は哺乳類の身体外での医療を目的とした医薬組成物およ
び医薬製剤(本出願では医薬組成物と呼んでいる)にも
関する。このような組成物および製剤は主に身体外での
循環または処理(たとえば対外循環や腎臓機械による透
析)、保存または修正(たえとば血液分離)にかけられ
る血液における凝血防止添加物として使用される。
薬組成物およびその類似体に加え、′本発明はヒトまた
は哺乳類の身体外での医療を目的とした医薬組成物およ
び医薬製剤(本出願では医薬組成物と呼んでいる)にも
関する。このような組成物および製剤は主に身体外での
循環または処理(たとえば対外循環や腎臓機械による透
析)、保存または修正(たえとば血液分離)にかけられ
る血液における凝血防止添加物として使用される。
これらの組梠物において、保存用溶液などの製剤または
単一投与形の製剤は上述の注射用製剤と同じである。し
かし活性物質の量または濃度は処理する血液の容積に基
づいている。

(材料と方法)
VACはEPA 0181465 もしくはEPA
0293567に類似する方法で調製した。以下の実験
はVACαを用いて行ったがその結果は別のアネキシン
類、特にVACβにも転換し得る。

%DOPCからなるリン脂質膜に結合する。

高濃度の亜鉛イオン(1mM)は上限はあるがVAC吸
着を促進する(第3図)。50μMでは吸着に全く効果
がない、驚くべきことにこの濃度は(’ a 3 *存
在下では結合に影響する。すなわち相乗効果が観察され
る。わずか1%DOPSを含む二重層に対する((:
a 2 ゛) 、 、2値は8.6から2.7mMに減
少する( (Zn”〕= 50 μM) (第4図)

Claims (1)
- 【特許請求の範囲】 1、アネキシンおよび天然のまたは化学合成的もしくは
遺伝子工学的に製造されたその誘導体または類似体から
なる群から選ばれた抗凝血物質およびCa^2^+、C
d^2^+、Zn^2^+、Mn^2^+およびCo^
2^+からなる群から選ばれた少なくとも1つの二価カ
チオンを含み、また場合によっては賦形剤および、また
はキャリヤーおよび、または安定剤を含むことを特徴と
する医薬組成物。 2、前記血管抗凝血物質が、式 【遺伝子配列があります。】 【遺伝子配列があります。】 【遺伝子配列があります。】 (式中、XXは、GluまたはAspを示し、また場合
によっては部位1のメチオニンが切断されており、また
場合によっては部位2のアラニンがブロックされており
、および、または場合によってはたとえば部位316の

て引き起こされる凝固が存在する。) で表される物質であるか、またはその生物学的に活性な

1乃至3および5のいずれか1項記載の医薬組成物。 7、0.1から100μM、好ましくは18から30μ
MのZn^2^+を含むことを特徴とする請求項1、2
、4、5または6のいずれか1項記載の医薬組成物。 8、Zn^2^+濃度が通常の血漿亜鉛濃度の範囲内に
あることを特徴とする請求項7記載の医薬組成物。 9、0.01から100mM、好ましくは0.03から
10mMのCa^2^+かつ0.1から100μM、好
ましくは1.8から30μMのZn^2^+、特に好ま
しくは通常の血漿亜鉛濃度の範囲の亜鉛を含むことを特
徴とする請求項5記載の医薬組成物。 10、アネキシンおよび天然のもしくは化学合成的また
は遺伝子工学的に製造したその誘導体または類似体から
なる群から選ばれる血管抗凝血物質を好ましくは請求項
6乃至8のいずれか1項で特定される濃度の、Ca^2
^+、Cd^2^+、Zn^2^+、Mn^2^+およ
びCo^2^+からなる群から選ばれる少なくとも1つ
の二価カチオンと混合し、また場合によっては賦形剤お
よび、またはキャリヤーおよび、または安定剤と混合し
、また場合によっては凍結乾燥することを特徴とする請
求項1記載の医薬組成物の製造法。 11、血栓症または塞栓症の治療および、または予防を
目的とした請求項1乃至9のいずれか1項記載の医薬組
成物の使用。 12、身体外部の医薬を目的とした請求項1乃至9のい
ずれか1項記載の医薬組成物の使用。
