発明の分野
本発明は、哺乳動物宿主の結合組織を再生する際の使用のために、トランスフォーミング増殖因子βスーパーファミリーのメンバーをコードする少なくとも1種の遺伝子を少なくとも1種の哺乳動物細胞に導入する方法に関する。本発明はまた、哺乳動物宿主の結合組織を再生する際の使用のために、トランスフォーミング増殖因子βスーパーファミリーの少なくとも1種の遺伝子産物と、少なくとも1種の結合組織細胞と、を導入する方法に関する。本発明はまた、トランスフォーミング増殖因子βスーパーファミリーのメンバーをコードする遺伝子を含有するDNAベクター分子を宿す哺乳動物細胞株に関する。
整形外科の分野において、変形性関節症または骨関節炎、および運動活動への参加により引き起こされた傷害が、最も頻繁に遭遇する軟骨損傷関連の病気である。骨関節炎に関しては、膝、腰、肩および手首などの体内のほとんど全ての関節が、罹患する。この疾患の病原は、硝子関節軟骨の変性である(Mankin et al., J Bone Joint Surg, 52A:460-466,1982)。関節の硝子軟骨は、変形し、フィブリル化して、最終的に陥凹する。変性した軟骨をどうにかして再生することができれば、ほとんどの患者が、消耗性の痛みを感じることなく生活を楽しむことができるであろう。今日までに、損傷された硝子軟骨を再生するために報告された方法はない。
本発明は、本明細書の先に記載された要件をかなえた。哺乳動物宿主を処置する際の使用のために、生成物をコードする少なくとも1種の遺伝子を哺乳動物細胞の少なくとも1種の細胞に導入する方法が、本発明で提供される。この方法は、該生成物をコードする遺伝子を含有するDNAベクター分子を生成するために組換え技術を利用すること、および該生成物をコードする遺伝子を含有する該DNAベクター分子を哺乳動物細胞に導入すること、を含む。該DNAベクター分子は、標的細胞または組織に送達され、そこで維持されることが可能な任意のDNA分子であり得るため、該当する生成物をコードする遺伝子が安定して発現され得る。好ましくは本発明内で使用される該DNAベクター分子は、ウイルスまたはプラスミドのどちらかのDNAベクター分子である。この方法は、好ましくは治療的使用のために生成物をコードする遺伝子を哺乳動物の結合組織の細胞内に導入することを含む。
a)タンパク質のトランスフォーミング増殖因子スーパーファミリーのメンバーを作製または獲得すること;
b)遺伝子をコードするベクターを含有し得る培養哺乳動物細胞の集団、または遺伝子をコードするいずれのベクターも含有しない培養結合組織細胞の集団を作製または獲得すること;および
c)関節内注射によりステップa)の細胞またはタンパク質と、ステップb)の結合組織細胞と、を哺乳動物宿主の関節炎の関節腔に移入することで、関節腔内での該組み合わせの活性が結合組織の再生をもたらすこと、
を含む、骨関節炎を処置する方法を対象とする。
a)タンパク質のトランスフォーミング増殖因子スーパーファミリーのメンバーを作製または獲得すること;
b)遺伝子をコードするベクターを含有し得る培養哺乳動物細胞の集団、または遺伝子をコードするいずれのベクターも含有しない培養結合組織細胞の集団を作製または獲得すること;および
c)関節内注射によりステップa)のタンパク質または細胞と、ステップb)の結合組織細胞と、を哺乳動物宿主の関節炎の関節腔に移入することで、関節腔内での該組み合わせの活性が硝子軟骨の再生をもたらすこと、
を含む、硝子軟骨を再生する方法を対象とする。
本明細書で用いられる用語「患者」は、非限定的に人類をはじめとする動物界のメンバーを包含する。
