さらに優性な卵胞も存在しないこと
が観察された。
の150mcg/日、i.v.+メトロジン(Metrodin )の150
I.U./日、i.m.の投与の下に実施した。卵胞の活性化、
優性な卵胞および卵胞の裂開のエコーグラフィックおよ
び生化学的な徴候の存在を示すことができた。
卵胞の成長を促進し、そして排卵(裂開)を期待できる
か、または排卵の誘発さえも示す。さらに、GRFは、
ほぼ卵胞のレベルで無排卵が実証されている患者におけ
るFSH作用を相互に補助しあう(coadjuvating)ことが
できる。これらの後者では、FSHとの組み合せたGR
Fによる処置は、常に卵胞を排卵まで成長せしめる効能
をもたらす。
全症を病む患者を、選択されるプロトコールの判定基準
に従い、1週3度の5mcg/kg、s.c.量のGRF-29で3ヶ月
間処置した。
者は、正常または平均以上のゴナドトロピンレベルを有
する25〜40才の範囲内にあり、そして精液パラメーター
を妨害し得る炎症反応または他の疾病を有していない。

Claims (6)
- 【請求項1】ソマトトロピン放出因子の薬学上の有効量
および1以上の賦形剤を含有する不妊症療法のための医
薬製剤。 - 【請求項2】ソマトトロピン放出因子が、GRF-29である
請求項1記載の医薬製剤。 - 【請求項3】医薬として許容され得る1以上の賦形剤の
存在下で、ソマトトロピン放出因子および卵胞刺激ホル
モンを含有し、不妊症療法においてその活性成分を同時
にか、別々にかまたは連続的に使用するための医薬製
剤。 - 【請求項4】医薬として許容され得る1以上の賦形剤の
存在下で、GRF-29および卵胞刺激ホルモンを含有する請
求項3記載の医薬製剤。 - 【請求項5】ソマトトロピン放出因子およびメノトロピ
ン(Menotrophin)を含有する請求項3記載の医薬製剤。 - 【請求項6】ソマトトロピン放出因子がGRF-29である請
求項5記載の医薬製剤。
