Description
(1)白子抽出物を有効成分として含有する、糖の吸収抑制用剤であって、該白子抽出物が、分子量5000以下の画分を50~100%含むように低分子化された魚類白子の加水分解物であることを特徴とする糖の吸収抑制用剤。
(2)白子抽出物が、白子由来DNAの加水分解物及び白子由来タンパク質の加水分解物を含むものである、(1)に記載の糖の吸収抑制用剤。
(3)糖の吸収抑制が、糖輸送体を介するものである、(1)又は(2)に記載の糖の吸収抑制用剤。
(4)(1)~(3)の何れかに記載の糖の吸収抑制用剤を含んでなる、糖の吸収抑制用飲食品。
1.トランスポーターに関する情報
先ず、本実験で用いた糖輸送体(グルコーストランスポーター)について説明する。グルコースをはじめとした単糖類は生体維持に必須の有機物質であるが、水溶性分子であるためこれらの細胞内外への輸送は細胞膜上に存在する膜タンパク質 (トランスポーター) に依存する。ナトリウム依存性グルコース輸送体(SGLT)1(配列番号1)はSLC5A1遺伝子(配列番号2)によってコードされる14回膜貫通型のトランスポーターであり、小腸上皮や腎近位尿細管の管腔側に発現している1,2)。
2-1.使用細胞
ヒト結腸腺癌由来細胞株Caco-2細胞はDSMZ(Deutsche Sammlung fur Mikroorganismen und Zellkulturen)より購入した。
Caco-2細胞の培養は、37℃-5%CO2インキュベーター内で行い、非働化した10% FBS、100 IU/mL ペニシリン-100μg/mL ストレプトマイシン、及び1%NEAA(non-essential amino acids)を含むDMEM(Dulbecco's modified Eagle's medium)を培養液とした。細胞は、100mmシャーレ(TPP)で培養し、播種後7日目に継代を行った。継代は、滅菌PBS(phosphate buffered saline;137mM NaCl、2.68mM KCl、8.1mM Na2HPO4・12H2O、1.47mMKH2PO4)で細胞を洗浄後、0.25% トリプシン及び0.02% EDTAを含むPBSにより細胞をシャーレから剥離させて行った。
SMEは培養液に直接溶解し、0.22μmフィルターでろ過滅菌した後実験に用いた。
本実施例においてSMEは日生バイオ株式会社のサケ白子抽出物を使用した。この製品は、特開2004-16143号公報の実施例1の記載に準じて、サケ白子を前処理し、続いてプロテアーゼ及びヌクレアーゼで加水分解することにより調製されたものである。本品のGPC分析結果を図1に、保持時間と分子量の関係を表1に示す。分子量目安の12,000はチトクロームC(MW12,400)の保持時間を基とした。
分子量12,000~5,000の画分(溶出時間20分~25分未満):0.3%
分子量5,000以下の画分(溶出時間25分以降):99.6%
以上のことから、実施例で用いたサケ白子抽出物における、DNA成分の分子量が12,000以下である画分は99.9%である。
分子量12,000~5,000の画分(溶出時間20分~25分未満):0.5%
分子量5,000以下の画分(溶出時間25分以降):99.4%
以上のことから、実施例で用いたサケ白子抽出物における、タンパク質成分の分子量が12,000以下である画分は99.9%である。
1)細胞からのtotal RNAの抽出
Caco-2細胞を12穴プレートに20万cells/wellで播種し、2日おきに培養液の交換を行った。播種後13日後に、各種濃度(100μg/mL又は1000μg/mL)のSMEを含む培養液、又はSMEを含まない培養液を添加した。24時間後、培養液を吸引しISOGENII(ニッポンジーン)を加え、このプロトコル(RNA抽出用試薬ISOGENIIマニュアル(第5版) Code No.311-07361)に従いtotal RNAを抽出した。なお、RNA濃度はNanodropにより測定した。
逆転写反応はReverTra Ace(TOYOBO)を使用し、表2に示した反応液中で、30℃で10分間、42℃で60分間、99℃で5分間、及び4℃で保温の順の条件下で行った。
Caco-2細胞を24穴プレートに10万 cells/wellで播種し、2日おきに培養液の交換を行った。播種後13日後にSME(100μg/mL)を含む培養液、又はSMEを含まない培養液を添加し、24時間後に取り込み実験に使用した。培養液を除去し、37℃のglucose-free Hank’s balanced salt solution(HBSS)(pH 7.4)500μLにより細胞を2回洗浄した後、同様のHBSS500μLを添加し、37℃で10分間プレインキュベートした。HBSSを除去した後、0.4μM(0.1μCi/mL)[14C]αMDG(α-メチル-D-グルコピラノシド)を500μL添加し、37℃で10分間インキュベートして細胞内に[14C]αMDGを取り込ませた。
1) Caco-2細胞からのタンパク質抽出
Caco-2細胞からのタンパク質の抽出及び定量はRIPAbuffer及びPierce BCA Protein Assay Kitを用いて、添付のプロトコルに準拠して行った。
サンプル(30μg)を同量の2×Sodium Dodecyl Sulfate(SDS) sample buffer (0.1 M Tris-HCl(pH6.8)、4% SDS、10% 2-メルカプトエタノール、20% グリセロール、0.004% bromophenol blue(BPB))と混合し、100℃で5分間加熱処理を行いタンパク質サンプルとして用いた。
SGLT1及びGLUT2のmRNA発現量に及ぼすSMEの影響を評価したところ、いずれの発現量も、SME未処理の場合と比べ、SME(100μg/mL及び1000μg/mL)処理により有意に減少した(図2)。また、SGLT1の基質であるαMDGを用いてCaco-2細胞内への取り込みを評価したところ、SME未処理の場合と比べ、100μg/mLのSME処理(24時間)によりαMDG取り込み量は有意に減少した(図3)。
以上より、SME処理によるSGLT1発現の減少により、その結果αMDGの取り込みが減少することが示された。

- 白子抽出物を有効成分として含有する、糖の吸収抑制用剤であって、該白子抽出物が、分子量5000以下の画分を50~100%含むように低分子化された魚類白子の加水分解物であることを特徴とする糖の吸収抑制用剤。
- 白子抽出物が、白子由来DNAの加水分解物及び白子由来タンパク質の加水分解物を含むものである、請求項1に記載の糖の吸収抑制用剤。
- 糖の吸収抑制が、糖輸送体を介するものである、請求項1又は2に記載の糖の吸収抑制用剤。
- 請求項1~3の何れか1項に記載の糖の吸収抑制用剤を含んでなる、糖の吸収抑制用飲食品。
