Description
(発明の利用分野)
本発明は、5−および7−位で塩基性基により
置換された新規なトリアゾロピリミジンおよびそ
の製造方法に関する。この新規化合物は医薬とし
て使用することができる。
(発明の開示)
東独特許第61269号明細書で記載された塩基性
置換化合物が冠作用(Coronarwirkungen)を有
するという知見に基づいて、従来知られていなか
つたこの系統の化合物が心臓−循環作用を示し、
この作用が公知の化合物の作用より数段優れかつ
同時に毒性がないこと、さらに医薬として使用で
きることが以外にも見い出された。
この新規な化合物は下記一般式()に対応す
る。
ここで、置換基は次の意味を有する。
R2およびR3は水素原子、鎖長C1〜C3のアルキ
ル基またはハロゲン原子である。
R4は直鎖もしくは分枝鎖の鎖長C4〜C9のアル
キル基、鎖長C1〜C3のアルコキシ基もしくはア
ルキル基で置換されたアラルキル基、2,5−ジ
オキサヘプチル残基または3−オキサヘキシル残
基である。
R5は水素原子、鎖長C1〜C3のアルキル基、ヒ
ドロキシエチル基またはヒドロキシプロピル基で
ある。
R6およびR7は水素原子、または直鎖もしくは
分枝鎖の鎖長C1〜C5のアルキル基であるか、も
しくはR6とR7とがヘテロ原子と共に環を形成す
るものである。
5−位および7−位の置換基を相互に交換して
もよい。
式()から置換基を決めることにより可能な
多数の化合物のうち、次の化合物が、心臓−循環
作用について著しく有効であることがわかつた。

式()に対応する化合物は、好ましくは生理
的に親和性のあるその塩の形で用いられる。特
に、塩化水素酸、臭化水素酸、ヨウ化水素酸、硫
酸、硝酸、シユウ酸、マロン酸および酒石酸が塩
生成に適している。
東独特許第61269号明細書記載の化合物に類似
する本発明による化合物は、下記一般式()の
トリアゾロピリミジンと、
(式中、R2およびR3は前記と同じ意味を有し、
Halは塩素原子または臭素原子を示す)
下記一般式()のアミンと
(式中、R4、R5、R6およびR7は前記の意味を示
す)
の反応によりつくられる。この反応は水、水−ア
ルコール混合物、アルコール類、トルエンまたは
ベンジンなどの溶剤中で行われる。この反応はそ
の時の置換アミンと共に二段階を経由する。
まず第一に、293K以下の温度で7−位のハロ
ゲンがアミンにより置換され、次いで、この時に
用いられた溶剤の沸点までの温度で5−位のハロ
ゲンが置換される。
この反応の際生じたハロゲン化水素酸を取り除
くために、過剰量の一般式()のアミンまたは
トリエチルアミン、炭酸アルカリ、もしくは水酸
化アルカリが加えられる。粗生成物の回収は通常
の方法で処理される。目的生成物が抽出、蒸留ま
たは再結晶によつて副生物から分離されて精製さ
れる。活性炭、酸化アルミニウムまたは酸性白土
などの吸着剤の添加によつて無用の着色を取り除
く。
得られた化合物を酸でその塩にしてもよい。立
体異性体の化合物が含まれているならば、これを
その成分に公知の方法で分離することができる。
本発明による化合物、特に前記の各々の化合物
は、動物実験において良好な有効性および治療上
の重要な特性を示す。試験官内および生体内の血
小板凝集抑制はアセチルサリチル酸もしくはトラ
ピデール(Trapidil)などの公知の薬物より数段
すぐれている。分離した心臓試料および心房試料
は冠状拡張および筋収縮力の増大作用を示す。特
に高血圧症状におけるその血圧を下げる。本発明
の化合物は高浸透状態で誘発される赤血球の変形
死並びに凝集傾向を抑制し、それにより毛細循環
領域における流動状態を改善する。アラキドン酸
代謝の影響については、トロンボキサンA2の生
成を緩和し、または凝集阻害性プロスタグランジ
ン、特にフロスタサイクリンの合成を促進する。
この化合物は、また抗不整派作用を示す。この新
