本開示は、他の方法の中でとりわけ、頭部への損傷を負った、又は負った可能性があるヒト対象に対して、MRIスキャン又はCTスキャンなどのイメージング手順を実施するのかどうかについて決定する一助となる方法に関する。本明細書において使用された「ヒト対象に対して、MRIスキャン又はCTスキャンなどのイメージング手順を実施するのかどうかについての決定」とは、対象のMRIが陽性となる、又は頭部CTスキャンが陽性となる、すなわち、頭蓋内病変が存在する可能性が、そうでない場合より高いことを決定するのに、他の情報(例えば、臨床評価データ)と共に、前述の方法が使用されうるという事実を指す。具体的に、このような方法は、(a)推測される頭部への損傷後約24時間以内に対象から採取された試料に対するアッセイを実施して、試料中の、ユビキチンカルボキシ末端ヒドロラーゼL1(UCH-L1)、グリア原線維性酸性タンパク質(GFAP)又はこれらの組合せを含む早期バイオマーカーの試料中レベルを測定又は検出するステップ;及び(b)早期バイオマーカーの試料中レベルが、早期バイオマーカーの基準レベルより高い場合、対象に対する、MRIスキャン又はCTスキャンなどのイメージング手順を実施し(イメージング手順の実施「を組み入れ」)、早期バイオマーカーの試料中レベルが、早期バイオマーカーの基準レベルより低い場合、対象に対するイメージング手順を実施しない(イメージング手順の実施「を除外する」)ステップを含みうる。試料は、生物学的試料でありうる。
本開示は、他の方法の中でとりわけ、頭部への損傷を負った、又は負った可能性があるヒト対象に対して、MRIスキャン又はCTスキャンなどのイメージング手順を実施するのかどうかについて決定する一助となる方法に関する。本明細書において使用された「ヒト対象に対して、MRIスキャン又はCTスキャンなどのイメージング手順を実施するのかどうかについての決定」とは、対象のMRIが陽性となる、又は頭部CTスキャンが陽性となる、すなわち、頭蓋内病変が存在する可能性が、そうでない場合より高いことを決定するのに、他の情報(例えば、臨床評価データ)と共に、前述の方法が使用されうるという事実を指す。具体的に、このような方法は、推測される損傷から約24時間以内に対象から採取された第1の試料及び第1の試料が採取された約3~約6時間後に対象から採取された第2の試料である、対象から得られた、少なくとも2つの試料に対して、アッセイを実施するステップ;少なくとも2つの試料中において、ユビキチンカルボキシ末端ヒドロラーゼL1(UCH-L1)、グリア原線維性酸性タンパク質(GFAP)又はこれらの組合せのような、外傷性脳損傷についての早期バイオマーカーを検出するステップ及び早期バイオマーカーのレベルが、第1の試料から、第2の試料へと、少なくともある絶対量だけ、低下又は上昇する場合に、MRIスキャン又はCTスキャンなどのイメージング手順を実施し、早期バイオマーカーのレベルの、第1の試料から、第2の試料への、少なくともある絶対量だけの、低下又は上昇が見られない場合に、イメージング手順を実施しない(イメージング手順の実施「を除外する」)ステップを含みうる。試料は、生物学的試料でありうる。
上記された方法において同定された対象は、処置又はモニタリングされうる。一部の実施形態において、本方法は、ヒト対象を、当技術分野において公知である、任意の処置のような、外傷性脳損傷処置により処置するステップをさらに含む。例えば、外傷性脳損傷の処置は、頭部への損傷の重症度に応じて、様々な形態を取りうる。例えば、軽度TBIを患う対象の場合、処置は、休息、スポーツのような、身体活動を控えること、日向に出るときの、光線の回避又はサングラスの着用、頭痛又は偏頭痛を緩和するための投薬、抗悪心薬などのうちの1つ以上を含みうる。中等度、重度又は中等度~重度のTBIを患う患者のための処置は、1つ以上の適切な薬物(例えば、利尿剤、抗痙攣薬物、個体を鎮静させ、薬物誘導性昏睡へと導く薬物など)又は他の医薬若しくは生物医薬(TBIの処置のために公知の医薬又は将来開発される医薬)、1つ以上の手術手順(例えば、血腫の除去、頭蓋骨骨折の修復、減圧開頭術など)、気道の保護及び1つ以上の治療(例えば、1つ以上のリハビリ、認知行動学的治療、アンガーマネージメント、心理カウンセリングなど)を含みうる。一部の実施形態において、本方法は、ヒト対象をモニタリングするステップをさらに含む。一部の実施形態において、対象は、CTスキャン又はMRIによりモニタリングされうる。
上記された方法において、UCH-L1レベルは、イムノアッセイ、タンパク質免疫沈降、免疫電気泳動、化学解析、SDS-PAGE及びウェスタンブロット解析若しくはタンパク質免疫染色のような抗体依存的方法のような、電気泳動解析、タンパク質アッセイ、競合的結合アッセイ、機能的タンパク質アッセイ、又は高速液体クロマトグラフィー(HPLC)若しくは液体クロマトグラフィー-質量分析(LC/MS)のような、クロマトグラフィー法若しくは分光法のような、任意の手段により測定されうる。また、当業者に公知の臨床化学フォーマットのアッセイも利用されうる。
本明細書において記載された方法は、「UCH-L1抗体」と称される、ユビキチンカルボキシ末端ヒドロラーゼL1(「UCH-L1」)(又はこの断片)に特異的に結合する単離抗体を使用しうる。UCH-L1抗体は、UCH-L1状態を、外傷性脳損傷の尺度として評価するか、又はUCH-L1の、試料中の存在を検出する、UCH-L1の、試料中量を定量する、若しくはUCH-L1の、試料中の存在を検出し、かつ、UCH-L1の、試料中量を定量するのに使用されうる。
「ユビキチンC末端ヒドロラーゼ」としてもまた公知のユビキチンカルボキシ末端ヒドロラーゼL1(UCH-L1)(「UCH-L1」)は、脱ユビキチン化酵素である。UCH-L1は、その産物が、ユビキチンの、小型のC末端付加物を加水分解して、ユビキチン単量体を発生させる遺伝子ファミリーのメンバーである。UCH-L1の発現は、ニューロン及びびまん性神経内分泌系の細胞並びにそれらの腫瘍に、高度に特異的である。UCH-L1は、全てのニューロン内に、豊富に存在し(全脳タンパク質のうちの、1~2%を占める)、とりわけ、ニューロン及び精巣/卵巣において発現される。UCH-L1の触媒三残基は、そのヒドロラーゼ活性の一因となる、90位におけるシステイン、176位におけるアスパラギン酸及び161位におけるヒスチジンを含有する。
