Description
本発明の別の目的は、調節可能なLCSTを有し、且つ温度に応じてナノ粒子を形成可能なブロックコポリマーを提供することである。
本発明の別の目的は、特定の調節可能なLCST以上の水性条件において、正確な生物活性及び刺激応答性自己組織化特性を備えた材料を提供することである。
R’は、プロリン及びその誘導体以外の天然もしくは合成アミノ酸の側鎖である]
を有する少なくとも1つの繰り返し単位を含む。
「プロリン誘導体」なる語は、プロリン骨格に基づく任意の非標準アミノ酸、すなわちα、β、γ、またはδ炭素原子に置換基を有するプロリン、例えば4-ヒドロキシプロリンまたはα-メチルプロリンを企図する。
好ましくは、Xaaアミノ酸は、バリンまたはメチオニンである。

材料
アクロレイン(95%)、アジ化ナトリウム(NaN3、99.5%)、酢酸(AcOH、99.8%)、メトキシアミン塩酸塩(98%)、ナトリウムシアノボロハイドライド(NaBH3CN、95%)、塩酸(HCl、37%)、4-ペンチン酸(97%)、N,N’-ジシクロヘキシルカルボジイミド(DCC, 99%)、N-ヒドロキシスクシンイミド(NHS、98%)、トリメチルアミン(TEA、99%)、硫酸銅(II)五水和物(CuSO4、99%)、ジクロロメタン(DCM、99.9%)、N,N-ジメチルホルムアミド(DMF、99.8%)、ジメチルスルホキシド(DMSO、99.7%)、メタノール(MeOH、99.8%)、ジエチルエーテル(99.9%)、及び無水硫酸マグネシウム(MgSO4、99.5%)を、Sigma-Aldrichから購入した。N,N-ジイソプロピルエチルアミン(DIPEA,99%)、酢酸ナトリウム(AcONa、99%)、及びアスコルビン酸ナトリウム(NaAsc、99%)を、Alfa Aesarから入手した。メトキシポリエチレングリコール(mPEG)、トリス(ベンジルトリアゾリルメチル)アミン(TBTA、97%)及び4-トルエンスルホニルクロライド(TsCl、99%)を、TCIから購入した。Cuprisorb(登録商標)をSeachemから購入した。デキストラン(Dex、T10)をpharmacosmosから購入した。ラミナリヘキサオース(Hex)をMegazymeから購入した。ヒアルロナン酸ナトリウム(HA)をLifecore Biomedicalから購入した。水を、ELGA PURELAB Classicシステムを用いて精製した。溶媒を、Innovative TechnologyのPureSolv MD-5溶媒精製システムを用いて精製した。透析は、Spectra/Por(登録商標)6透析膜を用いて行った。
アジドリンカーの調製
丸底フラスコ内の酢酸(4mL)を、-20℃に冷却し、アクロレイン(1.84mL、27.4mmol)を加え、続いてH2O(10.4mL)中のアジ化ナトリウム(2.38g、41.2mmol)の溶液を滴下した。混合物を-20℃で1.5時間攪拌し続けた。その後これに飽和炭酸水素ナトリウム水溶液(sat.aq.NaHCO3 80mL)を加えて急冷し、得られた混合物をDCM(2×100mL)で抽出した。合わせた有機抽出液を、飽和NaHCO3水溶液(150mL)で洗浄し、無水MgSO4上で乾燥させ、ろ過し、真空濃縮して100mLとした。DCM中の溶液に、メトキシアミン塩酸塩(2.68g、31.68mmol)及び酢酸ナトリウム(4.42g、54mmol)を加え、混合物を室温で一晩撹拌した。飽和NaHCO3水溶液(150mL)を加えて反応を急冷し、得られた混合物をDCM(2×100mL)で抽出した。合わせた有機抽出液を飽和NaHCO3水溶液(150mL)で洗浄し、無水MgSO4上で乾燥させ、ろ過し、真空濃縮して100mLとした。DCM中の溶液に、NaBH3CN(2g、32mmol)を加え、続いて1Mのエタノール性HCl(32mL、エタノールに塩化アセチルを加えて新たに調製したもの)を滴下した。得られた混合物を室温で1.5h撹拌した。その後、溶媒をエバポレーターで除去し、得られた白色固体を飽和NaHCO3水溶液(150mL)中に懸濁させ、DCMで抽出した(2×100mL)。合わせた有機抽出物を、飽和NaHCO3水溶液(150mL)で洗浄し、無水MgSO4上で乾燥させ、ろ過し、真空中で濃縮して、粗製のN-(3-アジドプロピル)-O-メチルヒドロキシルアミンを黄色油として得た。粗生成物をシリカゲルカラムクロマトグラフィー(DCM中1-3%のMeOH)で精製し、N-(3-アジドプロピル)-O-メチルヒドロキシルアミン(アジドリンカー、1.1g、3ステップで62%)が無色のオイルとして得られた。

