Description
a)PGA、好ましくはTLC分析によって決定される純度が98%を超えるPGAを準備する工程と、
b)PGAを脂肪酸又はその塩と共に極性溶媒に懸濁する工程であって、該脂肪酸又はその塩がPGAに対して2重量%~20重量%又は4重量%~10%重量の量で存在する、工程と、
c)工程b)の懸濁液を50℃~90℃の温度又は溶媒の還流温度で、15分又は30分又は1時間よりも長い時間にわたって加熱する工程と、
d)結晶化混合物をゆっくりと10℃未満の温度にする工程と、
e)PGAと脂肪酸との均質混合物の形態の結晶化物(crystallisate)を回収する工程と、
を含む。
滑膜の機能不全に起因する炎症性疾患、
腸粘膜の機能不全に起因する炎症性疾患、
尿路上皮粘膜の機能不全に起因する炎症性疾患、
ケラチン性上皮の機能不全に起因する炎症性疾患、
から選択されるのが好ましい。
794.3gのグルコサミン塩酸塩(3.68モル)を、6700mlの水に撹拌下で可溶化する。

TLC純度:98%超
950gの高純度N-パルミトイル-D-グルコサミン(HPPGA)に、50gのパルミチン酸ナトリウム塩(混合物の総重量に対して5重量%)を添加する。混合物を4リットルのメタノールに効率的な撹拌下で懸濁し、2時間還流させる。次いで、撹拌を行いながら、混合物を1時間のうちに5℃まで徐々に冷却する。次いで、固体を濾過によって分離し、高真空下で乾燥させる。

PGAを上記のような均質PGA/脂肪酸混合物として微粒子化した。
微粒子化チャンバ内径300mm、
流体ジェット圧8バール、
生成物供給量20kg/時間~25kg/時間。
PGA(脂肪酸を添加した)のサブミクロン化プロセスを、WA BACHOFEN(スイス、ムッテンツ)のDYNO-MILL MULTILABミクロスフェアミル、1259.7gの0.3mm径ジルコニアビーズを用いた「湿式粉砕」法によって行った。
粒径分布の決定を、1分間の超音波処理後に湿潤サンプルに対して行った。
上皮機能不全依存性炎症に対する種々の物理的形態のN-パルミトイル-D-グルコサミン(PGA)の薬理効果
上皮機能不全が足レベルで決定されるラットの足におけるカラギーナン浮腫の実験モデルを用いた。
非微粒子化N-パルミトイル-D-グルコサミン(PGA)、
微粒子化N-パルミトイル-D-グルコサミン(PGA-m)、
クルクミンと共微粒子化したN-パルミトイル-D-グルコサミン(PGACUR-m)、
サブミクロン形態のN-パルミトイル-D-グルコサミン(PGA-sm)。
微粒子化クルクミン(CUR-m)。
30mg/kg用量のPGA(非微粒子化)による処置は、薬理効果を生じず、
20mg/kg用量のPGA-mによる処置は、明らかな薬理効果を生じ、
20mg/kg用量のPGA-smによる処置は、顕著な薬理効果を生じず、
10mg/kg用量の微粒子化CUR単独による処置は、顕著な薬理効果を生じず、
30mg/kg(20mg/kgのPGA-m及び10mg/kgのCUR-mに相当する)用量のPGACUR-mによる処置は、PGA-m単独で得られるよりも明らかに優れた顕著な薬理効果を生じる。
この場合にも、上皮機能不全が足レベルで決定されるラットの足におけるカラギーナン浮腫の実験モデルを用いた。
群A)カラギーナン
群B)カラギーナン+微粒子化パルミトイルグルコサミン(10mg/kg)
群C)カラギーナン+微粒子化ポリダチン(1mg/kg)
群D)カラギーナン+パルミトイルグルコサミン(10mg/kg)+ポリダチン(1mg/kg)(混合)
群E)カラギーナン+パルミトイルグルコサミン及び共微粒子化ポリダチン(10:1)(11mg/kg)