抗体は、UCH-L1、この断片、UCH-L1のエピトープ又はこの変異体に結合する抗体である。抗体は、抗UCH-L1抗体の断片又はこの変異体若しくは誘導体でありうる。抗体は、ポリクローナル又はモノクローナル抗体でありうる。抗体は、キメラ抗体、単鎖抗体、アフィニティー成熟抗体、ヒト抗体、ヒト化抗体、完全ヒト抗体又はFab断片のような抗体断片又はこれらの混合物でありうる。抗体の断片又は誘導体は、F(ab’)2断片、Fv断片又はscFv断片を含みうる。抗体の誘導体は、ペプチド模倣体により作製されうる。さらに、単鎖抗体を作製するために記載されている技法は、単鎖抗体を作製するように適合させられうる。
抗体は、UCH-L1(配列番号1)、この断片又はこの変異体に免疫特異的に結合しうる。抗体は、エピトープ領域内の、少なくとも3アミノ酸、少なくとも4アミノ酸、少なくとも5アミノ酸、少なくとも6アミノ酸、少なくとも7アミノ酸、少なくとも8アミノ酸、少なくとも9アミノ酸又は少なくとも10アミノ酸を免疫特異的に認識することが可能であり、これらに結合しうる。抗体は、エピトープ領域のうちの、少なくとも3連続アミノ酸、少なくとも4連続アミノ酸、少なくとも5連続アミノ酸、少なくとも6連続アミノ酸、少なくとも7連続アミノ酸、少なくとも8連続アミノ酸、少なくとも9連続アミノ酸又は少なくとも10連続アミノ酸を有するエピトープを免疫特異的に認識することが可能であり、これらに結合しうる。
抗体は、当業者に周知の技法を含む、様々な技法のうちのいずれかにより調製されうる。一般に、抗体は、従来の技法を介するか、又は抗体の遺伝子、重鎖及び/若しくは軽鎖の、適切な細菌細胞宿主若しくは哺乳動物細胞宿主へのトランスフェクションであって、組換えでありうる抗体の作製を可能とするためのトランスフェクションを介する、モノクローナル抗体の作出を含む細胞培養法により作製されうる。「トランスフェクション」という用語の多様な形態は、外因性DNAの、原核宿主細胞又は真核宿主細胞への導入のために一般に使用されている、多種多様な技法、例えば、電気穿孔、リン酸カルシウム沈殿、DEAE-デキストラントランスフェクションなどを包含することが意図される。抗体を、原核宿主細胞又は真核宿主細胞において発現させることが可能であるが、そのような真核細胞(そして、特に、哺乳動物の細胞)は、原核細胞より、アセンブルし、適正にフォールディングされ、免疫学的に活性である抗体を分泌する可能性が高いため、抗体の、真核細胞内の発現が好ましく、哺乳動物宿主細胞内の発現が最も好ましい。
モノクローナル抗体は、当技術分野において公知の、多種多様な技法であって、ハイブリドーマ、組換え及びファージディスプレイ技術又はこれらの組合せの使用を含む技法を使用して調製されうる。例えば、モノクローナル抗体は、当技術分野において公知であり、例えば、Harlowら、「Antibodies:A Laboratory Manual」、2版(Cold Spring Harbor Laboratory Press、Cold Spring Harbor、1988);Hammerlingら、「Monoclonal Antibodies and T-Cell Hybridomas」(Elsevier、N.Y.、1981)において教示されているハイブリドーマ法を含むハイブリドーマ法を使用して作製されうる。また、本明細書において使用された「モノクローナル抗体」という用語は、ハイブリドーマ技術を介して作製された抗体に限定されないことも留意される。「モノクローナル抗体」という用語は、任意の真核生物クローン、原核生物クローン又はファージクローンを含む単一クローンに由来する抗体を指し、それが作製された方法を指すものではない。
本発明の別の態様において、組換え抗体は、米国特許第5,627,052号;PCT公開第92/02551号及びBabcookら、Proc.Natl.Acad.Sci.USA、93:7843~7848(1996)において記載されている通り、当技術分野において、選択リンパ球抗体法(SLAM)と称される手順を使用して、単一の単離リンパ球から作出される。この方法において、ビオチンのようなリンカーを使用して、抗原であるUCH-L1、UCH-L1のサブユニット又はこの断片が、ヒツジ赤血球へとカップリングされ、UCH-L1に対する特異性を伴う抗体を分泌する単一細胞を同定するのに使用される、抗原特異的溶血プラークアッセイを使用して、目的の抗体を分泌する、単一の細胞、例えば、免疫化された動物のうちのいずれか1つに由来するリンパ球がスクリーニングされる。目的の抗体分泌細胞を同定した後、重鎖可変領域及び軽鎖可変領域のcDNAが、逆転写酵素PCR(RT-PCR)により、細胞からレスキューされ、次いで、これらの可変領域を、COS細胞又はCHO細胞のような、哺乳動物宿主細胞内の、適切な免疫グロブリン定常領域(例えば、ヒト定常領域)の文脈において発現させる。次いで、インビボにおいて選択されたリンパ球に由来する、増幅された免疫グロブリン配列をトランスフェクトされた宿主細胞は、例えば、UCH-L1に対する抗体を発現する細胞を単離するように、トランスフェクト細胞をパニングすることにより、インビトロにおいて、さらなる解析及び選択を経る場合がある。増幅された免疫グロブリン配列は、インビトロアフィニティー成熟法によるように、インビトロにおいて、さらに操作されうる。例えば、PCT公開第97/29131号及びPCT公開第00/56772号を参照されたい。
