Description
本開示は、部位特異的な突然変異誘発によって熱的に安定化されたFGF7ポリペプチドを提供する。本開示では、生物情報分析およびコンピュータを用いたタンパク質設計を通じて、従来知られていなかった新規位置で最も適したアミノ酸を合理的に予測した後、部位-特異的な突然変異誘発によって、突然変異させた配列で構成された遺伝子およびポリペプチドを製造した。
表1に開示されている多様な変異体は、薬剤学的にまたは化粧用に許容可能な担体と共に、薬剤学的および/または化粧品組成物として提供できる。
表1に開示されている多様な変異体は、少なくとも5回の継代培養の間に未分化された状態で万能性幹細胞を維持させるのに必要な量に該当する「培地学的有効量」で含まれて、ヒト万能性幹細胞培地に提供できる。
FGF7について、1つの位置の突然変異(A120C)、2つの位置の突然変異(A120C、K126E)、2つの位置の突然変異(A120C、K178E)、3つの位置の突然変異(A120C、K126E、K178E)を合成し、His-Tagを有するpCold Iベクターにサブクローニングした。FGF7が挿入された組換えベクターを、BL21(DE3)細胞に形質転換して、発現させた。
組換えFGF7タンパク質に対してスペクトルデータを測定した。図3を参照すれば、還元(Reduced Condition)および非還元(Non-reduced Condition)サンプルに対して得られた基本ピーククロマトグラム(base peak chromatogram)は、ペプチド(14.4、26.3、and 32.63min)が二硫化結合と関連があることを示す。
図5および図6に示されているように、3D構造を基盤にして分析した結果、175番目アルギニン(R)残基(R175)が126番目残基と178番目残基との間の中間に位置しており、アミノ酸の間の距離が塩橋を形成する程度に十分に近いことを確認することができる。スイスモデルサーバーを用いて予測した結果、126番目リシン(K)がグルタミン酸(E)で置換された(K126E)場合に、K126Eの陰イオン性OE2とR175のNH2との間の距離が、2.56Åで維持されることが分かる。そして、178番目リシン(K)がグルタミン酸(E)で置換された(K178E)の場合にも、K178EのOE1とR175のNH1との間の距離は、2.76Åで維持されることが分かる。これら分析は、R175の陽に荷電されたグアニジニウム(guanidinium)が、K125EおよびK178Eの陰に荷電されたカルボキシル基と相互作用して、水素結合とイオン結合との2種類の非共有相互作用の組み合わせである塩橋を形成することができることを示唆する。
Δ53_hFGF7(palifermin)とFGF7突然変異体との熱力学的安定性を比較するために、190nmでCD分光器を使用して、FGF7突然変異体の熱による形態変化をモニターした。その結果が図7に示されている。中間点転移温度(Tm)は、熱変性曲線を使用して分析した。Δ53_hFGF7(palifermin)のTm値は、51.9℃であって、野生型FGF7の45.5℃よりも6.4℃高かった。反面、FGF7突然変異体(A120C、A120C/K126E、A120C/K178E、A120C/K126E/K178E)のTm値は、それぞれ、52.6℃、60.4℃、58.8℃、67.1℃であった。FGF7突然変異体を従来のΔ53_hFGF7(palifermin)と比較すれば、Tm値が、それぞれ、0.67℃、8.53℃、6.89℃および15.24℃増加した。特に、K126E/K178E二重突然変異を含む場合のTm値は、K126EまたはK178Eの単一突然変異のTm値よりも高く示されることを、確認することができた。
精製されたFGF7タンパク質について、1X PBSバッファーを基本使用して0.5mg/ml濃度で37℃で0、4、8、12、15日間反応させ、クマシーブルー染色試薬で染色して、15%SDS-PAGE電気泳動を実施した。SDS-PAGE GELに残っているタンパク質バンドについて、ImageJプログラム(Wayne Rasband、NIH)を使用して、定量的に密度(density)を測定した。
精製されたFGF7タンパク質について、1X PBSバッファーを基本使用して0.5mg/ml濃度で45℃で0、1、2、3、4、5、6、7日間反応させ、クマシーブルー染色試薬で染色して、15% SDS-PAGE電気泳動を実施した。図10にその結果が例示されている。
突然変異体に対してHaCaT細胞を使用し、10%ウシ胎仔血清(fetal bovine serum)を含むDMEM培地で、培養および維持した。FGF7による細胞増殖活性を確認するために、細胞には、0.03%BSAを含む無血清(serum-free)DMEM培地を使用した。

- 配列番号1で、120番目のアラニン(A)がシステイン(C)で置換され、
126番目のリシン(K)がグルタミン酸(E)またはアスパラギン酸(D)で置換されることか、178番目のリシン(K)がグルタミン酸(E)またはアスパラギン酸(D)で置換されることのうちから選択された少なくとも一つ以上の置換を含んで175番目のアルギニン(R)と塩橋を形成し、
133番目のシステイン(C)と137番目のシステイン(C)とが二硫化結合され、
配列番号1のN末端で2~54番目のアミノ酸を欠失した(Δ53N)配列を有し、
FGF7活性を有する温度安定性が向上した(thermally stable)ポリペプチド。 - 前記126番目のリシン(K)がグルタミン酸(E)で置換され、
前記178番目のリシン(K)がグルタミン酸(E)で置換された配列を有する、請求項1に記載の温度安定性が向上したポリペプチド。 - 請求項1または2に記載の温度安定性が向上したポリペプチド、および、薬剤学的にまたは化粧用に許容可能な担体を含む、組成物。