滑膜の機能不全に起因する炎症性疾患、
腸粘膜の機能不全に起因する炎症性疾患、
尿路上皮粘膜の機能不全に起因する炎症性疾患、
ケラチン性上皮の機能不全に起因する炎症性疾患。
実施例1 - 経口用シロップ
1本の100ml容ボトルが以下のものを含有する:
PGACUR-m 4000mg、
カルボキシメチルセルロース 3000mg、
Noveon AA1 150mg、
甘味料 10mg、
蒸留水 100mlまで適量。
100gの顆粒が以下のものを含有する:
PGACUR-m 6000mg、
ソルビトール粉末 400mg、
パルミチン酸サッカロース 20mg、
動物にとって魅力的な成分 30mg。
1つの錠剤が以下のものを含有する:
PGA-m 600mg、
微結晶性セルロース 200mg、
クロスカルメロースナトリウム 70mg、
ポリビニルピロリドン 10mg、
ステアリン酸マグネシウム 4mg。
1つの錠剤が以下のものを含有する:
PGACUR-m 600mg、
微結晶性セルロース 200mg、
クロスカルメロースナトリウム 70mg、
ポリビニルピロリドン 10mg、
ステアリン酸マグネシウム 4mg。
1つの錠剤が以下のものを含有する:
PGACUR-m 300mg、
微結晶性セルロース 180mg、
クロスカルメロースナトリウム 60mg、
ポリビニルピロリドン 10mg、
ステアリン酸マグネシウム 4mg。
1つのカプセルが以下のものを含有する:
PGACUR-m 400mg。
1つのカプセルが以下のものを含有する:
PGACUR-m 300mg、
植物油 200mg、
ダイズレシチン 50mg。
1つのカプセルが以下のものを含有する:
PGACUR-m 400mg、
植物油 200mg、
ダイズレシチン 50mg。
PGACUR-m 200mg、
坐剤用の脂肪塊(Fat mass) 1200mg。
100gのクリームが以下のものを含有する:
PGACUR-m 2000mg、
脱塩水 71g、
ワセリン油 10g、
PEG-5植物性ステロール 4g、
ピログルタミン酸ナトリウム 2g、
ステアリン酸 2.5g、
セトステアリルアルコール 2.5g、
ポリソルベート80 2%、
モノステアリン酸グリセリル 1.5g、
シリコーン油 1%、
カルボマー 0.5%、
2-フェノキシエタノール 1%。
1つの錠剤が以下のものを含有する:
PGAPOL-m 400mg、
微結晶性セルロース 250mg、
クロスカルメロースナトリウム 80mg、
ポリビニルピロリドン 15mg、
ステアリン酸マグネシウム 5mg。
Claims (18)
- 微粒子化形態のN-パルミトイル-D-グルコサミン。
- 少なくとも96体積%の粒子が0.6ミクロン~10ミクロンの粒径を有する、ミー計算アルゴリズムを用いるMalvern Mastersizer 3000ツールによって測定された粒径分布を有する、請求項1に記載のN-パルミトイル-D-グルコサミン(PGA)。
- 3.1ミクロン~3.5ミクロンのモードを有し、少なくとも97体積%の粒子が10ミクロンよりも小さい粒径分布曲線を有する、請求項2に記載のN-パルミトイル-D-グルコサミン。
- PGAに対して3%~20%、又は4%~15%、又は4.5%~6%の重量百分率の脂肪酸が添加されている、請求項1~3のいずれか一項に記載のN-パルミトイル-D-グルコサミン。
- クルクミン(CUR)及び/又はポリダチン(POL)との共微粒子化形態である、請求項1~4のいずれか一項に記載のN-パルミトイル-D-グルコサミン。