本発明の別の実施形態において、抗体は、ヒト免疫グロブリン遺伝子座の一部又は全部を含む非ヒト動物を、UCH-L1抗原により免疫化することにより作製される。ある実施形態において、非ヒト動物は、ヒト免疫グロブリン遺伝子座の大型断片を含み、マウス抗体の作製が欠損する操作マウス株である、XENOMOUSE(登録商標)トランスジェニックマウスである。例えば、Greenら、Nature Genetics、7:13~21(1994)並びに米国特許第5,916,771号;同第5,939,598号;同第5,985,615号;同第5,998,209号;同第6,075,181号;同第6,091,001号;同第6,114,598号及び同第6,130,364号を参照されたい。また、PCT公開第91/10741号;PCT公開第94/02602号;PCT公開第96/34096号;PCT公開第96/33735号;PCT公開第98/16654号;PCT公開第98/24893号;PCT公開第98/50433号;PCT公開第99/45031号;PCT公開第99/53049号;PCT公開第00/09560号及びPCT公開第00/37504号も参照されたい。XENOMOUSE(登録商標)トランスジェニックマウスは、完全ヒト抗体の成人様ヒトレパートリーを産生し、抗原特異的ヒトモノクローナル抗体を発生させる。XENOMOUSE(登録商標)トランスジェニックマウスは、ヒト重鎖遺伝子座及びx軽鎖遺伝子座の、メガ塩基サイズの、生殖細胞系列構成のYAC断片を導入することにより、ヒト抗体レパートリーのうちの約80%を含有する。その開示が参照により組み込まれた、Mendezら、Nature Genetics、15:146~156(1997);Green及びJakobovits、J.Exp.Med.、188:483~495(1998)を参照されたい。

抗体は、当技術分野において公知である、多数の技法のうちのいずれかにより作製されうる。例えば、重鎖及び軽鎖をコードする発現ベクターが、標準的技法により、宿主細胞へとトランスフェクトされた、宿主細胞からの発現である。「トランスフェクション」という用語の多様な形態は、外因性DNAの、原核宿主細胞又は真核宿主細胞への導入のために一般に使用されている、多種多様な技法、例えば、電気穿孔、リン酸カルシウム沈殿、DEAE-デキストラントランスフェクションなどを包含することが意図される。本発明の抗体を、原核宿主細胞又は真核宿主細胞において発現させることが可能であるが、そのような真核細胞(そして、特に、哺乳動物の細胞)は、原核細胞より、アセンブルし、適正にフォールディングさせられ、免疫学的に活性である抗体を分泌する可能性が高いため、抗体の、真核細胞内の発現が好ましく、哺乳動物宿主細胞内の発現が最も好ましい。
ヒト化抗体は、目的の抗原に免疫特異的に結合し、ヒト抗体のアミノ酸配列を実質的に有するフレームワーク(FR)領域及び非ヒト抗体のアミノ酸配列を実質的に有する相補性決定領域(CDR)を含む、抗体又はこの変異体、誘導体、類似体若しくは部分でありうる。ヒト化抗体は、所望の抗原に結合する非ヒト種抗体であって、非ヒト種に由来する、1つ以上の相補性決定領域(CDR)及びヒト免疫グロブリン分子に由来するフレームワーク領域を有する非ヒト種抗体に由来しうる。
抗UCH-L1抗体は、上記された技法を使用して、並びに、当技術分野において公知である、規定の技法を使用して作出されうる。一部の実施形態において、抗UCH-L1抗体は、United State Biological(型番:031320)、Cell Signaling Technology(型番:3524)、Sigma-Aldrich(型番:HPA005993)、Santa Cruz Biotechnology,Inc.(型番:sc-58593又はsc-58594)、R&D Systems(型番:MAB6007)、Novus Biologicals(型番:NB600-1160)、Biorbyt(型番:orb33715)、Enzo Life Sciences,Inc.(型番:ADI-905-520-1)、Bio-Rad(型番:VMA00004)、BioVision(型番:6130-50)、Abcam(型番:ab75275又はab104938)、Invitrogen Antibodies(型番:480012)、ThermoFisher Scientific(型番:MA1-46079、MA5-17235、MA1-90008 、又はMA1-83428)、EMD Millipore(型番:MABN48)、又はSino Biological Inc.(型番:50690-R011)から市販されているUCH-L1抗体のような非コンジュゲートUCH-L1抗体でありうる。抗UCH-L1抗体は、BioVision(型番:6960-25)又はAviva Systems Biology(型番:OAAF01904-FITC)から市販されている、コンジュゲートUCH-L1抗体のように、フルオロフォアへとコンジュゲートされうる。
上記された方法において、GFAPレベルは、イムノアッセイ、タンパク質免疫沈降、免疫電気泳動、化学分析、SDS-PAGE及びウェスタンブロット解析若しくはタンパク質免疫染色のような抗体依存的方法、電気泳動解析、タンパク質アッセイ、競合的結合アッセイ、機能的タンパク質アッセイ又は高速液体クロマトグラフィー(HPLC)若しくは液体クロマトグラフィー-質量分析(LC/MS)のような、クロマトグラフィー法若しくは分光法のような、任意の手段により測定されうる。また、当業者に公知のフォーマットのような、臨床化学フォーマットのアッセイも利用されうる。
本明細書において記載された方法は、「GFAP抗体」と称される、グリア細胞線維性酸性タンパク質(「GFAP」)(又はこの断片)に特異的に結合する単離抗体を使用しうる。GFAP抗体は、GFAP状態を、外傷性脳損傷の尺度として評価する、GFAPの、試料中の存在を検出する、GFAPの、試料中に存在する量を定量するか、又はGFAPの、試料中の存在を検出し、かつ、GFAPの、試料中量を定量するのに使用されうる。
グリア細胞線維性酸性タンパク質(GFAP)とは、星状細胞の細胞骨格の部分を構成し、星状細胞由来の細胞についての、最も特異的マーカーであることが実証されている、50kDaの細胞質内線維性タンパク質である。GFAPタンパク質は、ヒトにおいて、GFAP遺伝子によりコードされる。GFAPとは、成熟星状細胞の、主要な中間体線維である。分子中央のロッドドメインにおいて、GFAPは、他の中間体線維との、大きな構造的相同性を共有する。GFAPは、星状細胞突起へと、構造的安定性をもたらすことにより、星状細胞の運動性及び形状に関与する。グリア細胞線維性酸性タンパク質及びその分解産物(GFAP-BDP)とは、外傷性脳損傷(TBI)後において、病態生理学的応答の一部として、血液へと放出された脳特異的タンパク質である。外傷、遺伝障害又は化学物質により引き起こされたヒトCNSへの損傷に続き、星状細胞は、増殖し、細胞体及び細胞突起の、多大な肥大を示し、GFAPは、顕著に上方調節される。これに対し、星状細胞の悪性度が増大すると、進行性の、GFAP産生の喪失が見られる。GFAPはまた、シュワン細胞、消化管グリア細胞、唾液腺新生物、転移性腎癌、喉頭蓋軟骨、下垂体細胞、未成熟希突起膠細胞、乳頭状髄膜腫及び乳腺の筋上皮細胞においても検出されうる。