本発明は、哺乳動物宿主の結合組織細胞への該当するDNA配列の送達のためのエクスビボおよびインビボ技術を開示する。エクスビボ技術は、該当する遺伝子産物のインビボ発現を実行するために、標的哺乳動物細胞の培養、哺乳動物細胞への該当するDNA配列、DNAベクターまたは他の送達ビヒクルのインビトロトランスフェクションと、続いての哺乳動物宿主の標的関節への修飾哺乳動物細胞の移植を伴う。
本発明は、軟骨再生に関する。具体的実施形態において、本発明の方法は、トランスフォーミング増殖因子βスーパーファミリーのメンバー、またはその生物活性誘導体もしくは断片である遺伝子産物を用いることを含む。TGF-βスーパーファミリータンパク質は、同種異系軟骨細胞をはじめとする軟骨細胞などの結合組織細胞と併せて、投与される。軟骨が処置部位で再生される限り、TGF-βタンパク質は、該細胞と同時に投与されてもよく、またはそれは、該細胞の投与の前もしくは後に投与されてもよい。
様々な送達システム、例えば、リポソームへのカプセル化、マイクロ粒子、マイクロカプセル、化合物を発現することが可能な組換え細胞、受容体介在性エンドサイトーシス、レトロウイスルまたは他のベクターの一部としての核酸の構築などが公知であり、本発明の組成物を投与するために用いられ得る。導入方法としては、皮内、筋肉内、腹腔内、静脈内、皮下、鼻内、硬膜外および経口経路が挙げられるが、これらに限定されない。該化合物または組成物は、任意の簡便な経路により、例えば輸注またはボーラス注射により、上皮または皮膚粘膜の内壁(例えば、口腔粘膜、直腸および腸管粘膜など)を通した吸収により投与されてもよく、他の生物活性剤と共に投与されてもよい。投与は、全身または局所であり得る。加えて、脳室内および髄腔内注射をはじめとする任意の適切な経路により中枢神経系に本発明の医薬化合物または組成物を導入することが望ましく成り得、脳室内注射は、例えばOmmayaリザーバなどのリザーバに取り付けられた、脳室内カテーテルによって容易に行われてもよい。肺投与もまた、例えば吸入器またはネブライザの使用およびエアロゾル化剤の配合によって、用いられ得る。
実施例I 材料と方法
プラスミドの構築
メタロチオネイン発現構築物(pM)を作製するために、メタロチオネインIプロモータ(-660/+63)を、増幅に用いられるオリゴヌクレオチド中に組み込んだXba IおよびBam HI制限部位を利用したゲノムDNAを用いたポリメラーゼ連鎖増幅により作製した。増幅された断片をpBluescript(Stratagene、カリフォルニア州ラホヤ)のXba I-Bam HI部位にサブクローニングした。3’末端にTGF-β1コード配列および成長ホルモンポリA部位を含有する1.2kb Bgl II断片を、pMのBam HI-Sal I部位にサブクローニングすることにより、プラスミドpmTβ1を作製した。
安定した細胞株 - トランスフェクションを、リン酸カルシウム共沈法を利用することにより実行した(図1)。生存するコロニーの約80%が、導入遺伝子のmRNAを発現した。これらの選択されたTGF-β1生成細胞を、硫酸亜鉛溶液中でインキュベートした。細胞を100μM硫酸亜鉛溶液中で培養すると、それらはmRNAを生成した。TGF-β分泌速度は、約32ng/106細胞/24時間であった。
対照NIH 3T3またはNIH 3T3-TGF-β1のどちらかの細胞(5~7×105)に、6000rad(60Gy)を放射線照射して、ウサギ膝関節に注射した。これらの放射線照射された細胞は、組織培養皿の中で3週間内に完全に死滅した。注射の手順は、未処置細胞での過去のプロトコルと同様であった。膝関節を、注射後3または6週目に採取した。標本をホルマリンに固定し、硝酸で脱灰した。標本の切片を作製し、パラフィンで包埋し、その後、0.5μm厚の薄片に切り出した。図10では、サフラニン-O染色(A~D&A’~D’)およびヘマトキシリン-エオジン染色(E~F&E’ ~F’)を切片で実施して、再生軟骨組織を顕微鏡で観察した。(本来の倍率:(A、B、A’&B’)×12.5;(C~F&C’ ~F’)×400)。
