本実施形態のAPTT測定用試薬キット(以下、「試薬キット」ともいう)は、エラグ酸化合物と、上記の式(I)で表される化合物とを含む第1試薬と、カルシウム塩を含む第2試薬とを含む。第1試薬として、上記の本実施形態のAPTT試薬を用いることができる。第1試薬の詳細は、本実施形態のAPTT試薬について述べたことと同じである。エラグ酸化合物の詳細は、上記のとおりである。
別の実施形態では、エラグ酸化合物の沈殿を抑制する方法が提供される。この方法では、エラグ酸化合物と、上記の式(I)で表される化合物とを水溶液中で共存させることにより、エラグ酸化合物の沈殿を抑制する。エラグ酸化合物及び式(I)で表される化合物の詳細は、上記のとおりである。水溶液は、エラグ酸化合物及び式(I)で表される化合物を溶解可能な水性溶媒であればよい。そのような水性溶媒の詳細は、本実施形態のAPTT試薬に用いられる水性溶媒について述べたことと同じである。
本実施形態のAPTTの測定方法は、(以下、「測定方法」ともいう)は、血液検体と、エラグ酸化合物及び上記の式(I)で表される化合物を含む第1試薬と、カルシウムイオンを含む第2試薬とを混合して、凝固時間を測定することを含む。第1試薬として、上記の本実施形態のAPTT試薬を用いることができる。あるいは、第1試薬及び第2試薬として、上記の本実施形態の試薬キットを用いることができる。
活性化剤としてエラグ酸の金属イオン錯体を含むAPTT試薬では、塩濃度が増加すると、塩析効果によって錯体粒子径が増加して、最終的に沈殿が生じる。この塩析の原理を利用して、エラグ酸金属イオン錯体溶液における各種の添加剤の沈殿抑制効果を確認した。
緩衝液成分として50 mM HEPES、8 mM TAPS及び1質量%グリシン、エラグ酸の金属イオン錯体として100μMエラグ酸、60μM塩化亜鉛及び50μM塩化アルミニウム、防腐剤として0.05% ProClin(商標)300(Sigma-Aldrich社)を含むエラグ酸金属イオン錯体溶液を調製した。この溶液と、添加剤として没食子酸、ピロガロール、ヒドロキノン、5-メチルピロガロール、レゾルシノール、及びシクロヘキサントリオールから選択されるいずれか1つとを混合して、各添加剤を100μMの濃度で含む錯体溶液を得た。対照として、添加剤を含まないエラグ酸金属イオン錯体溶液を用いた。
上記(1)で調製した各錯体溶液と5 M塩化ナトリウム溶液とを混合して、塩化ナトリウム濃度が10 mM、50 mM又は100 mMである混合液(試験区)を得た。ゼータサイザーナノ(マルバーン社)を用いた動的散乱光法によって、各混合液の塩濃度と、錯体の平均粒子径との関係性を確認した。結果を図3に示す。図3に示されるように、没食子酸又はピロガロールを添加した試験区では、塩濃度の増加に対する粒子径の増加の度合いが小さいことがわかる。実際、塩化ナトリウム濃度が100 mMの場合、没食子酸又はピロガロールを添加した試験区では、沈殿は目視で確認できなかったが、それ以外の添加物を含む試験区及び添加剤なしの試験区では、沈殿が目視で確認された。
錯体粒子の沈殿を抑制する効果が認められた没食子酸及びピロガロールを、エラグ酸金属イオン錯体溶液に図4A及び4Bに示す濃度となるように添加した。没食子酸又はピロガロールを種々の濃度で含む錯体溶液を用いて、上記(2)と同様にして沈殿抑制効果を確認した。結果を図4A及びBに示す。いずれの濃度でも、沈殿抑制効果が認められることが示唆された。
ピロガロール又は没食子酸を含むAPTT試薬を用いることで、初期反応異常が抑制されるかを検討した。
緩衝液成分として50 mM HEPES、8 mM TAPS及び1質量%グリシン、エラグ酸の金属イオン錯体として100μMエラグ酸、60μM塩化亜鉛及び50μM塩化アルミニウム、防腐剤として0.05% ProClin(商標)300(Sigma-Aldrich社)を含むエラグ酸金属イオン錯体溶液を調製した。錯体溶液と、添加剤として没食子酸又はピロガロールとを混合して、没食子酸又はピロガロールを100μMの濃度で含む錯体溶液を得た。対照として、添加剤を含まないエラグ酸金属イオン錯体溶液を用いた。リン脂質成分として、PE(L-α-ホスファチジルエタノールアミン)、PC(L-α-ホスファチジルコリン)及びPS(L-α-ホスファチジルセリン)を、それぞれの終濃度が10 mg/dL、45 mg/dL及び5 mg/dLとなるように各錯体溶液に添加して、APTT試薬を調製した。各APTT試薬における没食子酸及びピロガロールの含有量は、それぞれ0.013質量%及び0.009質量%であった。また、各APTT試薬において、エラグ酸100質量部当たりの没食子酸及びピロガロールの含有量は、それぞれ43質量部及び32質量部であった。
凍結血漿に、超低比重リポタンパク質(VLDL)及びC反応性タンパク質(CRP)をそれぞれ100μg/mL及び300μg/mLの終濃度となるように添加して、凝固の初期反応異常を誘引する疑似試料を調製した。この疑似試料について、上記(1)で調製した試薬及びカルシウムイオン含有試薬をセットした全自動凝固測定装置CS-5100(シスメックス株式会社)によりAPTT測定を行い、その凝固波形を取得した。図5に、各APTT試薬を用いた測定で得られた凝固波形を示す。

Claims (13)
- 前記式(I)で表される化合物が、ピロガロール及び没食子酸からなる群より選択される少なくとも1つである請求項1に記載の試薬。
- 前記式(I)で表される化合物の含有量が、0.001質量%以上0.02質量%以下である請求項1又は2に記載の試薬。
- 前記式(I)で表される化合物の含有量が、前記エラグ酸化合物100質量部当たり5質量部以上65質量部以下である請求項1~3のいずれか1項に記載の試薬。
- 金属イオン形成化合物をさらに含む請求項1~4のいずれか1項に記載の試薬。
- 前記金属イオン形成化合物が、亜鉛イオン形成化合物とアルミニウムイオン形成化合物との混合物、又はマンガンイオン形成化合物とアルミニウムイオン形成化合物との混合物である請求項5に記載の試薬。
- 非極性アミノ酸をさらに含む請求項1~6のいずれか1項に記載の試薬。
- 前記非極性アミノ酸が、グリシン、アラニン及びフェニルアラニンからなる群より選択される少なくとも1つである請求項7に記載の試薬。
- 前記非極性アミノ酸の含有量が、0.75質量%以上である請求項7又は8に記載の試薬。
- リン脂質をさらに含む請求項1~9のいずれか1つに記載の試薬。
- 前記リン脂質が、ホスファチジルセリン、ホスファチジルエタノールアミン及びホスファチジルコリンからなる群より選択される少なくとも1つである請求項10に記載の試薬。