抗体は、GFAP(配列番号2)、この断片又はこの変異体に免疫特異的に結合しうる。抗体は、エピトープ領域内の、少なくとも3アミノ酸、少なくとも4アミノ酸、少なくとも5アミノ酸、少なくとも6アミノ酸、少なくとも7アミノ酸、少なくとも8アミノ酸、少なくとも9アミノ酸又は少なくとも10アミノ酸を免疫特異的に認識することが可能であり、これらに結合しうる。抗体は、エピトープ領域のうちの、少なくとも3連続アミノ酸、少なくとも4連続アミノ酸、少なくとも5連続アミノ酸、少なくとも6連続アミノ酸、少なくとも7連続アミノ酸、少なくとも8連続アミノ酸、少なくとも9連続アミノ酸又は少なくとも10連続アミノ酸を有するエピトープを免疫特異的に認識することが可能であり、これらに結合しうる。
抗体は、当業者に周知の技法を含む、様々な技法のうちのいずれかにより調製されうる。一般に、抗体は、従来の技法を介するか、又は抗体の遺伝子、重鎖及び/若しくは軽鎖の、適切な細菌細胞宿主若しくは哺乳動物細胞宿主へのトランスフェクションであって、組換えでありうる抗体の作製を可能とするためのトランスフェクションを介する、モノクローナル抗体の作出を含む細胞培養法により作製されうる。「トランスフェクション」という用語の多様な形態は、外因性DNAの、原核宿主細胞又は真核宿主細胞への導入のために一般に使用されている、多種多様な技法、例えば、電気穿孔、リン酸カルシウム沈殿、DEAE-デキストラントランスフェクションなどを包含することが意図される。抗体を、原核宿主細胞又は真核宿主細胞において発現させることが可能であるが、そのような真核細胞(特に、哺乳動物の細胞)は、原核細胞より、アセンブルし、適正にフォールディングされ、免疫学的に活性である抗体を分泌する可能性が高いため、抗体の、真核細胞内の発現が好ましく、哺乳動物宿主細胞内の発現が最も好ましい。
モノクローナル抗体は、当技術分野において公知の、多種多様な技法であって、ハイブリドーマ、組換え及びファージディスプレイ技術又はこれらの組合せの使用を含む技法を使用して調製されうる。例えば、モノクローナル抗体は、当技術分野において公知であり、例えば、Harlowら、「Antibodies:A Laboratory Manual」、2版(Cold Spring Harbor Laboratory Press、Cold Spring Harbor、1988);Hammerlingら、「Monoclonal Antibodies and T-Cell Hybridomas」(Elsevier、N.Y.、1981)において教示されているハイブリドーマ法を含むハイブリドーマ法を使用して作製されうる。また、本明細書において使用された「モノクローナル抗体」という用語は、ハイブリドーマ技術を介して作製された抗体に限定されないことも留意される。「モノクローナル抗体」という用語は、任意の真核生物クローン、原核生物クローン又はファージクローンを含む、単一クローンに由来する抗体を指し、それが作製された方法を指すものではない。
本発明の別の態様において、組換え抗体は、米国特許第5,627,052号;PCT公開第92/02551号及びBabcookら、Proc.Natl.Acad.Sci.USA、93:7843~7848(1996)において記載されている通り、当技術分野において、選択リンパ球抗体法(SLAM)と称される手順を使用して、単一の単離リンパ球から作出される。この方法において、ビオチンのようなリンカーを使用して、抗原であるGFAP、GFAPのサブユニット又はこの断片が、ヒツジ赤血球へとカップリングされ、GFAPに対する特異性を伴う抗体を分泌する単一細胞を同定するのに使用される、抗原特異的溶血プラークアッセイを使用して、目的の抗体を分泌する、単一の細胞、例えば、免疫化された動物のうちのいずれか1つに由来するリンパ球がスクリーニングされる。目的の抗体分泌細胞を同定した後、重鎖可変領域及び軽鎖可変領域のcDNAが、逆転写酵素PCR(RT-PCR)により、細胞からレスキューされ、次いで、これらの可変領域が、COS細胞又はCHO細胞のような、哺乳動物宿主細胞内の、適切な免疫グロブリン定常領域(例えば、ヒト定常領域)の文脈において発現される。次いで、インビボにおいて選択されたリンパ球に由来する、増幅された免疫グロブリン配列をトランスフェクトされた宿主細胞は、例えば、GFAPに対する抗体を発現する細胞を単離するように、トランスフェクト細胞をパニングすることにより、インビトロにおいて、さらなる解析及び選択を経る場合がある。増幅された免疫グロブリン配列は、インビトロアフィニティー成熟法によるように、インビトロにおいて、さらに操作されうる。例えば、PCT公開第97/29131号及びPCT公開第00/56772号を参照されたい。

抗体は、当技術分野において公知である、多数の技法のうちのいずれかにより作製されうる。例えば、重鎖及び軽鎖をコードする発現ベクターが、標準的技法により、宿主細胞へとトランスフェクトされた、宿主細胞からの発現である。「トランスフェクション」という用語の多様な形態は、外因性DNAの、原核宿主細胞又は真核宿主細胞への導入のために一般に使用されている、多種多様な技法、例えば、電気穿孔、リン酸カルシウム沈殿、DEAE-デキストラントランスフェクションなどを包含することが意図される。本発明の抗体を、原核宿主細胞又は真核宿主細胞において発現させることが可能であるが、そのような真核細胞(特に、哺乳動物の細胞)は、原核細胞より、アセンブルし、適正にフォールディングされ、免疫学的に活性である抗体を分泌する可能性が高いため、抗体の、真核細胞内の発現が好ましく、哺乳動物宿主細胞内の発現が最も好ましい。
ヒト化抗体は、目的の抗原に免疫特異的に結合し、ヒト抗体のアミノ酸配列を実質的に有するフレームワーク(FR)領域及び非ヒト抗体のアミノ酸配列を実質的に有する相補性決定領域(CDR)を含む、抗体又はこの変異体、誘導体、類似体若しくは部分でありうる。ヒト化抗体は、所望の抗原に結合する非ヒト種抗体であって、非ヒト種に由来する、1つ以上の相補性決定領域(CDR)及びヒト免疫グロブリン分子に由来するフレームワーク領域を有する非ヒト種抗体に由来しうる。