対照ヒト包皮線維芽細胞(hFSF)またはhFSF-TGF-β1細胞のどちらかを、大腿顆の部分的軟骨欠損(3mm×5mm、1.5mm深)を含有するウサギ膝関節に注射した。これらの細胞(2×106細胞/mlの0.5ml)を過去のプロトコルの通り注射するか、または同じ濃度の20~25μl細胞を、欠損の最上部に載せた。後者の場合、細胞を欠損内に15~20分間放置して、それらを欠損の底部に定着させた後、縫合した。両方の例で、類似のレベルの軟骨再生が、得られた。標本を注射後6週目に採取して、顕微鏡で観察した。図11A&Bは、hFSF(A)またはhFSF-TGF-β1(B)のどちらかの細胞の注射の6週後の大腿顆の写真を示す。C、E&Gは、対照hFSF細胞を注射された大腿顆の切片のサフラニン-O染色(C&E)およびH&E染色(G)を示す。D、F、&Hは、hFSF-TGF-β1細胞を注射された大腿顆の切片のサフラニン-O染色(D&F)およびH&E染色(H)を示す。(本来の倍率:(C&D)×12.5;(E~H)×400)。

再生軟骨組織におけるTGF-β1タンパク質の発現を調査するために、注射の3週後の修復組織の免疫組織化学的染色を、TGF-β1抗体で実施した。結果から、再生軟骨の細胞のみで高レベルのTGF-β1タンパク質発現が示され、それらの多くは、新たに作製された軟骨細胞と思われた(図13A&B)。二次抗体のみでプローブされた同組織からの切片には、染色は見られなかった(図13C)。(本来の倍率:A×12.5;(B~C)×40)。
インビボで同時注射された組換えTGF-β1タンパク質により刺激された正常軟骨細胞が軟骨組織の再生を誘導し得る可能性を、探索した。ヒト軟骨細胞を様々な量の組換えTGF-β1タンパク質と混合した。この混合物を、ウサギまたはイヌの大腿顆の中間層軟骨欠損を含有する膝関節に注射した。
図15A~15Fは、イヌにおける正常軟骨細胞(hChon)と組換えTGF-β1タンパク質との混合物での軟骨再生を示す。hChonと組換えTGF-β1タンパク質との混合物またはhChon対照のどちらかを、大腿顆の中間層軟骨欠損(3mm×10mm、1~2mm深)を含有するイヌ膝関節に注射した。混合物(20~25μlの2×106hChon細胞/mlおよび100、200または400ngの組換えTGF-β1タンパク質)を欠損の最上部に載せ、その後、欠損中で15~20分間放置して混合物を創傷に浸透させた後、縫合した。標本を注射後8週目に採取して、顕微鏡で観察した。図2A、2Cおよび2Eは、hChonと200ng(A)または400ng(B)組換えTGF-β1タンパク質との混合物またはhChonのみ(E)のどちらかの注射の8週後の大腿顆の写真を示す。図2B、2Dおよび4Fは、hChonと200ng(A)または400ng(B)組換えTGF-β1タンパク質との混合物またはhChonのみ(F)のどちらかを注射された大腿顆からの切片のマッソントリクローム染色を示す。

Claims (18)

b)前記トランスフォーミング増殖因子βスーパーファミリーのメンバーをコードするベクターを含有する培養哺乳動物細胞の集団であって、前記哺乳動物細胞が、ヒト胎児腎臓細胞または上皮細胞である、培養哺乳動物細胞の集団;および
c)医薬的に許容できる担体、
を含む、骨関節炎を処置するのに使用するための組成物であって、
関節内注射により前記組成物を哺乳動物宿主の関節炎の関節腔に移入することで、結合組織の再生をもたらす、組成物。 - 前記哺乳動物細胞が、ウイルスベクターを含有する、請求項1に記載の組成物。
- 前記ウイルスベクターが、レトロウイルスベクターである、請求項2に記載の組成物。
- 前記ベクターが、プラスミドベクターである、請求項1に記載の組成物。
