Description
本発明は、生体中から抽出される糖タンパクの
一種であるフイブロネクチンを配合した化粧料に
関するものである。さらに詳しくは皮膚に対して
皮膚賦活、保湿効果及び美白作用を有し、頭髪に
対しては養毛効果及び栄養作用を有するフイブロ
ネクチンを配合してなる化粧料に関する。
従来皮膚の代謝促進剤及び養毛剤としては、一
般にホルモン類が使用されてきた。すなわち卵胞
ホルモンもしくは副腎皮膚ホルモン等が肌を若返
らせ、しわを防ぐ目的で、又頭髪の育毛を促がす
目的で配合されてきた。
しかしながらホルモン類は多量に使用すると全
身的な副作用が発現したり表皮の肥厚化が起つた
りするなどの問題点があつた。
本発明者らは、こうした上記の事情にかんが
み、安全でしかも薬効性の高い物質の探索及び化
粧品への配合について鋭意研究を重ねた結果、生
体抽出物であるフイブロネクチンが上記の欠点も
なく、優れた効果を有することを見い出し、本発
明を完成するに至つた。
すなわち本発明はフイブロネクチンを配合する
ことを特徴とする化粧料を提供するものである。
以下、本発明の構成について詳述する。本発明
で用いられるフイブロネクチン(fibronectin、
線維芽細胞膜蛋白)は、生体中から抽出される糖
タンパクの一種で、別名、寒冷不溶性グロブリン
(cold insoluble globulin、CIG)とも呼ばれる。
これまでは大外面トリプシン感受性蛋白、細胞表
面蛋白、粘着固子あるいはオブソニツクα2表面結
合糖蛋白などとして呼ばれてきたが、最近では概
ねフイブロネクチン、あるいは寒冷不溶性グロブ
リンと呼ばれている。フイブロネクチンは、血奨
の他、線維芽細胞などの間葉細胞や表皮などの基
底膜に存在する。血奨中に存在するものは分子量
20万〜22万の単量体が二つついた二量体の構造を
なしており、血奨中に0.3mg/ml程度含まれてい
る。間葉細胞や基底膜に存在するものは20万〜25
万の分子量を持つ単量体が二量体もしくはそえ以
上の多量体をなした構造をしている。前者は通
称、血奨フイブロネクチン、後者は通称細胞性フ
イブロネクチンと呼ばれている。フイブロネクチ
ンについて他に知られている物理化学的性状とし
ては、易動度がα2グロブリンであり、等電点は
5.0、分子吸光係数A1%
1cm280nmが12.9〜13.0、
S0 20、Wが11〜14S、糖含量が5%、などをあげる
ことができる。
なお、フイブロネクチンはいくつかのドメイン
から成り立つており、それぞれがたとえば、トラ
ンスグルタミナーゼ、ヘパリン、アクチン、フイ
ブリン、細胞、コラーゲンなどと作用する特異構
造を有している。
上記の通りフイブロネクチンの人または動物の
細胞または血奨中に存在する。抽出は常法に従つ
て行なえばよい。たとえば0.2Mの尿素で培養細
胞を処理すれば5〜10%のフイブロネクチン溶液
が得られる。またゼラチン変性アフイニテイクロ
マトで処理すれば血奨中のフイブロネクチンが抽
出される。
本発明におけるフイブロネクチンの配合量は一
般的に該化粧料全量中の0.00001〜0.1重量%が適
当である。
本発明の化粧料は上記の必須成分に加えて必要
に応じて、本発明の効果を損わない範囲内で油
分、界面活性剤、顔料、香料、防腐剤、保湿剤、
紫外線吸収剤、色素、酸化防止剤、他の薬剤等を
配合できる。
本発明で得られた化粧料は頭皮に塗布した場
合、優れた育毛・発毛作用を発揮し、た皮膚に塗
布した場合は保湿性を与え、美白及び皮膚賦活作
用を発揮する。
次に実施例によつて本発明をさらに詳細に説明
する。なお本発明はこれにより限定されるもので
はない。配合量は重量%である。

製造法:精製水にクエン酸、クエン酸ソーダ、
グリセリン、フイブロネクチン、1−3ブチレン
グリコールを溶解する。別にエタノールにポリオ
キシエチレンオレイルアルコール、香料、メチル
パラベンを溶解し、これを前述の精製水溶液に加
えて可溶化しろ過して化粧水を得た。
〔美白効果〕
フイブロネクチンの美白効果をみるために実施
例1で得た化粧水のメラニン生成抑制効果を調べ
た。
まず実施例1で製造した化粧水を試料として用
い、またフイブロネクチンを配合しないもの(精
製水で置換)をブランクとして調製し、それぞれ
にL−チロジン水溶液及びマツクルベイン氏の緩
衝液を有効成分とするリニメント剤を添加し、10
分間恒温槽に入れる。次にチロジナーゼ水溶液を
加えてかくはんし、分光光度計をセツトしたのち
475mμにおける吸光度を経時的に測定した。475
mμの吸光度が上昇することはL−チロジンが酸
化され、メラニンの前駆体であるドーパークロム
が生成していることを示す。すなわち、ドーパー
クロムの生成量が少ないことは、メラニンの生成
抑制効果があるということである。測定結果を図
1に示す。この結果よりフイブロネクチンはメラ
ニン抑制効果を有する事がわかる。
〔創傷治癒効果〕
生後6週齢のウイスター系ラツトを5匹1群と
し剃毛した後試験に供した。ラツトは麻酔後背部

The present invention relates to cosmetics containing fibronectin, which is a type of glycoprotein extracted from living organisms. In view of the above-mentioned circumstances, the present inventors have conducted intensive research to search for safe and highly medicinal substances and to incorporate them into cosmetics. As a result, we have discovered that fibronectin, a biological extract, has excellent properties without the above-mentioned drawbacks.