抗GFAP抗体は、上記された技法を使用して、並びに、当技術分野において公知である、規定の技法を使用して作出されうる。一部の実施形態において、抗GFAP抗体は、Dako(型番:M0761)、ThermoFisher Scientific(型番:MA5-12023、A-21282、13-0300、MA1-19170、MA1-19395、MA5-15086、MA5-16367、MA1-35377、MA1-06701又はMA1-20035)、AbCam(型番:ab10062、ab4648、ab68428、ab33922、ab207165、ab190288、ab115898又はab21837)、EMD Millipore(型番:FCMAB257P、MAB360、MAB3402、04-1031、04-1062、MAB5628)、Santa Cruz(型番:sc-166481、sc-166458、sc-58766、sc-56395、sc-51908、sc-135921、sc-71143、sc-65343又はsc-33673)、Sigma-Aldrich(型番:G3893又はG6171)又はSino Biological Inc.(型番:100140-R012-50)から市販されているGFAP抗体のような非コンジュゲートGFAP抗体でありうる。抗GFAP抗体は、ThermoFisher Scientific(型番:A-21295又はA-21294)、EMD Millipore(型番:MAB3402X、MAB3402B、MAB3402B又は MAB3402C3)又はAbCam(型番:ab49874又はab194325)から市販されている、コンジュゲートGFAP抗体のように、フルオロフォアへとコンジュゲートされうる。
試料中に存在する目的の解析物(UCH-L1及び/又はGFAP)の存在又は量を決定する、開示された方法は、上記された通りでありうる。本方法はまた、解析物を解析するための他の方法を念頭に置いて、適合させられる場合もある。周知の変化形の例は、酵素による検出(酵素イムノアッセイ(EIA)又は酵素免疫測定アッセイ(ELISA))を含む、サンドイッチイムノアッセイ(例えば、モノクローナル-ポリクローナルサンドイッチイムノアッセイ)、競合阻害イムノアッセイ(例えば、フォワード競合阻害イムノアッセイ及びリバース競合阻害イムノアッセイ)、酵素多重化イムノアッセイ法(EMIT)、競合結合アッセイのようなイムノアッセイ、生物発光エネルギー移動(BRET)、ワンステップ抗体検出アッセイ、同種アッセイ、異種アッセイ、キャプチャーオンザフライアッセイなどを含むがこれらに限定されない。
目的の解析物及び/又はペプチド若しくはこれらの断片(例えば、UCH-L1及び/若しくはGFAP並びに/又はペプチド若しくはこれらの断片、すなわち、UCH-L1及び/又はGFAP断片)は、UCH-L1及び/又はGFAP抗体を、イムノアッセイにおいて使用して解析されうる。解析物(例えば、UCH-L1及び/又はGFAP)の存在又は量は、抗体を使用し、解析物(例えば、UCH-L1及び/又はGFAP)への特異的結合を検出して決定されうる。例えば、抗体又はこの抗体断片は、解析物(例えば、UCH-L1及び/又はGFAP)に特異的に結合しうる。所望の場合、抗体のうちの1つ以上は、1つ以上の、市販のモノクローナル/ポリクローナル抗体と組み合わせて使用されうる。このような抗体は、R&D Systems,Inc.(Minneapolis、MN)及びEnzo Life Sciences International,Inc.(Plymouth Meeting、PA)のような企業から市販されている。
サンドイッチイムノアッセイは、抗体(すなわち、少なくとも1つの捕捉抗体)と、検出抗体(すなわち、少なくとも1つの検出抗体)とからなる、2つの層の間の抗原の量を測定する。捕捉抗体及び検出抗体は、抗原、例えば、UCH-L1及び/又はGFAPのような、目的の解析物上の、異なるエピトープに結合する。捕捉抗体の、エピトープへの結合は、検出抗体の、エピトープへの結合に干渉しないことが所望される。モノクローナル抗体又はポリクローナル抗体のいずれも、サンドイッチイムノアッセイにおける、捕捉抗体及び検出抗体として使用されうる。
任意選択的に、被験試料を、少なくとも1つの、第1の捕捉抗体と接触させる前に、少なくとも1つの、第1の捕捉抗体は、第1の抗体-解析物(例えば、UCH-L1及び/又はGFAP)複合体の、被験試料からの分離を容易とする固体支持体に結合されうる。当技術分野において公知である、任意の固体支持体であって、ウェル、チューブ又はビーズ(マイクロ粒子など)の形態のポリマー材料から作られた固体支持体を含むがこれらに限定されない固体支持体が使用されうる。抗体(antibody(又はantibodies))は、そのような結合が、解析物(例えば、UCH-L1及び/又はGFAP)に結合する抗体の能力に干渉しないという条件において、吸着により、化学的カップリング剤を使用する共有結合により、又は当技術分野において公知である他の手段により、固体支持体に結合されうる。さらに、必要な場合、固体支持体は、抗体上の多様な官能基との反応性を可能とするように、誘導体化されうる。このような誘導体化は、無水マレイン酸、N-ヒドロキシスクシンイミド及び1-エチル-3-(3-ジメチルアミノプロピル)カルボジイミドのようであるがこれらに限定されない、ある特定のカップリング剤の使用を必要とする。
次いで、第1の/複数の捕捉抗体-解析物(例えば、UCH-L1及び/又はGFAP)複合体の形成後、複合体は、少なくとも1つの、第2の検出抗体(第1の/複数の抗体-解析物(例えば、UCH-L1及び/又はGFAP)抗原-第2の抗体複合体の形成を可能とする条件下において)と接触される。一部の実施形態において、被験試料は、捕捉抗体と同時に、検出抗体と接触される。第1の抗体-解析物(例えば、UCH-L1及び/又はGFAP)複合体が、1つを超える検出抗体と接触される場合、第1の/複数の捕捉抗体-解析物(例えば、UCH-L1及び/又はGFAP)-複数の抗体検出複合体が形成される。少なくとも第2の(及び後続の)抗体が、第1の抗体-解析物(例えば、UCH-L1及び/又はGFAP)複合体と接触される場合、第1の抗体と同様に、上記された条件と同様の条件下におけるインキュベーション時間が、第1の/複数の抗体-解析物(例えば、UCH-L1及び/又はGFAP)-第2の/複数の抗体複合体の形成に要求される。好ましくは、少なくとも1つの、第2の抗体は、検出可能な標識を含有する。検出可能な標識は、第1の/複数の抗体-解析物(例えば、UCH-L1及び/又はGFAP)-第2の/複数の抗体複合体の形成の前に、これと同時に、又はこの後に、少なくとも1つの、第2の抗体に結合されうる。当技術分野において公知である、任意の検出可能な標識が使用されうる。
