「リポソーム」とは、一般に、内部空洞を含む球状又はほぼ球状の粒子を指す。リポソームの壁は、脂質の二重層を含み得る。これらの脂質はリン脂質であり得る。多数の脂質及び/又はリン脂質が、リポソームを作るために使用され得る。一例は、疎水性及び極性頭部基部分を有する両親媒性脂質であり、これは、リン脂質によって例示されるように、水中の二重層小胞に自発的に形成され得るか、又は、その疎水性部分が二重層膜の内部の疎水性領域と接触し、その極性頭部基部分が膜の外部の極性表面に向いた状態で、脂質二重層に安定に組み込まれ得る。リポソームは、本明細書の例、参照によりその全体が本明細書に組み込まれる国際公開第2016057812A1号及び国際公開第2012139080A1号に記載されているものを含む、任意の公知の方法によって調製され得る。図1は、アミロイド沈着のMRIのための標的造影剤を含むリポソームの例示的な断面図を提供する。
ADx-001の動物モデルにおける用量応答、薬物動態及び体内分布を研究した。用量範囲の有効性研究を、早期発症ADのTgマウスモデルにおいて行った。イメージングは、T1w-SE及びFSE-IR配列を使用して1テスラ永久磁石MRスキャナーで実施した。ADx-001を、0.10、0.15、及び0.20mmol Gd/kgの3つの用量レベルで試験した。Tg及び年齢が一致するWT対照動物(n=6/用量レベル/遺伝子型)を、造影前及びADx-001の投与の4日後(遅延造影後)にイメージングした。造影前及び遅延造影後画像を定性的及び定量的に分析して、アミロイドβ斑の死後組織学的同定に対する感度、特異性及び精度を決定した。ADx-001の薬物動態をサル及びイヌにおいて研究した。ADx-001の投与後の複数の時点で血液試料を採取し、GdレベルをICP-MSによって分析した。ADx-001の体内分布をラットにおいて研究した。標的器官(肝臓、脾臓、腎臓、皮膚、骨及び脳)中のGdレベルを、ADx-001の投与後4日目及び28日目にICP-MSによって測定した。
図4を参照して、出発材料(E)-2-((2-アミノエチル)(4-(2-(ピリミジン-4-イル)ビニル)フェニル)アミノ)エタン-1-オール(SM1)をDIPEAと反応させてET3-73中間体1を形成した。ET3-73中間体1をTBDMS-OTf(TBDMS-トリフラート)と反応させてET3-73中間体2を得た。ET3-73中間体2をHO-PEG3500-NH2(SM2)とカップリングさせてET3-73中間体3を形成した。ET3-73中間体3をビス-ペンタフルオロフェニル-カルボナートと反応させ、活性化したET3-73中間体3を、ビス-トリメチルシリル-アセトアミドを使用してシリル化したDSPEと合わせて、ET3-73中間体4を得た。最後に、TBAFを使用した脱保護、続いて酢酸ナトリウムアンモニウムとの反応によってET3-73アンモニウム塩を形成した。
図5を参照して、合成スキームを、DOTA-トリス(tert-ブチルエステル)(SM1)とペンタフルオロフェノール(HOpFP)とのさらなるエステル化から始める。TFA及びトリメチルクロルシラン(TMSCl)を使用して、tert-ブチルエステル保護基を除去した。BSA及びNMMの存在下でペンタフルオロフェニルエステルをDSPEにカップリングさせて、DSPE-DOTAを形成した。ガドリニウムのキレート化及び塩形成は、酢酸塩としてのガドリニウム[Gd(OAc)3]の添加、続いてスカベンジャーSiliaMetS TAAcONa(トリアミンテトラアセタート、ナトリウム塩官能化シリカゲル)の添加によって起こり、最終的なDSPE-DOTA-Gdナトリウム塩を形成した。
図6を参照して、ADx-001を以下のように調製した。
研究を、早期発症ADのAPPswe/PSEN1dE9(C57BL/6J背景、11~18月齢)二重Tgマウスモデル(JAX MMRRC Stock#005864)において行った。Tgマウスは、6~7月齢前後の脳にアミロイド斑を発症する。例3で調製したADx-001を3つの用量レベル、0.10、0.15及び0.20(mmol Gd/kg)で試験した。各用量レベルで、ADx-001をTgマウス(n=6)及びWTマウス(両変異を欠くマウス)(n=6)で試験した。ADx-001を尾静脈を介して低速ボーラス注射として静脈内投与した。

ADx-001の薬物動態(「PK」)をカニクイザル及びビーグル犬で評価した。非ナイーブ雄カニクイザル(n=3、2~5歳、2.3~3.1kg体重)に、較正された注入ポンプを使用して0.30mmol Gd/kgで約60分間かけてADx-001を静脈内投与した。血液試料を、投与前、注入終了直後、ならびに注射開始(「SOI」)の4、8、24、48、96、168、336及び672時間後にすべての動物から採取した。ビーグル犬におけるPK分析のために、動物(n=5、5~7月齢、6.2~7.9kg体重)に、ADx-001を0.30mmol Gd/kgで約60分間かけて静脈内注入した。血液試料を血漿に処理し、分析の準備ができるまで凍結保存した。
ADx-001の体内分布をラットモデルで研究した。Wistar Hanラット(10週齢、257~296g体重、n=13/処置群)に、ADx-001を0.15mmol Gd/kg(n=13)又は0.30mmol Gd/kgで単回静脈内ボーラス注射として投与した。動物を、ADx-001の投与後4日目(n=7/用量レベル)及び28日目(n=6/用量レベル)に安楽死させた。組織を採取して、標的器官(肝臓、脾臓、腎臓、皮膚、骨及び脳)におけるGdレベルを決定した。組織試料を液体窒素中で直ちに凍結し、分析の準備ができるまで-20℃で保存した。