規な化合物の本質的な特徴は、カルシウム物質代
謝、特に生体細胞中の圧力依存透過膜性Ca2+流
(Ca2+−Einstroms)の影響にある。これは、フ
リケンスタイン(A.Fleckenstein著「心筋および
平滑筋中のカルシウム拮抗」、ジエー・ウイリ
ー・アンド・サンズ社(ニヨー・ヨーク)1983)
によれば、この際に高い選択性を持つカルシウム
拮抗を示す。既知のCa−拮抗に対する特別の長
所は、高い選択性および作用強度、すなわち、軽
い可変的効果、ならびに、敏感な細胞の高頻度の
刺激に際しての作用の発展にある。その広汎な薬
理的作用スペクトルおよび低い急性毒性から、局
処貧血性の心臓−循環疾患の治療法として本発明
の化合物を含有せしめることができ、従来から知
られている化合物よりも優れている。5%の水性
溶液の注射溶液として、または58%の作用物質、
20%のラクトース、19%の水分、3%のステアリ
ン酸マグネシウムの組成の公知法で被覆された粒
剤として使用できる。
例 1
18.9gの5,7−ジクロロ−s−トリアゾロ
〔1,5−a〕ピリミジンを50mlのエタノール中
に溶解もしくは懸濁し、25mlのエタノール中の
26.1gのn−アミルエタノールアミンの溶液によ
つてゆつくりと添加する。その際、撹拌し、添加
終了後なお1時間、283〜288Kに維持する。次い
で、得られた懸濁液に293〜298Kのエタノール20
ml中のピペリジン17.0gの溶液を添加し、313〜
324Kで3時間維持して沈殿を防ぐ。その後、100
mlの塩化メチレンを添加し、75mlの水で3度洗浄
してその水を取り除く。塩化メチレンを濃縮し、
残渣を結晶化する。ベンゼンによる再結晶化か
ら、融点が390〜391Kである26.5gのピペリジノ
−7−(N−(n−ペンチル)−N−(β−ヒドロキ
シエチル)−アミノ)−s−トリアゾロ〔1,5−
a〕ピリミジン(理論値の80%)が得られる。
例 2
18.9gの5,7−ジクロロ−s−トリアゾロ
〔1,5−a〕ピリミジンを50mlのエタノール中
に溶解もしくは懸濁し、25mlのエタノール中の
26.1gのn−アミルエタノールアミンの溶液によ
つてゆつくりと添加する。その混合物を撹拌し、
添加終了後なお1時間、283〜288Kに放置する。
得られた懸濁液を取り出し、50mlのエタノール中
で懸濁して293〜298Kで14.6gのジエチルアミン
の溶液を添加する。
次いで、3時間還流下その混合物を維持し、引
き続き真空下濃縮する。残渣を100mlの塩化メチ
レンに添加し、50mlの水で塩化メチレン層を洗つ
て分離する。この塩化メチレンを濃縮し、残渣を
結晶化してエーテルから再結晶化する。得られた
融点352〜353Kの5−ジエチルアミノ−7−(N
−(n−ペンチル)−N(β−ヒドロキシエチル)−
アミノ)−s−トリアゾロ〔1,5−a〕ピリミ
ジンは225gの収量(理論値の70%)となる。
例 3
18.9gの5,7−ジクロロ−s−トリアゾロ
〔1,5−a〕ピリミジンを50mlのエタノール中
に懸濁する。5〜10℃で撹拌下に29.4gのn−ヘ
キシルアミノエタノールを滴下し、沈降物の温度
を293Kにしてその温度で1時間撹拌する。次い
で、得られた化合物を取り出し、再度150mlのエ
タノール中で懸濁する。室温で15gのジエチルア
ミンを添加し、次いで混合物を3時間沸騰させ
る。沈降物を真空下濃縮し、その残渣に75mlのク
ロロホルムを添加して水で洗浄する。洗浄後有機
相を濃縮し、得られた5−ジエチルアミノ−7−
(n−ヘキシル)−N−(β−ヒドロキシエチル)−
アミノ)−s−トリアゾロ〔1,5−a〕ピリミ
ジンを石油エーテルから再結晶する。融点は335
〜336Kを示し、収量は162g(論理値の50%)と
なる。
例 4
例3に記載されたようにこの反応を起させる。
ジエチルアミンの代りに17.1gのピペリジンを用
いる。得られた5−ピペリジノ−7−(N−(n−