フォワード競合フォーマットにおいて、公知の濃度の、標識された目的の解析物(例えば、蛍光標識、切断可能なリンカーと接合されたタグなどを有する解析物(例えば、UCH-L1及び/又はGFAP))のアリコートが、被験試料中の目的の解析物(例えば、UCH-L1及び/又はGFAP)と、目的の解析物抗体(例えば、UCH-L1及び/又はGFAP抗体)への結合について競合するように使用される。
リバース競合アッセイにおいて、固定化された目的の解析物(例えば、UCH-L1及び/又はGFAP)は、被験試料及び少なくとも1つの標識された抗体と、逐次的に、又は同時に接触されうる。
キャプチャーオンザフライアッセイにおいて、固体基質は、固定化剤によりあらかじめコーティングされている。捕捉剤、解析物(例えば、UCH-L1及び/又はGFAP)及び検出剤が、固体基質へと併せて添加されるのに続き、検出の前に、洗浄ステップがなされる。捕捉剤は、解析物(例えば、UCH-L1及び/又はGFAP)に結合することが可能であり、固定化剤のためのリガンドを含む。捕捉剤及び検出剤は、本明細書において記載された、又は当技術分野で公知である、捕捉又は検出が可能な、抗体又は他の任意の部分でありうる。リガンドは、ペプチドタグを含むことが可能であり、固定化剤は、抗ペプチドタグ抗体を含みうる。代替的に、リガンド及び固定化剤は、キャプチャーオンザフライアッセイのために利用されるように、併せて結合することが可能な薬剤の任意の対(例えば、特異的結合対及び当技術分野で公知であるような他の対)でありうる。1つを超える解析物が測定されうる。一部の実施形態において、固体基質は、抗原によりコーティングされる場合があり、解析される解析物は、抗体である。
上記された、診断法、予後診断法及び/又は評価法は、診断、予後診断及び評価のための他の因子の使用をさらに含みうる。一部の実施形態において、外傷性脳損傷は、GCS(Glasgow Coma Scale)又はGOSE(Extended Glasgow Outcome Scale)を使用して診断されうる。他の検査、スケール又は指標もまた、単独で、又はGCS(Glasgow Coma Scale)と組み合わせて使用されうる。例は、RLAS(Ranchos Los Amigos Scale)である。RLAS(Ranchos Los Amigos Scale)は、意識、認知、行動及び環境との相互作用のレベルを測定する。RLAS(Ranchos Los Amigos Scale)は、レベルI:応答なし;レベルII:一般的な応答;レベルIII:局所的応答;レベルIV:混乱(興奮);レベルV:混乱(不適切);レベルVI:混乱(適切);レベルVII:自動的(適切)及びレベルVIII:意図的(適切)を含む。
一部の実施形態において、試料は、ヒト対象が、物理的振盪、閉鎖性頭部外傷若しくは開放性頭部外傷をもたらす外部の機械的力若しくは他の力、1回以上の転倒、爆発若しくは爆破による鈍的衝撃により引き起こされた頭部損傷又は他の種類の鈍的外傷を負った後において得られる。一部の実施形態において、ヒト対象が、化学物質、毒素又は化学物質と毒素との組合せを摂取した後又はこれらへと曝露された後において、試料が得られる。このような化学物質及び/又は毒素の例は、火、カビ、アスベスト、害虫駆除剤、殺虫剤、有機溶剤、塗料、糊、ガス(一酸化炭素、硫化水素及びシアン化物など)、有機金属(メチル水銀、テトラエチル鉛及び有機スズなど)及び/又は1つ以上の乱用薬物を含む。一部の実施形態において、試料は、自己免疫疾患、代謝性障害、脳腫瘍、低酸素症、1つ以上のウイルス、髄膜炎、水頭症又はこれらの組合せを患うヒト対象から得られる。
本明細書において使用された「試料」、「被験試料」、「生物学的試料」とは、UCH-L1及び/又はGFAPを含有するか、又はこれを含有することが推測される体液試料を指す。試料は、任意の適切な供給源に由来しうる。場合によって、試料は、液体、流動性の粒子状固体又は固体粒子の懸濁流体を含みうる。場合によって、試料は、本明細書において記載された解析の前に処理されうる。例えば、試料は、解析の前に、その供給源から、分離又は精製されうるが、ある特定の実施形態において、UCH-L1及び/又はGFAPを含有する非処理試料が、直接的にアッセイされうる。特定の例において、UCH-L1及び/又はGFAPの供給源は、ヒト体内物質(例えば、体液、全血、血清、血漿のような血液、尿、唾液、汗、痰、精液、粘液、涙液、リンパ液、羊水、間質液、肺洗浄液、脳脊髄液、糞便、組織、臓器など)である。組織は、骨格筋組織、肝臓組織、肺組織、腎臓組織、心筋組織、脳組織、骨髄、子宮頸部組織、皮膚などを含みうるがこれらに限定されない。試料は、液体試料又は固体試料の液体抽出物でありうる。ある特定の場合、試料の供給源は、組織の分解/細胞の溶解により可溶化されうる、臓器又は生検試料のような組織でありうる。
対照試料を含むことが所望でありうる。対照試料は、上記された通り、対象に由来する試料と共時的に解析されうる。対象試料から得られた結果は、対照試料から得られた結果と比較されうる。試料についてのアッセイ結果が比較されうる、検量線が用意されうる。このような検量線は、マーカーのレベルを、アッセイ単位、すなわち、蛍光標識が使用される場合なら、蛍光のシグナル強度の関数として提示する。複数のドナーから採取された試料を使用して、正常健常組織におけるUCH-L1及び/又はGFAPの基準レベル並びに上記において明示された特徴のうちの1つ以上を有しうるドナーから採取された組織内の、「危険性がある」UCH-L1及び/又はGFAPのレベルについての検量線が用意されうる。

i-STAT(登録商標)GFAPアッセイ