- N-パルミトイル-D-グルコサミン及びクルクミンが3:1~1:3又は2:1~1:1の重量比である、請求項5に記載のN-パルミトイル-D-グルコサミン。
- N-パルミトイル-D-グルコサミン及びポリダチンが20:1~5:1又は12:1~8:1の重量比である、請求項5に記載のN-パルミトイル-D-グルコサミン。
- 共微粒子化PGA/CUR混合物が、ミー計算アルゴリズムを用いるMalvern Mastersizer 3000ツールによって測定したときに、3.1ミクロン~3.8ミクロンのモードの粒径分布曲線を有し、少なくとも98体積%の粒子が10ミクロンよりも小さい、請求項5又は6に記載のN-パルミトイル-D-グルコサミン。
- 共微粒子化PGA/POL混合物が、ミー計算アルゴリズムを用いるMalvern Mastersizer 3000ツールによって測定したときに、3.1ミクロン~3.8ミクロンのモードの粒径分布曲線を有し、少なくとも98体積%の粒子が10ミクロンよりも小さい、請求項5又は7に記載のN-パルミトイル-D-グルコサミン。
- 請求項1~9のいずれか一項に記載のN-パルミトイル-D-グルコサミンを含むヒト用の又は動物用の医薬組成物。
- 経口投与、口腔投与、非経口投与、直腸投与又は経皮投与用の剤形に配合されている、請求項10に記載の医薬組成物。
- 請求項1~9のいずれか一項に記載のN-パルミトイル-D-グルコサミンを含む、食事療法用組成物、ダイエタリーサプリメント若しくは特別医療目的用食品(FSMP)、又は動物用の飼料若しくは栄養補給剤。
- 請求項1~9のいずれか一項に記載のN-パルミトイル-D-グルコサミンを含む化粧品組成物。
- 上皮及び滑膜の機能不全に起因するヒト及び動物の慢性全身性炎症性疾患の治療における使用のための、
請求項1~9のいずれか一項に記載のN-パルミトイル-D-グルコサミン;
請求項10若しくは11に記載の医薬組成物;又は
請求項12に記載の食事療法用組成物、ダイエタリーサプリメント若しくは特別医療目的用食品(FSMP)、又は動物用の飼料若しくは栄養補給剤。 - 前記疾患が、
滑膜の機能不全に起因する炎症性疾患、
腸粘膜の機能不全に起因する炎症性疾患、
尿路上皮粘膜の機能不全に起因する炎症性疾患、
ケラチン性上皮の機能不全に起因する炎症性疾患、
から選択される:
請求項14に記載の使用のための、
請求項1~9のいずれか一項に記載のN-パルミトイル-D-グルコサミン;
請求項10若しくは11に記載の医薬組成物;又は
請求項12に記載の食事療法用組成物、ダイエタリーサプリメント若しくは特別医療目的用食品(FSMP)、又は動物用の飼料若しくは栄養補給剤。 - N-パルミトイル-D-グルコサミンを脂肪酸の存在下で結晶化させる方法であって、
a)N-パルミトイル-D-グルコサミンを準備する工程と、
b)N-パルミトイル-D-グルコサミンを脂肪酸又はその塩と共に極性溶媒に懸濁する工程であって、該脂肪酸又はその塩がN-パルミトイル-D-グルコサミンに対して2重量%~20重量%又は4重量%~10%重量の量で存在する工程と、
c)工程b)の懸濁液を50℃~90℃の温度又は溶媒の還流温度で、15分又は30分又は1時間よりも長い時間にわたって加熱する工程と、
d)結晶化混合物をゆっくりと10℃未満の温度にする工程と、
e)N-パルミトイル-D-グルコサミンと脂肪酸との均質混合物の形態の結晶化物を回収する工程と、
を含む、方法。 - 前記極性溶媒がメタノール、エタノール、アセトン及び酢酸エチルから選択される、請求項16に記載の方法。
- 前記脂肪酸がパルミチン酸である、請求項16又は17に記載の方法。