デキストラン(MW8000)(1g、0.125mmol)の酢酸緩衝液(AcOH/AcONa、2M、pH4.6、4.2mL)中の溶液に、アジドリンカー(380mg、2.9mmol)を加え、反応混合物を40℃のサーモミキサー上で9日間振とうした(1日に3回ボルテックス)。その後、混合物を、純水で透析バッグ(MWCO 1000)を用いて24時間透析することにより精製した(1日に3回水を交換)。最終生成物を、凍結乾燥によって得た(白色粉末、805mg、収率79%)。

ラミナリヘキサオース(500mg、0.5mmol)の酢酸緩衝液(AcOH/AcONa、2M、pH4.6、5mL)中の溶液に、アジドリンカー(900mg、6.9mmol)を加え、反応混合物を40℃のサーモミキサー上で8日間振とうした(1日に3回ボルテックス)。その後、混合物を凍結乾燥させ、水5mLに再溶解させ、純水で透析バッグ(MWCO100)を用いて36時間透析することにより精製した(1日3回水を交換)。最終生成物を、凍結乾燥によって得た(白色粉末、302mg、収率54%)。

ヒアルロン酸ナトリウム(MW7000)(1g、0.14mmol)の酢酸緩衝液(AcOH/AcONa、2M、pH5.5、5mL)中の溶液に、アジドリンカー(520mg、4mmol)及びシアノ水素化ホウ素ナトリウム(65mg、1mmol)を加え、反応混合物を50℃のサーモミキサー上で5日間振とうした(1日に3回ボルテックス)。その後、混合物を水5mLで希釈し、続いて純水で透析バッグ(MWCO 1000)を用いて24時間透析した(1日に3回水を交換)。最終生成物を、凍結乾燥によって得た(白色粉末、610mg、収率60%)。

1.ペンチン酸-NHSエステルの合成
ペンチン酸(210mg、2.14mmol)のDCM(8mL)中の溶液に、N,N'-ジシクロヘキシルカルボジイミド(480mg、2.3mmol)を加えた。混合物を5分間撹拌した後、N-ヒドロキシスクシンイミド(260mg、2.3mmol)を加えた。この反応物を室温で3h継続して撹拌した。その後、析出したジシクロヘキシルカルバミドをセライトで濾過し、濾過ケーキを冷温DCMで洗浄した(2×10mL)。濾液を回収し、DCMを真空下で除去した。生成物をEtOAc(10mL)に再溶解させ、0℃の冷蔵庫で20分間冷却した。析出物をセライトで再度ろ過し、ろ液を飽和NaHCO3(2×50mL)及びブライン(2×50mL)で洗浄し、無水MgSO4上で乾燥させ、ろ過し、真空中で濃縮して、粗生成物を無色のオイルとして得た。残渣をシリカゲルカラムクロマトグラフィー(石油エーテル:EtOAc=2:1)で精製することにより、ペンチン酸NHS-エステルを白色固体として得た(300mg、収率72%)。