i-STAT(登録商標)GFAPアッセイを、TBI患者集団研究において使用した。捕捉モノクローナル抗体としての抗体A及び検出モノクローナル抗体としての抗体Bのようなモノクローナル抗体対を使用した。抗体A及び抗体Bは、Abbott Laboratories(Abbott Park、IL)社内において開発された、例示的な抗GFAP抗体である。GFAPアッセイデザインを、鍵となる性能属性に対して査定した。カートリッジ構成は、抗体構成:抗体A(捕捉抗体)/抗体B(検出抗体);試薬条件:0.8%の固体、250μg/mLのFabアルカリホスファターゼクラスターコンジュゲート及び試料注入口の印字:GFAP特異的であった。アッセイ時間は、10~15分間(試料捕捉時間を7~12分間とする)であった。
TBI集団研究(TRACK-TBI)
Transforming Research and Clinical Knowledge in Traumatic Brain Injury(TRACK-TBI)研究とは、大規模かつ複雑なプロジェクトである。その機関間連携及び官民連携は、11の臨床施設、7つのコア、のべ約50の共同研究機関、共同研究企業及び支援団体から構成される。3つの臨床施設からの臨床データに基づく、初期のTRACK-TBIパイロット研究は、TBI Common Data Elementsを精緻化する一助となり、TRACK-TBI研究のための、TBI Infomation Commonsの原型を創出した。
a)推測される頭部への損傷後約24時間以内に対象から採取された試料に対するアッセイを実施して、試料中の、ユビキチンカルボキシ末端ヒドロラーゼL1(UCH-L1)、グリア原線維性酸性タンパク質(GFAP)又はこれらの組合せを含む早期バイオマーカーの試料中レベルを測定又は検出するステップ;及び
b)対象に対して、イメージング手順を実施するのかどうかを決定するステップであって、早期バイオマーカーの試料中レベルが、早期バイオマーカーの基準レベルより高い場合、対象に対するイメージング手順を実施し、早期バイオマーカーの試料中レベルが、早期バイオマーカーの基準レベルより低い場合、対象に対するイメージング手順を実施しないステップ
を含み、
基準レベルが、(a)少なくとも約70%~約100%の間の感度及び少なくとも約30%~約100%の間の特異度を有するアッセイにより決定される;又は(b)少なくとも約20pg/mL~約200pg/mLの間である
方法。

c)対象に対して、イメージング手順を実施するのかどうかを決定するステップであって、早期バイオマーカーのレベルが第1の試料から第2の試料へ少なくとも絶対量の低下又は上昇をする場合に、対象に対するイメージング手順を実施し、早期バイオマーカーのレベルの第1の試料から第2の試料へ少なくとも絶対量の低下又は上昇が見られない場合に、対象に対するイメージング手順を実施しないステップ
を含む方法。
a)推測される損傷後、24時間以内に対象から採取された第1の試料及び第1の試料が採取された約3~約6時間後に対象から採取された第2の試料である、対象から得られた少なくとも2つの試料に対して、アッセイを実施するステップ;
b)少なくとも2つの試料中において、ユビキチンカルボキシ末端ヒドロラーゼL1(UCH-L1)、グリア細胞線維性酸性タンパク質(GFAP)又はこれらの組合せを含む外傷性脳損傷の早期バイオマーカーを検出するステップ;並びに

を含む方法。
A.試料を、同時に、又は逐次的に、任意の順序において、
(1)UCH-L1上又はUCH-L1断片上のエピトープに結合してUCH-L1捕捉抗体-UCH-L1抗原複合体を形成するUCH-L1捕捉抗体、及び
(2)検出可能な標識を含み、UCH-L1捕捉抗体が結合していないUCH-L1上のエピトープに結合してUCH-L1抗原-UCH-L1検出抗体複合体を形成するUCH-L1検出抗体
と接触させて、UCH-L1捕捉抗体-UCH-L1抗原-UCH-L1検出抗体複合体を形成すること並びに
B.UCH-L1の、試料中量又は試料中濃度を、UCH-L1捕捉抗体-UCH-L1抗原-UCH-L1検出抗体複合体内の検出可能な標識により発生したシグナルに基づき測定すること
を含む、条項43~69のいずれか一項に記載の方法。
A.試料を、同時に、又は逐次的に、任意の順序において、
(1)GFAP上又はGFAP断片上のエピトープに結合してGFAP捕捉抗体-GFAP抗原複合体を形成するGFAP捕捉抗体、及び
(2)検出可能な標識を含み、GFAP捕捉抗体が結合していないGFAP上のエピトープに結合してGFAP抗原-GFAP検出抗体複合体を形成するGFAP検出抗体と接触させて、GFAP捕捉抗体-GFAP抗原-GFAP検出抗体複合体を形成すること並びに
B.GFAPの、試料中量又は試料中濃度を、GFAP捕捉抗体-GFAP抗原-GFAP検出抗体複合体内の検出可能な標識により発生したシグナルに基づき測定すること
を含む、条項43~71のいずれか一項に記載の方法。
Claims (30)
- ヒト対象を査定して、頭部損傷についての磁気共鳴イメージング手順を実施するのかどうかを決定するための一助となる方法であって、
a)推測される頭部への損傷後24時間以内に、前記対象から得られた全血、血清、血漿又は脳脊髄液である試料に対するアッセイを実施して、ユビキチンカルボキシ末端ヒドロラーゼL1(UCH-L1)、又はUCH-L1とグリア原線維性酸性タンパク質(GFAP)の組合せを含む早期バイオマーカーの試料中レベルを測定するステップを含み、
b)前記早期バイオマーカーの基準レベルより高い前記早期バイオマーカーの試料中レベルは、軽度、中等度、重度又は中等度~重度の外傷性脳損傷(TBI)についての対象の査定の一助とするために、前記対象に対して磁気共鳴イメージング手順を実施すべきであることを示し、前記早期バイオマーカーの基準レベルより低い前記早期バイオマーカーの試料中レベルは、前記対象に対して磁気共鳴イメージング手順を実施すべきではないことを示し、
UCH-L1についての前記基準レベルが、80pg/mL~150pg/mLの間である
方法。 - 前記基準レベルが、(a)少なくとも70%の感度及び少なくとも30%の特異度を有するアッセイにより決定される;又は(b)少なくとも80%の感度及び少なくとも30%の特異度を有するアッセイにより決定される、請求項1に記載の方法。
- 前記対象が、アッセイが実施される前に、又は実施された後において、イメージング手順を受けている、請求項1又は2に記載の方法。