ヘキシル)−N−(β−ヒドロキシエチル)−アミ
ノ)−s−トリアゾロ〔1,5−a〕ピリミジン
をベンゼンから再結晶化する。収量は28gを、融
点は354K示す。
例 5
18.9の5,7−ジクロロ−s−トリアゾロ
〔1,5−a〕ピリミジンを、50mlのエタノール
と50mlの水との混合液中で懸濁する。278〜283K
の温度で23.6gのn−ブチルアミノエタノールを
ゆつくりと添加し、室温で3時間沈降物を撹拌す
る。得られた化合物を次いで取り出し、再び150
mlのメタノール中で懸濁し、17.4gのモルホリン
をゆつくり添加して還流下2時間沈降物を保持す
る。メタノールを濃縮し、その残渣を75mlの塩化
メチレンに添加して50mlの水で2度洗浄する。濃
縮後、5−モルホリノ−7−((n−ブチル)−N
−(β−ヒドロキシエチル)−アミノ)−s−トリ
アゾロ〔1,5−a〕ピリミジンは375Kの融点
を示し、16gの収量(理論値の50%)である。
例 6
200mlのメタノール中に18.9gの5,7−ジク
ロロ−s−トリアゾロ〔1,5−a〕ピリミジン
を懸濁させる。278〜283Kで26.6gのN−(エト
キシエチル)−N−(β−ヒドロキシエチル)−ア
ミンを添加して288〜293Kで2時間なお沈降物を
撹拌する。引き続き、反応生成物を取り出し、再
び75mlのメタノール中で懸濁させ、17gのピペリ
ジンを添加して沸騰温度で3時間保持する。この
懸濁液を濃縮し、その残渣を晶出させる。5−ピ
ペリジノ−7−(N−エトロキシエチル)−N−
(β−ヒドロキシエチル)−アミノ)−s−トリア
ゾロ〔1,5−a〕ピリミジンは酢酸エチルエス
テルから再結晶されて、393〜394Kの融点を示
し、その収量は22g(理論値の65%)である。
例 7
例6で示したように実験を行い、17gのピペリ
ジンの代りに14.6gのジエチルアミンを用いた。
ベンゼンの再結晶化から融点342〜343Kの5−ジ
エチルアミノ−7−(N−(エトキシエチル)−N
−(β−ヒドロキシエチル)−アミノ)−s−トリ
アゾロ〔1,5−a〕ピリミジンを得る。
例8〜15では、化学名称の代りに次の記号A〜
Cが当てられる。

ヒト血小板に関するアラキドン酸ないしU−
46619誘発の凝集
過去10日間にわたつて薬剤を服用しなかつた給
血者から血液9体積を、3.8%クエン酸三ナトリ
ウム溶液1体積と混合し、そして室温で、10分
間、200gの遠心分離した。血小板に富む血漿
(PRP)をシリコン製ピベツトで取り出し、そし
て最長3時間の試験期間中室温で保持した。
テスト化合物をエタノール/クロロホルム
(1:1)の溶剤混合物中に溶かし、計量器に注
入して、有機溶剤混合物を窒素流で吹き払つた。
0.3mlのPRPと生理食塩溶液の添加および2分間
の前潜伏期後、37℃で、アラキドン酸ナトリウム
(0.2〜0.8ミリモル/1)ないしTXA2拮抗筋U−
46619(0.2〜0.8ミリモル/1)による凝集が発現
した。凝集経過はボルンの方法(Born、9.V.R.
およびCross、U.J.:J.Physiol.168、178(1963))
によつて測定した。凝集を50%抑制するために必
要なテスト化合物の必須濃度がそれらとトラピジ
ルとの直接比較によつて調べられた。表1の結果
が示すように、これらの誘導体がトラピジルと比
較してかなり強い抗凝集作用を有している。
FIELD OF THE INVENTION The present invention relates to novel triazolopyrimidines substituted with basic groups at the 5- and 7-positions and a method for producing the same. This new compound can be used as a medicine.