ELP(225mg、13.2μmol)の無水DMSO(18mL)中の溶液に、ペンチン酸NHS-エステル(92mg、0.47mmol)及びN,N-ジイソプロピルエチルアミン(1.7mg、13.2μmol)を加えた。この反応物を室温で72h撹拌した。その後、混合物を水(20mL)で希釈し、得られた溶液を透析バッグ(MWCO15000)中の純水で2日間透析した(1日に3回水を交換)。最終生成物を、凍結乾燥によって得た(白色粉末、210mg、収率93%)。

1.デキストラン-ELP
アルキン-ELP(65mg、3.8μmol)、デキストラン-アジド(46mg、5.7μmol)、硫酸銅(6mg、22.8μmol)、アスコルビン酸ナトリウム(10mg、45.6μmol)、及びTBTA(12mg、22.8μmol)を、無水DMSO(8mL)にアルゴン雰囲気下で溶解させた。この混合物を室温で3日間撹拌した。その後、混合物を冷水(20mL)で希釈し、4℃の冷蔵庫で20分間冷却した。TBTAが析出し、これを遠心分離機で除去した。Cuprisorb(120mg)を上澄み液に加え、得られた溶液を室温で一晩振とうして銅を除去した。Cuprisorbを遠心分離で除去し、上澄み液を透析バッグ(MWCO 15000)中の純水で5日間透析した(1日に3回水を交換)。最終生成物を、凍結乾燥によって得た(白色粉末、92mg、収率90%)。

ヒアルロナン-アジドは、まず塩酸水溶液を加えることで酸性化し、DMSOに完全に溶解させた。アルキン-ELP(60mg、3.5μmol)、硫酸銅(9mg、36μmol)、アスコルビン酸ナトリウム(18mg、90μmol)、及びトリス(ベンジルトリアゾリルメチル)アミン(TBTA、22mg、41μmol)を、無水DMSO(4mL)にアルゴン雰囲気下で溶解させた。無水DMSO(2mL)に完全に溶解したヒアルロナン-アジド(18mg、3.5μmol)の溶液を加えた。この反応物を40℃で4日間撹拌した。その後、混合物を、冷水(20mL)で希釈し、4℃の冷蔵庫で20分間冷却した。TBTAが析出し、これを遠心分離で除去した。Cuprisorb(180mg)を上澄み液に加え、得られた溶液を室温で一晩振とうして銅を除去した。Cuprisorbを遠心分離によって除去し、上澄み液を透析バッグ(MWCO 15000)中の純水で5日間透析した(1日に3回水を交換)。粗生成物を凍結乾燥によって得た(白色粉末、53mg)。脱イオン水(5.3mL)を加え、溶液を40℃に加熱して1時間保持した。不溶性の未反応ELPを40℃での遠心分離によって除去し、上澄み液を凍結乾燥して最終生成物を得た(白色粉末、42mg、収率54%)。