- 前記対象が、前記イメージング手順に基づき、外傷性脳損傷を有すると推測される、請求項3に記載の方法。
- GFAPについての前記基準レベルが、20pg/mL~200pg/mLの間である、請求項1~4のいずれか一項に記載の方法。
- 前記試料が、前記推測される頭部への損傷後、0~12時間以内に採取される、請求項1~5のいずれか一項に記載の方法。
- ヒト対象を査定して、頭部損傷についての磁気共鳴イメージング手順を実施するのかどうかを決定するための一助となる方法であって、
a)推測される損傷後、24時間以内に前記対象から採取された第1の試料及び前記第1の試料が採取された3~6時間後に前記対象から採取された第2の試料である、前記対象から得られた全血、血清、血漿又は脳脊髄液である少なくとも2つの試料に対して、アッセイを実施するステップ;並びに
b)前記少なくとも2つの試料中において、ユビキチンカルボキシ末端ヒドロラーゼL1(UCH-L1)、グリア細胞線維性酸性タンパク質(GFAP)又はこれらの組合せを含む外傷性脳損傷の早期バイオマーカーを検出するステップを含み;
c)前記早期バイオマーカーのレベルにおける前記第1の試料から前記第2の試料への絶対量の低下又は上昇は、前記対象に対して磁気共鳴イメージング手順を実施すべきであることを示し、前記早期バイオマーカーのレベルにおける前記第1の試料から前記第2の試料への絶対量の低下又は上昇が見られないことは、前記対象に対して磁気共鳴イメージング手順を実施すべきではないことを示す、方法。 - 前記対象が、アッセイが実施される前に、又は実施された後において、イメージング手順を受けている、請求項7に記載の方法。
- 前記対象が、前記イメージング手順に基づき、外傷性脳損傷を有すると推測される、請求項8に記載の方法。
- 前記絶対量が、少なくとも70%~100%の間の感度及び少なくとも30%~100%の間の特異度を有するアッセイにより決定される、請求項7に記載の方法。
- 前記絶対量が、(a)少なくとも80%の感度及び少なくとも30%の特異度又は(b)少なくとも75%の感度及び少なくとも40%の特異度を有するアッセイにより決定される、請求項7に記載の方法。
- 前記絶対量が、少なくとも10pg/mL~150pg/mLの間である、請求項7~11のいずれか一項に記載の方法。
- 前記早期バイオマーカーがUCH-L1であり、前記絶対量が少なくとも30pg/mL~100pg/mLの間であるか、前記早期バイオマーカーがGFAPであり、前記絶対量が少なくとも10pg/mL~150pg/mLの間であるか、又はこれらの組合せである、請求項7~12のいずれか一項に記載の方法。
- 前記第1の試料が、前記推測される頭部への損傷後、0~12時間以内に採取される、請求項7~13のいずれか一項に記載の方法。
- 前記第2の試料が、前記第1の試料が採取された後、3時間~6時間の間に、前記対象から採取される、請求項7~13のいずれか一項に記載の方法。
- UCH-L1のレベルを測定するステップが、
A.前記試料を、同時に、又は逐次的に、任意の順序において、
(1)UCH-L1上又はUCH-L1断片上のエピトープに結合してUCH-L1捕捉抗体-UCH-L1抗原複合体を形成するUCH-L1捕捉抗体、及び
(2)検出可能な標識を含み、前記UCH-L1捕捉抗体が結合していないUCH-L1上のエピトープに結合してUCH-L1抗原-UCH-L1検出抗体複合体を形成するUCH-L1検出抗体
と接触させて、UCH-L1捕捉抗体-UCH-L1抗原-UCH-L1検出抗体複合体を形成すること、並びに
B.UCH-L1の、前記試料中量又は前記試料中濃度を、前記UCH-L1捕捉抗体-UCH-L1抗原-UCH-L1検出抗体複合体内の前記検出可能な標識により発生したシグナルに基づき測定すること
を含む、請求項1~15のいずれか一項に記載の方法。 - GFAPのレベルを測定するステップが、
A.前記試料を、同時に、又は逐次的に、任意の順序において、
(1)GFAP上又はGFAP断片上のエピトープに結合してGFAP捕捉抗体-GFAP抗原複合体を形成するGFAP捕捉抗体、及び
(2)検出可能な標識を含み、前記GFAP捕捉抗体が結合していないGFAP上のエピトープに結合してGFAP抗原-GFAP検出抗体複合体を形成するGFAP検出抗体
と接触させて、GFAP捕捉抗体-GFAP抗原-GFAP検出抗体複合体を形成すること、並びに
B.GFAPの、前記試料中量又は前記試料中濃度を、前記GFAP捕捉抗体-GFAP抗原-GFAP検出抗体複合体内の前記検出可能な標識により発生したシグナルに基づき測定すること
を含む、請求項1~16のいずれか一項に記載の方法。 - 前記対象が、物理的振盪、閉鎖性頭部外傷若しくは開放性頭部外傷をもたらす外部の機械的力若しくは他の力、1回以上の転倒、爆発若しくは爆破による鈍的衝撃又は他の種類の鈍力外傷により引き起こされた頭部への損傷を負った後において、前記試料が得られる、請求項1~17のいずれか一項に記載の方法。
- 前記試料が、前記対象が、化学物質、毒素又は化学物質と毒素との組合せを摂取したか、又はこれらへと曝露された後において得られる、請求項1~18のいずれか一項に記載の方法。
- 前記化学物質又は毒素が、火、カビ、アスベスト、害虫駆除剤、殺虫剤、有機溶剤、塗料、糊、ガス、有機金属、乱用薬物又はこれらの1つ以上の組合せである、請求項19に記載の方法。
- 前記試料が、自己免疫疾患、代謝性障害、脳腫瘍、低酸素症、ウイルス、髄膜炎、水頭症又はこれらの組合せを患う対象から得られる、請求項1~20のいずれか一項に記載の方法。
- 前記対象の臨床状態、前記対象の検査値、軽度、中等度、重度又は中等度~重度の外傷性脳損傷の罹患についての前記対象の分類、前記対象による低レベル又は高レベルのUCH-L1の呈示、及び前記対象が頭部への損傷を負った可能性がある任意の事象のタイミングからなる群から選択される因子に関わらず、任意の対象に対して実行されうる、請求項1~21のいずれか一項に記載の方法。
- 前記対象をモニタリングするステップをさらに含む、請求項1~22のいずれか一項に記載の方法。
- 前記試料が、全血試料である、請求項1~23のいずれか一項に記載の方法。
- 前記試料が、血清試料である、請求項1~23のいずれか一項に記載の方法。