- 前記哺乳動物細胞が、同種異系または自家細胞である、請求項1に記載の組成物。
- 前記トランスフォーミング増殖因子βスーパーファミリーのメンバーが、TGF-β1、TGF-β2、TGF-β3、BMP-2、BMP-3、BMP-4、BMP-6、BMP-7、またはBMP-9である、請求項1に記載の組成物。
- a)タンパク質のトランスフォーミング増殖因子βスーパーファミリーのメンバー;
b)前記トランスフォーミング増殖因子βスーパーファミリーのメンバーをコードするベクターを含有する培養哺乳動物細胞の集団であって、前記哺乳動物細胞が、ヒト胎児腎臓細胞または上皮細胞である、培養哺乳動物細胞の集団;および
c)医薬的に許容できる担体、
を含む、硝子軟骨を再生するのに使用するための組成物であって、
関節内注射により前記組成物を哺乳動物宿主の関節炎の関節腔に移入することで、結合組織の再生をもたらす、組成物。 - 前記哺乳動物細胞が、ウイルスベクターを含有する、請求項7に記載の組成物。
- 前記ウイルスベクターが、レトロウイルスベクターである、請求項8に記載の組成物。
- 前記ベクターが、プラスミドベクターである、請求項7に記載の組成物。
- 前記哺乳動物細胞が、同種異系または自家細胞である、請求項7に記載の組成物。
- 前記トランスフォーミング増殖因子βスーパーファミリーのメンバーが、TGF-β1、TGF-β2、TGF-β3、BMP-2、BMP-3、BMP-4、BMP-6、BMP-7、またはBMP-9である、請求項7に記載の組成物。
- a)プロモータに動作可能に連結されたタンパク質のトランスフォーミング増殖因子βスーパーファミリーのメンバーをコードするDNA配列を含む組換えウイルスまたはプラスミドベクター;
b)前記ベクターでトランスフェクトされた同種異系哺乳動物細胞の集団であって、前記哺乳動物細胞が、ヒト胎児腎臓細胞または上皮細胞である、同種異系哺乳動物細胞の集団;および
c)医薬的に許容できる担体、
を含む、硝子軟骨を再生するのに使用するための組成物であって、
関節内注射により前記組成物を哺乳動物宿主の関節炎の関節腔に移植することで、前記関節腔内の前記DNA配列の発現が硝子軟骨の再生をもたらす、組成物。 - タンパク質の前記トランスフォーミング増殖因子βスーパーファミリーのメンバーが、TGF-β1、TGF-β2、TGF-β3、BMP-2、BMP-3、BMP-4、BMP-6、BMP-7、またはBMP-9である、請求項13に記載の組成物。
- a)プロモータに動作可能に連結されたタンパク質のトランスフォーミング増殖因子βスーパーファミリーのメンバーをコードするDNA配列を含む組換えウイルスまたはプラスミドベクター;
b)前記ベクターでトランスフェクトされた同種異系哺乳動物細胞の集団であって、前記哺乳動物細胞が、ヒト胎児腎臓細胞または上皮細胞である、同種異系哺乳動物細胞の集団;および
c)医薬的に許容できる担体、
を含む、骨関節炎を処置するのに使用するための組成物であって、

- a)タンパク質のトランスフォーミング増殖因子βスーパーファミリーのメンバー;
b)前記トランスフォーミング増殖因子βスーパーファミリーのメンバーをコードするベクターを含有する培養哺乳動物細胞の集団であって、前記哺乳動物細胞が、ヒト胎児腎臓細胞または上皮細胞である、培養哺乳動物細胞の集団;および
c)医薬的に許容できる担体、
を含む、関節における結合組織への傷害を処置するのに使用するための組成物であって、
関節内注射により前記組成物を哺乳動物宿主の関節炎の関節腔に移入することで、結合組織の再生をもたらす、組成物。 - a)プロモータに動作可能に連結されたタンパク質のトランスフォーミング増殖因子βスーパーファミリーのメンバーをコードするDNA配列を含む組換えウイルスまたはプラスミドベクター;