温度誘起自己組織化研究
水中における多糖類-ELPバイオコンジュゲート(本発明によるブロックコポリマーに相当)の温度誘起自己組織化を、動的光散乱(DLS)、静的光散乱(SLS)、及び液体原子間力顕微鏡法(液体AFM)によって調べた。
動的光散乱測定(DLS)
動的光散乱測定は、NanoZS計器(Malvern、U.K.)を用い、キュベット内の定位置で90°の角度で行った(一定の散乱体積)。得られるカウントレート(DCR)を、減衰率で正規化した平均散乱強度として定義した。DCRを、温度に対してプロットし、Ttを、このプロットにおいてDCRが増加し始める点に相当する温度として定義した。
静的光散乱測定は、フルデジタル相関器を、Spectra Physicsレーザー(λ=632.8nmで垂直偏光を照射)及び恒温槽コントローラー(20から50℃の範囲)と組み合わせて備えた、ALV/CG6-8Fゴニオメーターを使用して実施した。データは、ALV相関器ソフトウェアを用いて得られ、計数時間は、30°から150°の範囲で10段階に分けた各散乱角で、典型的には15秒であった。流体力学的半径(Rh)を、見かけの拡散係数及びStokes-Einsteinの式から決定した。旋回半径(Rg)は、同じ角度での平均散乱強度の測定から得られたギニエプロットから決定した。
温度制御された液体原子間力顕微鏡法による測定は、Dimension FastScan Bruker AFMシステムを用いて実施した。バイオコンジュゲートのトポグラフィー画像を、典型的な先端半径5nmを有するシリコンカンチレバー(ScanAsyst-Fluid+、Bruker)を用い、Peak Forceタッピングモードで得た。カンチレバーの共振は150kHzであり、バネ定数は0.7N/mであった。基板はAgar Scientificから購入した。試料は、50μM(HA-ELPの場合は150μM)のバイオコンジュゲート水溶液の、新たに劈開したマイカまたはHOPG表面へのドロップキャスティングにより調製し、これを直接画像化に用いた。AFM画像処理は、特定温度の液体環境中で行われた。外部加熱ステージ(Bruker)を用いて、,基板表面に標的温度を実現させた。
Dex-ELPの自己組織化研究
DLSを高速で加熱したところ、Dex-ELP(水中125μM)の転移温度(Tt)が約40℃であることが判明したが、これはELP及び物理的混合物のTtに比べて高かった。実際のところ、親水性ブロックのELPへの結合により、Ttの上昇がもたらされた。
Hex-ELPは親水性の割合が少ないため、Hex-ELPの転移温度(Tt)は約33℃であって、ELPのTtに比べてやや高めであった。Hex-ELPのTt以下の低温では、小さな物体及び少数の凝集体が、非常に低い散乱強度をもって観察された(30℃)。散乱強度は転移温度(約33℃)で急激に増加し、自己組織化を引き起こして、流体力学直径(Dh)が約210nmである構造体を形成した。Hex-ELPの温度応答性は完全に可逆的である。温度を45℃以上に加熱すると、ナノ粒子は散乱強度にほとんど変化を示さず、約400~500nmの直径Dhを示した(図5)。
UV-vis測定と同様に、DLS加熱過程でのHA-ELPのTtは約45℃と判明した。HA-ELPのTt以下の低温では、非常に低い散乱強度で小さな物体が観察された(35℃)。散乱強度は転移温度で急激に増加し、自己組織化を引き起こして、48-50℃でDhが約300nmの構造を形成した(図10-11)。HA-ELPを55℃以上に加熱したところ、ナノ粒子の直径が不安定となり、2つのサイズ分布に分離した。このことは液体AFMでも確認された(図14C)。
Claims (11)
- 少なくとも1つのオリゴ糖もしくは多糖ブロック及び少なくとも1つのエラスチン様ポリペプチドブロックを有し、且つ下式(II):
・Rは、(C1-C6)アルキル基であり、
・Xは、オリゴ糖もしくは多糖であり、
・X’は、(C1-C6)アルキレン基であり、
・Yは、以下の基:
R3及びR4は、Hまたはこれらを担持する炭素原子と共にシクロヘキシル基を形成し、
R5は、Hまたはアルキル基である}
からなる群より選択される基であり、
・iは、1から6の整数であり、
・X1は、共有結合または式:
・X2は、共有結合または式-(AA)j-の基であり、jは、1から6の整数であり、AAは、個別に天然もしくは合成のアミノ酸であり、
・nは、1から200の整数であり、
・R’は、プロリン及びその誘導体以外の天然もしくは合成のアミノ酸の側鎖である]
を有するブロックコポリマー。 - R’が、-CH(CH3)2または-(CH2)2SCH3である、請求項1に記載されるブロックコポリマー。

- オリゴ糖または多糖が、ヒアルロナン、ラミナリヘキサオース、デキストラン、またはガラクタンである、請求項5に記載されるブロックコポリマー。
